Enhanced inhibition of Avian leukosis virus subgroup J replication by multi-target miRNAs

Abstract: In this study, the avian leukosis virus subgroup J (ALV-J) proviral genome, including the gag genes, were treated as targets for RNAi. Four pairs of miRNA sequences were designed and synthesized that targeted different regions of the gag gene. The screened target (i.e., the gag genes) was shown to effectively suppress the replication of ALV-J by 19.0-77.3%. To avoid the generation of escape variants during virus infection, expression vectors of multi-target miRNAs were constructed using the multi-target serial strategy (against different regions of the gag, pol, and env genes). Multi-target miRNAs were shown to play a synergistic role in the inhibition of ALV-J replication, with an inhibition efficiency of viral replication ranging from 85.0-91.2%.The strategy of multi-target miRNAs might be an effective method for inhibiting ALV replication and the acquisition of resistant mutations.Avian leukosis (AL) is the general term for a variety of neoplastic diseases of poultry caused by the Alpharetrovirus, Avien leukosis virus (ALV). ALV has been classified into 10 subgroups, designated A-J. The subgroup J virus (ALV-J) is a relatively new strain of ALV that was isolated from Dorking fowl in the early 1990s [1]. ALV is an RNA virus with a genome of approximately 7.6 kb. The proviral genome of ALV-J contains three major genes, gag, pol, and env, which encode the viral structural proteins, RNA-dependent DNA polymerase, and the envelope glycoprotein, respectively.RNA interference (RNAi) is a simple and effective tool for silencing target genes that involves endogenous or exogenous double-stranded RNA (dsRNA)-mediated degradation of the specific mRNA sequences. Abstract: In my research, the water supply network's macro model was established by NN method based on the corresponding theory and it was applied successfully to the dynamic simulation of the network's working states. Furthermore, a simulation software with great practicability for the water supply network's working states was developed through the integrative application of many computer technologies. The results showed that this model and the software were more precise and simpler than the methods before and they were proper to the application in water supply corporations.

Abstract: Taking account of the drawbacks in dynamicity and low latency of the existing MAC protocols for wireless sensor networks, an adaptive MAC protocol (AMAC) based on clustering was put forward. In this protocol, every member within a cluster could apply to the cluster head for slot according to its state. The cluster head arbitrated this application information, and then adjusted the frame time size in time to make it more suitable for the topology and traffic load of current network. The simulation results show that AMAC outperforms other MAC protocols in the real-time and mobile wireless sensor networks.
An Overall Ltcc Package Solution for X-Band Tile T/R Module

Abstract: An overall Low-Temperature Co-fired Ceramics (LTCC) package solution for X-band T/R module has been presented in this paper. This tile type package contributes to a dramatic reduction in size and weight of the T/R module. Moreover, an obvious merit of ceramic housing is better consistency of Coefficient of Thermal Expansion (CTE), compared with the traditional combination of ceramic board and metal housing. The schematic diagram and 3-D structure of the T/R module have been presented and a novel vertical interconnection based on Ball Grid Array (BGA) has been proposed to connect vias in the lid and those in the stage of the main LTCC pan. The LTCC T/R module has been fabricated and measured. It is compact in size (20×20× 2.6 mm) and has a weight of 3.5 g. The measured transmit output power is 33±1 dBm in the frequency range from 8.8 GHz to 10.4 GHz, and the measured receive gain and Noise Figure are 29-30.5 dB and 2.6-2.8 dB, respectively.

Abstract: Random amplified polymorphic DNA (RAPD) was used in analysing the relationship amongtwenty-six speices of Amanita from Mangshan,Hunan. of 40 primers, 6 ones generated repreducible RAPDprofiles, and each one produced 1-10 bands. Two primers (OPG15 and OPH04) could differentiate 26 species ofAmanita by its RAPD profiles. The similarity coefficients obtained from profiles genelated by six primers amongAmanita were about 20-60%. A dendrogram was constructed using average linkage clustering for clusteringanalysis. It indicated that there were tWo groups in 26 species and some species with ring and membrmousvolva clustered a group which coincided with morphological classification.

Abstract: -鹅膏毒(环)肽和二羟鬼笔毒(环)肽是剧毒的鹅膏菌和其它几种致死毒菌中由一些修饰氨基酸组成的环肽毒素。由于-鹅膏毒肽对真核生物的mRNA合成的专一性抑制和和二羟鬼笔毒肽对肌动蛋白的专一性束缚,因而它们在分子生物学和细胞学研究中具有重要应用,对其需求逐步增加。为此,作者使用了一种改良的毒素提取方法,以制备高效液相色谱从灰花纹鹅膏菌中分离制备-鹅膏毒肽和二羟鬼笔毒肽,并通过紫外吸收光谱和质谱进行鉴定,表明-鹅膏毒肽和二羟鬼笔毒肽的分离效果好,纯度高。本方法对其它毒菌中的-鹅膏毒肽和二羟鬼笔毒肽的分离制备具有同样的应用价值。

Comparison of Psychological Symptoms and Serum Levels of Neurotransmitters in Shanghai Adolescents with and without Internet Addiction Disorder: A Case-Control Study

Abstract: Background Internet addiction disorder (IAD) is now recognized internationally and is known to be linked with academic and social impairment. To date, we know little about its associated main biological factors. This study aimed to collect a carefully defined group of adolescents with IAD and an age- and gender-matched typically developing comparison group. We hypothesized that the young people with IAD would have higher rates of self-reported anxiety and depressive symptoms, have altered levels of peripheral blood dopamine, norepinephrine and serotonin. In addition, we hypothesized the hours spent online are correlated with the severity of depression and anxiety among these young people with IAD. Methodology/Principal Finding A cross-sectional study of 20 adolescents who met Beard’s criteria for IAD and 15 typically developing adolescents (comparison group) was conducted. All the participants completed the Self Rating Depression Scale (SDS), Self Rating Anxiety Scale (SAS), and the Screen for Child Anxiety Related Emotional Disorders (SCARED). Peripheral blood dopamine, serotonin and norepinephrine were assayed. The mean level of norepinephrine was lower in the IAD group than that in the typically developing participants, while dopamine and serotonin levels did not differ. The SDS, SAS and SCARED symptom scores were increased in the adolescents with IAD. A logistic regression analysis revealed that a higher SAS score and lower level of norepinephrine independently predicted IAD group membership. There was no significant correlation between hours spent online and scores of SAS/SDS in IAD group. Conclusions/Significance Increased self-reported anxiety and lower peripheral blood norepinephrine are independently associated with IAD.
Synovium Fragment-Derived Cells Exhibit Characteristics Similar to Those of Dissociated Multipotent Cells in Synovial Fluid of the Temporomandibular Joint

Abstract: Multipotent mesenchymal stem cells (MSCs) found in the synovial fluid (SFMSCs) of the tempromandibular joint (TMJ) remain poorly understood. During TMJ arthrocentesis, we discovered that synovial fluid collected from some patients with TMJ disorders contained not only SFMSCs but also synovium fragments (SFs). In this study, we attempted to characterize both the SFMSCs and SF-derived cells (SFCs) in order to further understand the role of MSCs in the synovial fluid of the TMJ. The SFs were membranous and translucent and consisted of several cell layers, indicating that their origin was only from the intima. SFCs were obtained by digestion of the SFs and subsequently expanded in vitro. SFMSCs were enriched by centrifugation of the synovial fluid and expanded in vitro. SFCs and SFMSCs displayed a similar fibroblast-like, spindle-shaped morphology, and we observed that some SFMSCs grew out of small tissue masses in culture. Flow cytometric analysis showed that both groups of cells expressed similar surface markers, including CD90, CD44, CD105, and CD73. However, both were negative for Stro-1, CD146, CD45, CD34, CD11b, CD19, and HLA-DR. Immunofluorescent staining showed that both SFs and SFMSCs expressed vascular cell adhesion molecule 1. Both SFCs and SFMSCs could be induced to differentiate down osteogenic, chondrogenic, adipogenic, and neurogenic lineages in vitro. Together, our results indicate that the intima is the most likely tissue origin of SFMSCs in the TMJ. Moreover, the SFs are composed of only intima and thus offer an improved source of synovium-derived MSCs compared to synovium specimens obtained by surgery, which contain both intima and subintima.
