Rapid Resolution Liquid Chromatography Coupled with Quadrupole Time-of-Flight Mass Spectrometry-Based Metabolomics Approach to Study the Effects of Jieduquyuziyin Prescription on Systemic Lupus Erythematosus

Abstract: Jieduquyuziyin prescription (JP), a traditional Chinese medicine (TCM) prescription, has been widely used for the clinical treatment of systemic lupus erythematosus (SLE). However, the complex chemical constituents of JP and the multifactorial pathogenesis of SLE make research on the therapeutic mechanism of JP in SLE challenging. In this paper, a serum metabolomics approach based on rapid resolution liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (RRLC-Q-TOF/MS) was employed to acquire the metabolic characteristics of serum samples obtained from mice in the SLE model group, JP-treated group, prednisone acetate (PA)-treated group and control group. The orthogonal partial least squares (OPLS) was applied to recognize metabolic patterns, and an obvious separation of groups was obtained. Thirteen metabolites, namely, phosphatidylethanolamine (PE 20:3), hepoxilin B3, lyso- phosphatidylethanolamine (lyso-PE 22:6), 12S-hydroxypentaenoic acid (12S-HEPE), traumatic acid, serotonin, platelet-activating factor (PAF), phosphatidylcholine (PC 20:5),eicosapentaenoic acid (EPA), 12(S)-hydroxyei- cosatetraenoic acid (12S-HETE), 14-hydroxy docosahexaenoic acid (14-HDOHE), lyso-phosphatidylcholine (lyso-PC 20:4), and indole acetaldehyde, were identified and characterized as differential metabolites involved in the pathogenesis of SLE. After treatment with JP, the relative content of 12(S)-HETE, PAF, 12(S)-HEPE, EPA, PE (20:3), Lyso-PE(22:6), and 14-HDOHE were effectively regulated, which suggested that the therapeutic effects of JP on SLE may involve regulating disturbances to the metabolism of unsaturated fatty acid, tryptophan and phospholipid. This research also demonstrated that metabolomics is a powerful tool for researching complex disease mechanisms and evaluating the mechanism of action of TCM.
Antioxidant, Antityrosinase and Antitumor Activity Comparison: The Potential Utilization of Fibrous Root Part of Bletilla striata (Thunb.) Reichb.f.

Abstract: This study was carried out to evaluate the utilization probability of the fibrous root part (FRP) of Bletilla striata, which was usually discarded and harvesting pseudobulb part (PSP). The chemical composition, total phenolic content, DPPH radical scavenging activity, Ferric-reducing antioxidant power and tyrosinase inhibition activity were compared between FRP and PSP. Antioxidant and pro-oxidant effect as well as antitumor effect of the extract of FRP and PSP were analyzed by in vitro cell system as well. Thin layer chromatography and high performance liquid chromatography analysis indicated that the chemical compositions in the two parts were similar, but the content in FRP was much higher than PSP. Meanwhile, the FRP extracts showed higher phenolic content, stronger DPPH scavenging activity, Ferric-reducing antioxidant capacity and tyrosinase inhibition activity. Sub-fraction analysis revealed that the distribution characteristic of phenolic components and other active constituents in FRP and PSP were consistent, and mainly deposited in chloroform and acetoacetate fractions. Especially, the chloroform sub-fraction (sch) of FRP showed extraordinary DPPH scavenging activity and tyrosinase inhibition activity, with IC50 0.848 mg/L and 4.3 mg/L, respectively. Besides, tyrosinase inhibition activity was even stronger than the positive compound arbutin (31.8 mg/L). Moreover, In vitro cell system analysis confirmed that FRP extract exerts comparable activity with PSP, especially, the sub-fraction sch of FRP showed better antioxidant activity at low dosage and stronger per-oxidant activity at high dosage, and both sch of FRP and PSP can dose-dependent induce HepG2 cells apoptosis, which implied tumor therapeutic effect. Considering that an additional 0.3 kg FRP would be obtained when producing 1.0 kg PSP, our work demonstrated that FRP is very potential to be used together with PSP.

Synthesis and photocurrent response of porphyrin-containing conjugated polymers

Abstract: Porphyrin-containing conjugated polymers with fluorene or carbazole as spacer groups were prepared by Sonogashira cross-coupling reactions. The polymers were of high molecular weight and the flexible alkyl chains on fluorene or carbazole units made the conjugated polymers soluble in common organic solvents, such as THF and methylene chloride. The polymers could form high quality durable films from solution casting. Their optical and photocurrent responsive properties were investigated. It was found that the photocurrent response was directly proportional to the content of porphyrin. The incorporation of carbazole units into the polymer chains also gave positive contribution to the photocurrent generation in some extent.

Review of the ecohydrological processes and feedback mechanisms controlling sand-binding vegetation systems in sandy desert regions of China

Abstract: Soil water is the key abiotic limiting factor in desert areas and hydrological processes determine the vegetation composition, patterns and processes in desert regions. The hydrological processes can be altered by vegetation succession. In this paper, we review the major advances in ecohydrological research and their potential impact on plant-water relations in revegetated desert communities. The major advances in ecohydrological research over the past 50 years in desert areas were analyzed using a case study that investigated the long-term ecosystem effects of sand-binding vegetation in the Tengger Desert. Key challenges and opportunities for ecohydrology research in the future are also discussed in the context of the major scientific issues affecting sand binding vegetation.
Hyperbranched polymer-cored star polyfluorenes as blue light-emitting materials

Abstract: Hyperbranched polymer-cored star polyfluorenes with high molecular weights and narrow molecular weight distribution were prepared by palladium-catalyzed one-pot Suzuki polycondensation of multi-functional cores and an AB-type monomer. The optical, electrochemical and thermal properties of the hyperbranched polymer-cored star polymers were investigated. These polymers exhibited good thermal and color stability in solid state, and there was no significant blue-green emission after the polymers had been annealed in air for 2.5 h. Their three-dimensional hyperbranched structures could effectively reduce the aggregation of the peripheral rigid linear conjugated polyfluorene chains.
The ecohydrology of the soil–vegetation system restoration in arid zones: a review

Abstract: Arid zones, which cover approximately 40 percent of the earth’s land surface, support complicated and widely varied ecological systems. As such, arid zones are an important composition of the global terrestrial ecosystem, and water is the key and abiotic limited factor in ecosystem-driven processes in these areas. Ecohydrology is a new cross discipline that provides, in an objective and comprehensive manner, novel ideas and approaches to the evaluation of the interaction and feedback mechanisms involved in the soil–vegetation systems in arid zones. In addition, ecohydrology provides a theoretical basis of ecological restoration that is centered on vegetation construction. In this paper, long-term monitoring and local observations in the transitional belt between a desertified steppe and a steppified desert at the Shapotou Desert Research and Experiment Station, Tengger Desert, in northern China, were evaluated. The primary achievements and related research progress regarding ecohydrology in arid zones were analyzed and summarized, as a keystone, and the response of soil ecohydrological processes to the changes in the species composition, structure, and function of sandland vegetation was discussed. Meanwhile, the long-term ecological effects and mechanism of regulation of vegetation on soil habitat and on water-cycling were considered. As a vital participant in the ecohydrological processes of soil–vegetation systems, the studies on biological soil crusts was also summarized, and related theoretical models of restoration based on the water balance was reviewed

Abstract: Taking I-Magic Cube - the entry of Shandong Jianzhu University for Solar Decathlon 2013 - for example, the paper analyzes the conceptual, technological, functional and aesthetic solutions of zero energy solar house coping with ecological, economic, social and cultural challenges. Passive strategies, the solar thermal and PV system, and the building integrated solar system tactics are elaborated. Abstract: Maintainability is an important character which is given by product design process. The maintainability design criteria and measure index used in product maintainability analysis are summarized and discussed in this paper. A product maintainability design method is studied by integrating the product feature model, maintainability design criteria with measure index. Product feature model can be built on the basis of the product feature library quickly. Product feature library for steam turbine design is created by using SolidWorks design library origination structure. A methodology which supports the design and development of product maintainability design support tool (PMDSTs) is put forward. The function of PMDSTs is designed by using UML (Unified Modeling Language) use case diagram, it is developed by using VC++ 6.0. The maintainability analysis application case of steam turbine-generator system is given at last.
