Ohmic Heating for Tofu Making—A Pilot Study  [PDF]

Abstract:

The aim of this study is to explore the relationship between temperature and electrical conductivity of soya milk under ohmic heating in tofu making. The soya milk of 10 Brix was heated to a steady temperature of 90?C. The applied voltage was increased and the temperature rising rate was investigated for adequate heating profiles in tofu making. Experimental results showed that the electrical conductivity of soya milk is proportional to the heating time. The temperature rising rate was increased from 1.46?C to 3.82?C/min as a result of increased voltage. Hence ohmic heating could be an efficient, convenient heating measure in tofu making.

Abstract: Indoor air quality has received increased attention. Surveying human thermal comfort and evaluating indoor airflow and ventilation can improve the integrity assessment of the indoor environment. In this study, indoor air quality measurements and spatial assessments were performed in a convenience store on a campus. The computational fluid dynamics model was used to simulate the indoor airflow and explore the spatial distribution of the CO2 concentration. The results showed the CO2 concentration easily surpassed the standard value during the weekdays and accumulated in short flows and dead zones. In the condition of no-building alterations, the preferred improvement strategy is to modify the air supply angle to 30°.
University Student Sample Is Unable to Accurately Assess Their Calorie Needs: Implications for Weight Management and Menu Labeling  [PDF]

Abstract: National surveys have shown that over 80% of adults do not know their recommended calorie levels. Lack of knowledge about calorie needs could be contributing to the high prevalence of obesity in the US. Young adulthood is a crucial period for the development of dietary behaviors that continue into later adulthood and influence the risk of obesity and chronic disease. This study examined university students’ knowledge of their recommended calorie needs. Subjects (N = 153) were students at Texas Tech University in Fall 2010. Students were given a survey to assess perceived daily calorie need (PDCN) and perceived daily calorie intake (PDCI). Their recommended daily calorie needs (RDCN) and actual calorie intakes (ACI) were determined using MyPyramid.gov. PDCN, PDCI, RDCN, and ACI were compared to determine students’ ability to accurately estimate and consume recommended daily calorie levels. The range of their PDCN was 120 kcal to 10,000 kcal. Only 19.7% of students estimated their RDCN accurately. There were significant differences between PDCN and RDCN (t [152] = ?3.223, P = 0.002); PDCI and ACI (t [114] = 3.246, P = 0.002); and ACI and RDCN (t [114] = ?5.6, P = 0.000). Nearly 40% of these university students were overweight. BMI had a significant effect (P = 0.001) on students’ estimation of their RDCN as students with underweight/normal BMI were more accurate. Nutrition education programs focused on calorie needs should be implemented with university students so they will be able to effectively use calorie information on food labels and menus for weight management.

Abstract: BICEP2's detection on the primordial B-mode of CMB polarization suggests that inflation occurred around GUT scale, with the tensor-to-scalar ratio r~0.2. Inspired by this discosvery, we study the topological inflation which was driven by a double/single/no well potential. We show that with proper choice of parameters, all these three types of topological inflationary models could be consistent with the constraints from current observations.
Novel Nuclear Localization and Potential Function of Insulin-Like Growth Factor-1 Receptor/Insulin Receptor Hybrid in Corneal Epithelial Cells

Abstract: Background Type I insulin-like growth factor receptor (IGF-1R) and insulin receptor (INSR) are highly homologous molecules, which can heterodimerize to form an IGF-1R/INSR hybrid (Hybrid-R). The presence and biological significance of the Hybrid-R in human corneal epithelium has not yet been established. In addition, while nuclear localization of IGF-1R was recently reported in cancer cells and human corneal epithelial cells, the function and profile of nuclear IGF-1R is unknown. In this study, we characterized the nuclear localization and function of the Hybrid-R and the role of IGF-1/IGF-1R and Hybrid-R signaling in the human corneal epithelium. Methodology/Principle Findings IGF-1-mediated signaling and cell growth were examined in a human telomerized corneal epithelial (hTCEpi) cell line using co-immunoprecipitation, immunoblotting and cell proliferation assays. The presence of Hybrid-R in hTCEpi and primary cultured human corneal epithelial cells was confirmed by immunofluorescence and reciprocal immunoprecipitation of whole cell lysates. We found that IGF-1 stimulated Akt and promoted cell growth through IGF-1R activation, which was independent of the Hybrid-R. The presence of Hybrid-R, but not IGF-1R/IGF-1R, was detected in nuclear extracts. Knockdown of INSR by small interfering RNA resulted in depletion of the INSR/INSR and preferential formation of Hybrid-R. Chromatin-immunoprecipitation sequencing assay with anti-IGF-1R or anti-INSR was subsequently performed to identify potential genomic targets responsible for critical homeostatic regulatory pathways. Conclusion/Significance In contrast to previous reports on nuclear localized IGF-1R, this is the first report identifying the nuclear localization of Hybrid-R in an epithelial cell line. The identification of a nuclear Hybrid-R and novel genomic targets suggests that IGF-1R traffics to the nucleus as an IGF-1R/INSR heterotetrameric complex to regulate corneal epithelial homeostatic pathways. The development of novel therapeutic strategies designed to target the IGF-1/IGF-1R pathway must take into account the modulatory roles IGF-1R/INSR play in the epithelial cell nucleus.
The Influence of Tobacco Smoking on the Relationship between Pressure and Flow in the Middle Cerebral Artery in Humans

Abstract: Background Cigarette smoking is associated with an increased risk of stroke but the mechanism is unclear. The study examined whether acute and chronic cigarette smoking alters the dynamic relationship between blood pressure and cerebral blood flow. We hypothesised that acute and chronic smoking would result in a cerebral circulation that was less capable of buffering against dynamic fluctuations in blood pressure. Further, these changes would be accompanied by a reduction in baroreflex sensitivity, which is reduced after smoking (acute smoking). Methods We recruited 17 non-smokers and 15 habitual smokers (13 ± 5 pack years). Continuous measurements of mean cerebral blood flow velocity (transcranial Doppler ultrasound), blood pressure (finger photoplethysmography) and heart rate enabled transfer function analysis of the dynamic relationship between pressure and flow (gain, normalised gain, phase and coherence) and baroreflex sensitivity during supine rest before and after smoking a single cigarette (acute smoking). Results There were no between-group differences in gain, phase or coherence before acute smoking. However, both groups showed a reduction in gain and coherence, associated with a reduction in baroreflex sensitivity, and increase in phase after acute smoking. Conclusions Contrary to our hypothesis, these findings suggest that in the face of a reduction in baroreflex sensitivity acute smoking may potentially improve the ability of the cerebral circulation to buffer against changes in blood pressure. However, chronic smoking did not alter the dynamic relationship between blood pressure and cerebral blood flow velocity. These results have implications on understanding mechanisms for attenuating stroke risk.
CISA: Contig Integrator for Sequence Assembly of Bacterial Genomes

Abstract: A plethora of algorithmic assemblers have been proposed for the de novo assembly of genomes, however, no individual assembler guarantees the optimal assembly for diverse species. Optimizing various parameters in an assembler is often performed in order to generate the most optimal assembly. However, few efforts have been pursued to take advantage of multiple assemblies to yield an assembly of high accuracy. In this study, we employ various state-of-the-art assemblers to generate different sets of contigs for bacterial genomes. A tool, named CISA, has been developed to integrate the assemblies into a hybrid set of contigs, resulting in assemblies of superior contiguity and accuracy, compared with the assemblies generated by the state-of-the-art assemblers and the hybrid assemblies merged by existing tools. This tool is implemented in Python and requires MUMmer and BLAST+ to be installed on the local machine. The source code of CISA and examples of its use are available at http://sb.nhri.org.tw/CISA/.
Identification of Cytotoxic T Lymphocyte Epitopes on Swine Viruses: Multi-Epitope Design for Universal T Cell Vaccine

Abstract: Classical swine fever (CSF), foot-and-mouth disease (FMD) and porcine reproductive and respiratory syndrome (PRRS) are the primary diseases affecting the pig industry globally. Vaccine induced CD8+ T cell-mediated immune response might be long-lived and cross-serotype and thus deserve further attention. Although large panels of synthetic overlapping peptides spanning the entire length of the polyproteins of a virus facilitate the detection of cytotoxic T lymphocyte (CTL) epitopes, it is an exceedingly costly and cumbersome approach. Alternatively, computational predictions have been proven to be of satisfactory accuracy and are easily performed. Such a method enables the systematic identification of genome-wide CTL epitopes by incorporating epitope prediction tools in analyzing large numbers of viral sequences. In this study, we have implemented an integrated bioinformatics pipeline for the identification of CTL epitopes of swine viruses including the CSF virus (CSFV), FMD virus (FMDV) and PRRS virus (PRRSV) and assembled these epitopes on a web resource to facilitate vaccine design. Identification of epitopes for cross protections to different subtypes of virus are also reported in this study and may be useful for the development of a universal vaccine against such viral infections among the swine population. The CTL epitopes identified in this study have been evaluated in silico and possibly provide more and wider protection in compared to traditional single-reference vaccine design. The Application of Data Mining Techniques and Multiple Classifiers to Marketing Decision

Abstract: Nowadays, the capability to collect data has been expanded enormously and provides enterprises huge amount of data. The interesting knowledge or the high-valued information about the customer can be extracted by data mining. By following the market segmentation strategy, an enterprise could increase the expected profits. However, as for the enterprise, the customer’s basic data including some demographic variables and geographic variables are easier to obtain than the behavior data of customers. The customer value may be predicted through the customer’s basic data. Followed by taking marketing activities to those customers with high value, the enterprises could avoid unnecessary marketing cost. The research constructed a marketing decision model which utilized the demographic and geographic variables as input of three individual classifiers - BP network, decision tree, and Mahalanobis distance - to predict a new customer’s value. In order to improve the accuracy of prediction, this study combined three classifiers to predict new customer’s value through the different combining methods. The results show the multiple classifier system combined by BP has the best prediction accuracy.
Quantification Bias Caused by Plasmid DNA Conformation in Quantitative Real-Time PCR Assay

Abstract: Quantitative real-time PCR (qPCR) is the gold standard for the quantification of specific nucleic acid sequences. However, a serious concern has been revealed in a recent report: supercoiled plasmid standards cause significant over-estimation in qPCR quantification. In this study, we investigated the effect of plasmid DNA conformation on the quantification of DNA and the efficiency of qPCR. Our results suggest that plasmid DNA conformation has significant impact on the accuracy of absolute quantification by qPCR. DNA standard curves shifted significantly among plasmid standards with different DNA conformations. Moreover, the choice of DNA measurement method and plasmid DNA conformation may also contribute to the measurement error of DNA standard curves. Due to the multiple effects of plasmid DNA conformation on the accuracy of qPCR, efforts should be made to assure the highest consistency of plasmid standards for qPCR. Thus, we suggest that the conformation, preparation, quantification, purification, handling, and storage of standard plasmid DNA should be described and defined in the Minimum Information for Publication of Quantitative Real-Time PCR Experiments (MIQE) to assure the reproducibility and accuracy of qPCR absolute quantification.
