Influence of Aloe polysaccharide on proliferation and hyaluronic acid and hydroxyproline secretion of human fibroblasts in vitro

Abstract: Objective: To investigate the effect of Aloe polysaccharide on proliferation and hyaluronic acid and hydroxyproline secretion of human fibroblasts in vitro. Methods: The fibroblasts were treated with different doses of polysaccharide (0, 25, 50, 100, 200, 400 mg/L). Subsequently, cell proliferation was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, cell cycle by flow cytometry, evaluation of the Aloe polysaccharide toxic effect by acridine orange-ethidium bromide staining, evaluation of the cell injury by lactate dehydrogenase (LDH) assay, and the collagen synthesis by 3H-proline incorporation. In addition, hyaluronic acid and hydroxyproline levels in the supernatants of cultured fibroblasts were measured by enzyme-linked immunosorbent assay. Results: The proliferation of fibroblasts was induced with polysaccharide in a dose-dependent manner, reaching its highest level on 5th day. Meanwhile, the percentage of cells at phase G0/G1 was decreased, while that at phases G2/M and S was increased significantly in Aloe polysaccharide-treated groups as compared with those in the control group (P<0.05). Additionally, the apoptosis of the fibroblasts showed no differences among all groups. The collagen synthesis was increased and cell injury decreased in polysaccharide-treated groups as compared with those in control group (P<0.05), while the levels of hyaluronic acid and hydroxyproline in the supernatants of fibroblasts treated with polysaccharide were significantly higher than those in the control group (P<0.05).Conclusion: The Aloe polysaccharide promotes both the proliferation of fibroblasts and the production of hyaluronic acid and hydroxyproline in fibroblasts. This indicates that the Aloe polysaccharide may play an important role in the extracellular matrix remodeling during wound healing.
A Study on the Relationship between Well-Being and Turnover Intentions among Rural School Teachers: School Organizational Climate as a Moderating Variable  [PDF]

Abstract: This study examined the effect of teacher well-being and the organizational climate in rural elementary schools on teachers’ turnover intentions, as well as the effect of the interaction between teacher well-being and organizational climate on teachers’ turnover intentions. Teachers from rural elementary schools in Nantou County, Taiwan, were the participants in this study. A questionnaire was distributed and surveys were collected from 254 teachers. SPSS statistical software was used to conduct confirmatory factor analysis, reliability analysis, descriptive statistics analysis, independent-samples t-tests, one-way ANOVA, Pearson’s correlation analysis, and hierarchical regression analysis. The findings in this study were as follows: 1) Male and female teachers in rural elementary schools showed a significant difference in the professional sharing construct of school organizational climate. 2) Teachers of different age, marital status, part-time position, years of service, and native land showed significant differences in the well-being constructs of life satisfaction and negative emotions. 3) Teachers of different marital status showed a significant difference in the school organizational climate constructs of work supervision, teaching constraints, and turnover intentions. 4) Teachers from different places showed a significant difference in the estrangement construct. 5) The influence of teacher well-being on turnover intentions was negatively affected by the school organizational climate constructs of support, work supervision, and comradeship. The constructs of teaching constraints and estrangement had a significant and positive effect on turnover intentions. Thus, school organizational climate had a moderating effect on the relationship between teacher well-being and turnover intentions.

Abstract: 摘要: 针对现有张量影响力模型未能充分考虑用户的时间特征以及在线学习等问题,提出了一种融合时间特征的社交媒介用户影响力分析方法。该方法首先将用户观点、活跃度、网络中心度等特征加入张量模型中,并将张量分解过程中的用户潜在特征矩阵加入时间特征约束;其次,采用随机梯度下降的方法进行张量的分解;最后,通过融合不同张量片的影响力信息得到用户影响力得分。该方法的优点是能够快速分解张量并准确预测特定话题领域下的用户社会影响力,同时能够在已有模型参数的基础上进行新数据的在线训练。实验结果表明,与现有TwitterRank、OOLAM、受限非负张量分解模型等相比, 该方法在平均预测准确率上提升了2%~6%。同时,该方法的时间消耗仅为受限非负张量分解模型的30%~50%。
Abstract: Since both the temporal characteristics and online learning are not fully considered in exsiting tensor influence models, a novel method with temporal characteristics is proposed in this paper. This method constructs tensor with users opinion, activity and network centrality information. Then, a factorizes tensor with stochastic gradient descent algorithm which is constrained by temporal characteristics matrix is deployed in our model. Base on these two steps above, this method calculates user influence by combining different slices of tensor in the end. The advantages of this method are that it can decompose tensor efficiently and satisfy the need of online learning. Experimental results show that the average accuracy of the proposed method is 2% to 6% better than the baseline method such as TwitterRank, OOLAM and constrained nonnegative tensor factorization method. Besides, the running time of the proposed method is only 30% to 50% of constrained nonnegative tensor factorization method

Abstract: Detection of sulfur-fumigated Paeoniae Alba Radix (PAR) in different complex preparations is challenging due to the relatively lower content of PAR and interference from more complicated components in complex preparations with different multiple constituent herbs. In this study, a high performance liquid chromatography- triple-quadrupole tandem mass spectrometry method was developed for detecting sulfur-fumigated PAR in different complex preparations. Paeoniflorin, the major component of PAR, and paeoniflorin sulfonate, the characteristic artifact transformed from paeoniflorin during sulfur-fumigation of PAR, were used as chemical markers. Multiple reaction monitoring (MRM) scan was employed to maximize sensitivity and selectivity. Through optimizing full mass scan and daughter ion scan conditions, two mass transitions were selected and employed respectively for unequivocal identification of paeoniflorin and paeoniflorin sulfonate. The detection limits for paeoniflorin and paeoniflorin sulfonate using MRM were much lower than those detected with UV 270 nm. Paeoniflorin and paeoniflorin sulfonate could be simultaneously detected in different commercial PAR-containing complex preparations without interference of other components using the established method, indicating that the newly established method was selective and sensitive enough for screening sulfur-fumigated PAR in commercial complex preparations.

Abstract: 为观察桑叶总黄酮的降糖作用及其对抗氧化酶、蛋白糖基化的影响.用桑叶总黄酮(8 g生药/kg体重)灌胃四氧嘧啶诱导的糖尿病大鼠4周,观察给药前后血糖变化,测定LPO、SOD、GSH-px及果糖胺水平;分别以0.001,0.01,0.1 g/L桑叶总黄酮浓度加入体外蛋白糖基化体系孵育1,4,7,14,21,28d,测定早期糖化蛋白产物的浓度.结果表明:与对照组比较,桑叶总黄酮组有明显降血糖作用.桑叶总黄酮组血糖由(18.33±3.33)mmol/L降至(10.19±4.45)mmol/L,P<0.05,LPO含量由(10.51±2.31)nmol/L,降至(6.01±2.04)nmol/L,P<0.05,SOD水平由(471.1±103.0)Nu/mL,升至(678.7±31.5)Nu/mL,P<0.001,果糖胺水平由(6.22±1.37)mmol/L,降至(1.70±0.32)mmol/L,P<0.05,体外糖化蛋白产量孵育时间呈正相关,0.1 g/L桑叶总黄酮在各时间点均能使糖化蛋白生成量降低.因此:桑叶总黄酮能显著降低糖尿病大鼠血糖、LPO含量并提高SOD水平,抑制蛋白糖基化.

Abstract: The system studied in the paper is one of the sub-systems of the 2010'Shanghai Expo Transportation System. It is one of the most important basic guarantees for safe and rapid converging and dispersing of visitors for the Shanghai Expo to locate scientifically transportation interchanges in the Expo Arena. The paper generally introduces the 2010'Shanghai Expo, analyses the characteristics of the visitors for the 2010'Shanghai Expo and gives the definition of Transportation Interchange. An index-system for evaluating alternatives of the transportation interchanges is introduced. The alternatives of the transportation interchanges can be arranged in order of importance by using AHP and would be altered according to the decision maker with different budgets.

Abstract: 介绍了一种用于航天立体摄影的大视场准远心光学系统,其光组结构形式为复杂化的双高斯型.主要光学参量为f′=23.3 mm;2ω=37.8°;F/数=F/5;MTF均值达到0.8时,对应的空间分辨率大于36lp/mm;光谱适应范围为0.5 ~0.75μm.在实验室进行了模拟推扫,合成了三维立体图像.模拟试验结果表明该光学系统满足航天立体摄影的要求.

Abstract: A kind of wide wave band, collimated optical system was designed, which the aperture locates in the front focus. In this design, auto-corrected chromatic aberration, long work distance, front aperture and collimated rays were concentrated in one. In this thesis, all these were discussed. Glasses partner were selected in Chinese, it is very cheap, and after correct the chromatic, the secondary spectrum of this wide wave band optical system also were corrected. In the same time, the system realized the long work distance both in its front and back side. The result show that the residual secondary spectrum of the system is less than one thousandth focal length, and the parallel error of rays of the wavefront less than 0.000 22 rad.
