Wrinkled 1 (WRI1) Homologs, AP2-Type Transcription Factors Involving Master Regulation of Seed Storage Oil Synthesis in Castor Bean (Ricinus communisL.)  [PDF]

Abstract: Among APETALA2 (AP2)-type plant specific transcription factor family, WRINKLED1 (WRI1), has appeared to be a master gene transcriptionally regulating a set of carbon metabolism- and fatty acid synthesis (FAS)-related genes responsible for seed specific triacylglycerols (TAGs) storage in oil plants. B3 type transcription factors, such as ABI3 and FUS3, are known to be involved in seed development, such as seed storage protein synthesis and maturation. Based on the recent whole genome sequence data of castor bean (Ricinus communis L.), putative WRI1 homologs (RcWRI1, RcWRI2) specifically expressed in castor bean seed have been identified by comparing organ specific expression profiles among seed development-related transcription factors, seed storage specific genes (Ricin, RcOleosin) and a set of FAS genes including genes for sucrose synthase (RcSUS2), biotin carboxyl carrier protein (a subunit of acetyl-CoA carboxylase, RcBCCP2) and ketoacyl-acyl carrier protein synthase (RcKAS1). Immunoreactive signals with WRI1, FUS3 and ABI5-related polypeptides were also detected in seed specifically, consistent with the expression profiles of seed development-related genes. The WRI1 binding consensus sites, [CnTnG](n)(7)[CG], designated as the AW-box, were found at the promoter region of RcBCCP2 and RcKAS1. Thus, RcWRI1 possibly play a pivotal role in seed specific TAGs storage during seed development by directly activating FAS -related genes.

Abstract: Silencing of gene expression by RNA interference (RNAi) has become a widely used tool. For the study of mammalian gene function expression vectors for short hairpin RNA (shRNA) were developed. However the standard methods of shRNA transgenic (Tg) mice production have not been established. Sry (sex-determining region on the Y chromosome) is a mammalian sex-determining gene on the Y chromosome. In mice, the transient expression of Sry in supporting cell precursor cells between 10.5 and 12.5 days post-coitus (dpc) triggers the differentiation of Sertoli cells from granulosa cells. Then high efficiency of Sry gene silencing in Tg mice should induce XY male-to-female sex reversal. An shRNA Tg mouse targeting Sry gene was attempted to be generated by pronuclear microinjection. A low rate (Tg pups/all pups born after microinjection = 2/154 to 7/178) of Tg pups was observed. These Tg mice showed no XY male-to-female sex reversal. The results suggest that exogenous expression of small RNA might exert a negative effect on embryonic development and another approach should be needed for RNAi transgenesis in mice.

Abstract: Background: Hyaluronic acids act upon keratinocytes via CD44 receptors and regulate proliferation and differentiation. Some cosmetic hyaluronan lotions manufactured based upon the fact are nowadays available. Aims: To evaluate a cosmetic hyaluronan lotion (Dr. Fukaya’s skin repair lotion or Hyaluprotect) from the viewpoint of anti-aging effects and to consider its mechanism. Patients/Methods: In ten healthy volunteers at the age over 60, immunohistochemical research of the biopsied skin was performed before and after the application of the hyaluronan lotion for two weeks. Results: Expression of PCNA in the lower epidermis increased in 8 of 10 subjects. Filaggrin expression of the upper epidermis increased in 6 subjects. 11βHSD1 decreased in 5 subjects and 11βHSD2 increased in 5 subjects. Conclusions: The proliferative and differentiative effects of the hyaluronan lotion upon keratinocytes were confirmed immunohistologically. There is a possibility that hyaluronic acids work through the regulation of corticosteroids by 11βHSD1 and 11βHSD2.

Abstract: Natural vitamin E is suggested to have an antioxidant function. However, the synthetic form of vitamin E, DL-tocopherol, which has been widely used in topical ointments, may cause allergic contact dermatitis. Here, we report a case of allergic contact dermatitis with erythema multiforme-like eruption caused by a topical ointment. Patch testing indicated a positive allergic reaction to an anti-inflammatory ointment the patient had been using and its ingredient, DL-alpha-tocopheryl acetate (vitamin E). In addition, a positive reaction to glycyrrhetinic acid was observed. Both vitamin E and glycyrrhetinic acid are useful ingredients of topical applications. However, the possibility that both can cause contact dermatitis, albeit rarely, should be considered.
Reconstruction of the Lacustrine Delta and Lake Level Change Analyzing Subsurface Geology and Geomorphology: Changes That Occurred during the Holocene in the Oguraike Reclaimed Land Area, Southern Kyoto, Japan  [PDF]

Abstract: A paleo-lacustrine delta in Kyoto, Japan was reconstructed on the basis of subsurface geological and geomorphological analysis, and paleo-lake level changes were estimated from the structure of the delta. These analyses of the study region, i.e., the Oguraike reclaimed land area provided evidence that Lake Ogura existed until about 60 years ago in southern Kyoto, Japan. The Uji river delta was provided influents to this lake until ca. 400 years ago, as is indicated by an upward-coarsening delta succession of about 2 - 4 m thickness. The lake level could also have changed in the past as a result of a change in altitude of the delta-front (foreset) and delta-plain boundary, which probably reflects the lake surface elevation. About 400 years ago, the Paleo-Uji River was separated from Ogura Lake because a levee was constructed along the river for building a castle and for constructing a waterway for transportation. As a result of this construction, the lake level that was more than 13.0 m in elevation was reduced by 1.5 m. In a more ancient times, the lake level experienced two stages—one in which the elevation was more than 13.5 m, and one in which the elevation was reduced to less than 10 m. These changes in the lake level are represented by a flat surface with four steps and small cliff of height ca. 0.5 - 2 m (relative elevation) separating them, recognized at the southern lakeshore. The observation of strata along with the archaeological survey in the north of Ogura Lake reveals that the lake level was decreased ca. 800 - 680 years ago. The lake level was at its highest during two periods, the first from before the 8th century to the end of the 8th century and the second from the 14th century to 400 years ago.

Abstract: A 73-year-old male patient suffered from very late stent thrombosis occurred 6 years after sirolimuseluting stent (SES) implantation in the ostial and proximal left anterior descending coronary artery (LAD). He presented emergently with cardiogenic shock and emergent coronary angiography showed thombus in the ostial stent and in the ostial left circumflex artery. Optical coherence tomography found delayed healing on the ostial stent. Acetylcholine provocation test had also shown severe provoked coronary spasm in all coronary arteries 28 months after SES implantation which suggested the association of severe coronary endothelial dysfunction as a potential mechanism of very late stent thrombosis.

Abstract:

This report attempts to ascertain the current state of, and outstanding issues relating to, nursing diagnosis, as taught in nursing education in Japan, and to obtain basic resources that will allow the improvement of said nursing diagnosis training. A self-completed, anonymous survey was carried out in regard to teaching staff responsible for classes in “nursing process” or “nursing diagnosis” at 183 university institutions involved in nursing education nationwide. Responses were received from 82 people, which clarified the following three points. 1) Of the 63 universities teaching nursing process as an independent subject, approximately 62% included nursing diagnosis. 2) A diverse range of educational materials were used in nursing diagnosis training, including NANDA-I nursing diagnosis. 3) In implementing nursing process using nursing diagnosis, issues raised included the following: the limitations of education “on paper”, using theoretical patients, insufficient skills among teaching staff, the difficulty of realizing practical training in a clinical setting, and the tendency to try to make a simple diagnosis fit the circumstances. In the future, this study suggests that it may be important to create a set of guidelines guaranteeing a minimum level of educational content in relation to nursing diagnosis, which must be learned before graduation.

Potentiality of Mindfulness Art Therapy Short Version on Mood of Healthy People  [PDF]

Abstract: The present study examined the potential of Mindfulness Art Therapy Short version on mood of healthy people. The Mindfulness Art Therapy Short version consisted of two sessions and included factors of both mindfulness and art therapy. The Art Therapy alone consisted of two sessions including factors of art therapy. Seventeen college students received the Mindfulness Art Therapy Short version and twenty-two students received Art Therapy alone. All participants completed the Profile of Mood States pre- and post-interventions in each session. In the Mindfulness Art Therapy Short version group, scores for Tension-Anxiety, Depression-Dejection, and Fatigue-Inertia reduced and those for Vigor-Activity significantly increased after the program. In the Art Therapy group, although scores for Tension-Anxiety and Fatigue-Inertia decreased, the scores of Depression-Dejection and Vigor-Activity did not significantly change. The score of Total Mood significantly decreased only in the Mindfulness Art Therapy Short version. These results suggest that the Mindfulness Art Therapy Short Version may be more affective for improving mood, particularly depression and vigor, compared to Art Therapy alone.

Abstract: Argentine ant invasion changed colour-polymorphic composition of ant-mimetic jumping spider Myrmarachne in southwestern Japan. In Argentine ant-free sites, most of Myrmarachne exhibited all-blackish colouration. In Argentine ant-infested sites, on the other hand, blackish morph decreased, and bicoloured (i.e. partly bright-coloured) morphs increased in dominance. Invasive Argentine ant drives away native blackish ants. Disappearance of blackish model ants supposedly led to malfunction of Batesian mimicry of Myrmarachne.
Familiar and Unfamiliar Face Recognition in a Crowd  [PDF]

Abstract:

The purpose of the present study was to investigate whether the visual search process for familiar faces differs from that for unfamiliar faces. We used a single-target visual search task and recorded eye movements of participants during the task. We employed three different types of face familiarity: personally familiar faces (friends and teachers), famous faces, and unfamiliar faces. Participants had to search through arrays of faces for a target face. In each trial, a target face and three distractor faces were horizontally aligned and presented at the same time. In the personally familiar or famous condition, the target was a personally familiar face or a famous face, respectively, and the distractor faces were unfamiliar. In the unfamiliar condition, the target was an unfamiliar face and the distractor faces were personally familiar. The reaction times to identify the target demonstrated that the visual search for unfamiliar faces is slower than that for personally familiar faces and famous faces, but there was no significant difference between reaction times to recognize the personally familiar and the famous face targets. Additionally, the eye movement results in the unfamiliar face condition showed that an exhaustive search of the entire array occurred more frequently than a self-terminating search, but this was not true for both the personally familiar faces and famous faces conditions. These results suggest that the visual search process for familiar faces (personally familiar and famous) in a crowd differs from that for unfamiliar faces.
