Aquaporin 1-expressing MCF-7 mammary carcinoma cells show enhanced migration in vitro  [PDF]

Abstract: Recent studies have demonstrated that aquaporin (AQP) expression facilitates cell migration and promotes angiogenesis and neutrophil motility. Migration of tumor cells is a crucial step in tumor invasion and metastasis. Here we investigated the expression of AQP in MCF-7 human mammary carcinoma cells and characterized its function in cell migration. Reverse Transcription–Polymerase Chain Reaction, Immunoblot and Immunofluorescence analysis demonstrated two populations of MCF-7 cell clones with low (AQP1L) and high (AQP1H) AQP1 expression and the AQP1 protein expression patterns in the plasma membrane of MCF-7 cells. MCF-7 cell clones (AQP1L and AQP1H) with low and about two-fold higher osmotic water permeability were identified by functional assays with corresponding low and high AQP1 expression. Cell migration rate was remarkably higher in AQP1H cells as compared to AQP1L cells, assessed by wound healing and transwell migration assays. Adenoviral-mediated mRNA and protein expression of AQP1 in AQP1L cells increased their water permeability and migration rate to the level similar to AQP1H cells. The results provided direct evidence that aquaporin-mediated plasma membrane water permeability played an important role in mammary carcinoma cell migration and may be associated with mammary carcinoma invasion and metastasis.
Business Negotiation Culture in China - A Game Theoretic Approach

Abstract: There are numerous researches on the relationship between culture and business activities. Based on the Chinese negotiating culture, this paper utilizes game theory modeling method to summarize the major style of negotiations between Chinese businessmen and foreign businessmen. First, this paper analyzes the impact of culture on business negotiations, including priorities issues, negotiation strategy and negotiating style. Second, the paper puts forward the concept of “acculturation” in the analysis of cultural adaptability of Chinese businessmen, and it also clarifies the general as well as the potential reasons for the low acculturation for Chinese businessmen. Etiquettes for general business communications in China are also discussed in detail. In theoretical part, the paper analyzes negotiating sytle of Chinese businessmen with cultural characteristics. Due to the strategic feature of this model, a specific game theoretical model is built, and then a specific example of Chinese and foreign business price negotiations is used to show the applicability of the model. Finally, the article states the conclusions and direction for further research.

Abstract: This paper develops a measure of “linguistic distance” between languages, which is used as a dummy variable in Gravity Model. By this model, the paper discusses the trade relationships of Canada and China with those countries which speak English as the first language and as the second language. The results show that the proportion of the population that use English as second language will be more important factor than those use English as second language for the bilateral trade with Canada and China. Considering the specification error in this model, it can also reach the same results. Key words: Linguistic distance; Bilateral trade; Gravity model Résumé Cet article développe une fa on d’indiquer la “distance linguistique” en tant que variable fictive dans le modèle de gravité. A travers les analyses sur le modèle, nous discutons la relation commerciale de la Chine et du Canada entre les pays où l’anglais est la langue maternelle et ceux où l’anglais est la langue seconde. Selon les résultats, du point de vue de la régression du commerce bilatéral, comme un facteur principal, la proportion de l’utilisation de l’anglais comme la langue seconde est plus importante que les facteurs dans les pays où l’anglais est la langue maternelle. Parametrizations of canonical bases and irreducible components of nilpotent varieties

Abstract: It is known that the set of irreducible components of nilpotent varieties provides a geometric realization of the crystal basis for quantum groups. For each reduced expression of a Weyl group element, Gei{\ss}, Leclerc and Schr\"{o}er has recently given a parametrization of irreducible components of nilpotent varieties in studying cluster algebras. In this paper we show that their parametrization coincides with Lusztig's parametrization of the canonical basis.

Abstract: With aqueous ammonia in the process of CO2 absorption and desorption to join sodium bicarbonate, the influence of HCO3- on CO2 absorption and desorption from ammonia solution was investigated through the experimental analysis of the desorption quantity of CO2, desorption rate, CO2 loading and the absorption rate. The experimental results showed that, in experimental conditions, The desorption rate decreased gradually with increasing ammonia concentrations. The desorption rate increased 12%, 17%, 19% and 28.8% when 1 mol/L of ammonia solution is added in 0.1 mol/L, 0.3 mol/L, 0.5 mol/L and 1 mol/L of sodium bicarbonate. The higher concentration of ammonium bicarbonate solution which was added sodium bicarbonate,the more observably the effect of CO2 desorption was promoted. The absorption rate had dropped when absorption process added 0.3 mol/L sodium bicarbonate, the CO2 loading was a little change.

Abstract: The asparagine-proline-alanine sequences (NPA motifs) are highly conserved in aquaporin water channel family. Crystallographic studies of AQP1 structure demonstrated that the two NPA motifs are in the narrow central constriction of the channel, serving to bind water molecules for selective and effi- cient water passage. To investigate the importance of the two NPA motifs in the structure, function and biogenesis of aquaporin water channels, we generated AQP1 mutations with NPA1 deletion, NPA2 de- letion and NPA1,2 double deletion. The coding sequences of the three mutated cDNAs were subcloned into the mammalian expression vector pcDNA3.1 to form expression plasmids. We established stably transfected CHO cell lines expressing these AQP1 mutants. Immunofluorescence indicated that all the three mutated AQP1 proteins are expressed normally on the plasma membrane of stably transfected CHO cells, suggesting that deletion of NPA motifs does not influence the expression and intracellular processing of AQP1. Functional analysis demonstrated that NPA1 or NPA2 deletion reduced AQP1 water permeability by 49.6% and 46.7%, respectively, while NPA1,2 double deletion had little effect on AQP1 water permeability. These results provide evidence that NPA motifs are important for water per- meation but not essential for the expression, intracellular processing and the basic structure of AQP1 water channel.
Mn-Rich Nanostructures in : Fabrication, Microstructure, and Magnetic Properties

Abstract: Magnetic semiconductors have attracted extensive attention due to their novel physical properties as well as the potential applications in future spintronics devices. Over the past decade, tremendous efforts have been made in the diluted magnetic semiconductors (DMS) system, with many controversies disentangled but many puzzles unsolved as well. Here in this paper, we summarize recent experimental results in the growth, microstructure and magnetic properties of Ge-based DMSs (mainly ), which have been comprehensively researched owing to their compatibility with Si microelectronics. Growth conditions of high-quality, defect-free, and magnetic bulks, thin films, ordered arrays, quantum dots, and nanowires are discussed in detail. 1. Introduction Spin-based electronics research in the 1990s boosted with lower power consumption and greater integration comparing with the current electronics based on charge [1, 2]. Among various potential material systems, diluted magnetic semiconductors (DMSs), possessing benefits from both the charge and the spin of electrons, are considered to be most promising. In particular, Mn-doped Ge DMSs discovered in the early 2000s [3, 4] have been extensively investigated [5–8] due mainly to their compatibility with existing complementary metal-oxide-semiconductor (CMOS) technologies. The fabrication of Ge-based DMS ( ) was firstly reported by Park et al. [4], with Curie temperature ( ) increasing linearly with the Mn concentration up to 116?K for . Since then, many modified or other methods have been put forward in order to obtain a high-quality, room-temperature (RT) ferromagnetic GeMn DMS with high Mn concentration. One of the key issues to achieve ideal GeMn DMSs is to uniformly dope Mn into the Ge matrix. However, undesirable Mn-rich clusters or precipitates, such as Mn5Ge3 [9, 10] and Mn11Ge8 [7, 11], commonly occur. Although Mn5Ge3 and Mn11Ge8 phases are also ferromagnetic, both of them have metallic character, considerably hampering the discovery of intrinsic magnetic properties of the GeMn nanostructures and complicating their use as spin injectors.
