Piper sarmentosum inhibits ICAM-1 and Nox4 gene expression in oxidative stress-induced human umbilical vein endothelial cells

Abstract: HUVECs were divided into four groups:- control; treatment with 180 μM hydrogen peroxide (H2O2); treatment with 150 μg/mL AEPS and concomitant treatment with AEPS and H2O2 for 24 hours. Total RNA was extracted from all the groups of HUVEC using TRI reagent. Subsequently, qPCR was carried out to determine the mRNA expression of NF-κB, VCAM-1, ICAM-1, E-selectin, Nox4, SOD1, CAT and GPx. The specificity of the reactions was verified using melting curve analysis and agarose gel electrophoresis.When stimulated with H2O2, HUVECs expressed higher level of ICAM-1 (1.3-fold) and Nox4 (1.2-fold) mRNA expression. However, AEPS treatment led to a reduction in the mRNA expression of ICAM-1 (p < 0.01) and Nox4 (p < 0.05) in the H2O2-induced HUVECs. AEPS also upregulated the mRNA expression of SOD1 (p < 0.05), CAT (p < 0.01) and GPx (p < 0.05) in oxidative stress-induced HUVECs. There was no significant change in the mRNA expression of VCAM-1 and E-selectin.The expressional suppression of ICAM-1 and Nox4 and induction of antioxidant enzymes might be an important component of the vascular protective effect of AEPS.Atherosclerosis has been recognized as a chronic inflammatory disease and oxidative stress plays a pivotal role in its initiation and progression [1]. Endothelial dysfunction is considered to be an early marker for atherosclerosis [2]. Evidence suggests that increased production of reactive oxygen species (ROS) and vascular inflammation play important roles in endothelial dysfunction.Vascular disorders, through over expression of adhesion molecules and cytokines are involved in the development of atherosclerosis. Endothelial cells in human atherosclerotic lesions have increased cell adhesion molecules expression such as intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1) and endothelial selectin (E-selectin) [3,4]. Abstract: objective: nitric oxide produced by endothelial nitric oxide synthase (enos) possesses multiple anti-atherosclerotic properties. hence, enhanced expression of enos and increased nitric oxide levels may protect against the development of atherosclerosis. piper sarmentosum is a tropical plant with antioxidant and anti-inflammatory activities. this study aimed to investigate the effects of piper sarmentosum on the enos and nitric oxide pathway in cultured human umbilical vein endothelial cells (huvecs). methods: huvecs were divided into four groups: control, treatment with 180 μm hydrogen peroxide (h2o2), treatment with 150 μg/ml aqueous extract of piper sarmentosum, and concomitant treatment with aqueous extract of ps and h2o2 for 24 hours. subsequently, huvecs were harvested and enos mrna expression was determined using qpcr. the enos protein level was measured using elisa, and the enos activity and nitric oxide level were determined by the griess reaction. results: human umbilical vein endothelial cells treated with aqueous extract of piper sarmentosum showed a marked induction of nitric oxide. treatment with ps also resulted in increased enos mrna expression, enos protein level and enos activity in huvecs. conclusion: aqueous extract of piper sarmentosum may improve endothelial function by promoting no production in huvecs.

Abstract: The large number of operations involving noisy machinery associated with construction site activities result in considerable variation in the noise levels experienced at receiver locations. This paper suggests an approach to predict noise levels generated from a site by using a Monte Carlo approach. This approach enables the determination of details regarding the statistical uncertainties associated with noise level predictions or temporal distributions. This technique could provide the basis for a generalised prediction technique and a simple noise management tool.
Anti-Diabetic Activity of Ferula assafoetida Extract in Normal and Alloxan-Induced Diabetic Rats

Abstract: The aim of the present study was to evaluate the possible anti-diabetic potential of Asafetida extract against the pancreatic β-cells damage from alloxan-induced diabetes in rats and some hormones related to diabetes mellitus. Asafetida induced significant reduction in blood glucose and increasing of serum insulin, our data indicate that the level of glucose in the animals that subjected with alloxan was 10.28 ±0.85 mmol L-1 p<0.05 comparing with control group 3.27±0.25 p<0.05, the level of blood glucose in diabetic group when subjected with Asafetida extract decreasing to 6.75±0.31 p<0.05. There was significant amount of insulin secretion in diabetic animals when subjected with Asafetida extract 0.48±0.05 p<0.05 in comparison with diabetic animal’s 0.33±0.06 p<0.05. These findings suggested that Asafetida extract treatment exerts therapeutic protective effect in diabetes by preserving pancreatic β-cells integrity and significant activity extract, which supports traditional usage to prevent diabetic complications.

Kaedah Berlelar Tidak Tersirat Kumpulan Berselang-seli (TTKS) digunakan kerana kaedah ini mempunyai tugas-tugas yang berasingan dan merdeka yang sesuai dilaksanakan dalam komputer selari. Willingness to Pay for Domestic Water Service Improvements in Selangor, Malaysia: A Choice Modeling Approach

Abstract: : The tasks and responsibilities of domestic water service management in Malaysia are handled by various government agencies. Sufficient water service and resources management is required for sustainable water resources conservation. In order to realized water resource conservation, economic effectiveness of water utilization (consumers), maintenance of water quality supply (source of water supply) and efficiency in allocating water resources (agencies) needs to be addressed. The objective of the study is to assess community preferences and values relating to alternative water service management with particular concentration on water service improvement. This study has applied Choice Experiment (CE) to investigate the current policies and potential alternative of water service management in Selangor. The studies are based on 230 respondents in Subang Jaya that were randomly interviewed for data collection in October to December 2008. The findings indicates that respondents are willing to pay higher for drinking water as compared to the current rate with improved in water quality (WQ), reduced the frequency of water interruption (WI) and increases in the consumer trust to tap water (CT). The finding of this study is very important in order to assists and recommends the policy makers towards efficiency of domestic water service management in Malaysia. Abstract: In mammalians, brain Limbic System is responsible for emotionalprocess. An important part of motor learning as well as attention isformed by the system. Brain Emotional Learning (BEL) process ofmammalians has been mathematically modeled. Based on the model,a controller architecture called Brain Emotional Learning BasedIntelligent Controller (BELBIC) has been developed. EmotionalLearning is a powerful methodology in real time control anddecision systems due to low computational complexity and fasttraining where the gradient based methods and evolutionaryalgorithms are hard to be applied because of their highcomputational complexity. In this paper, the emotional processesand the Limbic System will be reviewed. In addition, the utilizationof BELBIC model in variety of applications of control system will beexamined. The results of simulations demonstrate control systemsbased on BELBIC have good performances.

Abstract: A total of 57 Listeria species strains (19 isolates of Listeria monocytogenes and 38 isolates of Listeria innocua ) isolated from different location in the state of Selangor (Seri Serdang, Seri Kembangan, Kajang and Bangi) were characterized by using RAPD-PCR fingerprinting technique. Primer GEN15002 and GEN15009 were chosen whereby it produced reproducible and typeable results in L. monocytogenes (19) and L. innocua (38) isolates examined with the bands sizes ranging from 0.25 to 3.0 kilobase pairs. From the current study, it can be concluded that the RAPD profiles revealed a wide variability and high level of DNA sequence diversity within Listeria species strains tested. RAPD showed a very discriminatory power since several RAPD profiles were obtained among L. monocytogenes and L. innocua isolates and even slight difference between isolates can be detected.
