Effects of Astragalus Polysaccharide on Immune Responses of Porcine PBMC Stimulated with PRRSV or CSFV

Abstract: Background Astragalus polysaccharide (APS) has been used as an immunomodulator that can enhance immune responses, whereas the immunomodulatory effects of APS on porcine peripheral blood mononuclear cells (PBMCs) exposed to porcine reproductive and respiratory syndrome virus (PRRSV) and classical swine fever virus (CSFV) have not been investigated. Methodology/Principal Findings Porcine PBMCs were cultured in complete RPMI media in the presence of the R98-strain of PRRSV (5×104 TCID50/ml) or C-strain of CSFV (103 TCID50/ml) with or without APS. The expression of mRNA for CD28, cytotoxic T-lymphocyte antigen 4 (CTLA-4), transforming growth factor-β (TGF-β), interleukin 2 (IL-2) and IL-10 was assayed by TaqMan real-time RT-PCR. The expression of mRNA for CD28 and CTLA-4 increased at 24 h after stimulation of PBMCs with CSFV and the increased production of CTLA-4 was confirmed by western blot analysis, whereas the increases were inhibited by the addition of APS. In addition, APS alone upregulated IL-2 and TGF-β mRNA expression in PBMCs and the addition of APS had the capacity to prevent a further increase in IL-2 mRNA expression in PBMCs during CSFV or PRRSV infection, but had no effect on TGF-β mRNA expression. The production of tumor necrosis factor-alpha (TNF-α) increased at 12 h after stimulation with PRRSV or CSFV, but not with PRRSV plus APS or CSFV plus APS, whereas the addition of APS to PBMCs infected with PRRSV or CSFV promoted IL-10 mRNA expression. Conclusions We suggested that APS had immunomodulatory effects on cells exposed to PRRSV or CSFV. It might be that APS via different mechanisms affects the activities of immune cells during either PRRSV or CSFV infection. This possibility warrants further studies to evaluate whether APS would be an effective adjuvant in vaccines against PRRSV or CSFV.
Energy-Saving Potential of Building Envelope Designs in Residential Houses in Taiwan

Abstract: The key factors in the energy-saving design of a building’s exterior in Taiwan are the thermal performance of the roof and window glazing. This study used the eQUEST software to investigate how different types of roof construction, window glasses and sunshield types affect the energy consumption in residential buildings under common scenarios. The simulation results showed that the use of an appropriate window glass significantly reduced the annual energy consumption, followed by the shading device, whereas the roof construction produced less of an energy-efficiency benefit. By using a low-E glass and a 1.5 × 1.5 m box shading (e.g., balcony), this could save approximately 15.1 and 13.6% of the annual electricity consumption of air conditioners, respectively. Therefore, having control over the dominant factors in the building envelope is indeed an important step in the path to achieving energy savings and carbon reduction in residential houses.
Algorithm to improve long patent sentence machine translation

Abstract: This paper presented a method using sentence segmentation to improve the performance of long patent sentence translation. In this method, segmented the long Chinese sentence into separated short sentences using some semantic features from the hierarchical network of concepts theory (HNC theory). Integrated the segmentation algorithm with a rule-base MT system. After integrating, the BLEU value (0.1898) of MT system was improved about 30%. The experiment results indicate the proposed method can improve the performance of patent sentence translation effectually.
Risks Associated with High-Dose Lactobacillus rhamnosus in an Escherichia coli Model of Piglet Diarrhoea: Intestinal Microbiota and Immune Imbalances

Abstract: Probiotic could be a promising alternative to antibiotics for the prevention of enteric infections; however, further information on the dose effects is required. In this study, weanling piglets were orally administered low- or high-dose Lactobacillus rhamnosus ACTT 7469 (1010 CFU/d or 1012 CFU/d) for 1 week before F4 (K88)-positive Escherichia coli challenge. The compositions of faecal and gastrointestinal microbiota were recorded; gene expression in the intestines was assessed by real-time PCR; serum tumour necrosis factor-α (TNF-α) concentrations and intestinal Toll-like receptor 4 (TLR4) were detected by ELISA and immunohistochemistry, respectively. Unexpectedly, high-dose administration increased the incidence of diarrhoea before F4+ETEC challenge, despite the fact that both doses ameliorated F4+ETEC-induced diarrhoea with increased Lactobacillus and Bifidobacterium counts accompanied by reduced coliform shedding in faeces. Interestingly, L. rhamnosus administration reduced Lactobacillus and Bifidobacterium counts in the colonic contents, and the high-dose piglets also had lower Lactobacillius and Bacteroides counts in the ileal contents. An increase in the concentration of serum TNF-α induced by F4+ETEC was observed, but the increase was delayed by L. rhamnosus. In piglets exposed to F4+ETEC, jejunal TLR4 expression increased at the mRNA and protein levels, while jejunal interleukin (IL)-8 and ileal porcine β-defensins 2 (pBD2) mRNA expression increased; however, these increases were attenuated by administration of L. rhamnosus. Notably, expression of jejunal TLR2, ileal TLR9, Nod-like receptor NOD1 and TNF-α mRNA was upregulated in the low-dose piglets after F4+ETEC challenge, but not in the high-dose piglets. These findings indicate that pretreatment with a low dose of L. rhamnosus might be more effective than a high dose at ameliorating diarrhoea. There is a risk that high-dose L. rhamnosus pretreatment may negate the preventative effects, thus decreasing the prophylactic benefits against potential enteric pathogens. Our data suggest a safe threshold for preventative use of probiotics in clinical practice.
A Novel Image Coding Algorithm Based on Coefficients of Interest

Abstract: A novel image coding algorithm based on coefficients of interest is proposed in terms of the special needs of remote-sensing images. When coding rate is given, the entropy estimate is calculated from the coefficients of the given image, so that the last coding bit-plane can be derived when the image is coded normally. The difference of the algorithm from the one for region-of-interest algorithm lies in that the region and the scaled value need not be preappointed, and the most important is that all the coefficients in bands are not scaled up, while the coefficients of interest and the scaled value are deduced from the given image in the algorithm. This algorithm can be thought as the generalization from the region-of-interest algorithm, which is smarter and more valid than the region of interest coding algorithm. The simulation and the experimental results show that the small targets in the remote-sensing images can be saved validly while the whole image quality decreases a little.
Efficient FP-STREAM Algorithm Based on Vertical Compression Data Format

Abstract: 近年来由于信息的爆炸式增长,数据流频繁模式挖掘逐渐成为研究的热点。FP-Stream作为经典的数据流频繁模式的挖掘算法,实现了多时间粒度的挖掘,但是该算法并未对数据本身进行压缩,使其在一定时间内处理的数据量受到限制,存在有限内存和高速海量数据的矛盾。通过对数据流进行垂直和Dif-bits压缩变换来改进FP-Stream算法,大大降低了内存需求,提高了数据处理能力。经过实验证明,改进算法是有效的。
Effects of a Green Space Layout on the Outdoor Thermal Environment at the Neighborhood Level

Abstract: This study attempted to address the existing urban design needs and computer-aided thermal engineering and explore the optimal green space layout to obtain an acceptable thermal environment at the neighborhood scale through a series of building energy and computational fluid dynamics (CFD) simulations. The building-energy analysis software eQUEST and weather database TMY2 were adopted to analyze the electric energy consumed by air conditioners and the analysis results were incorporated to derive the heat dissipated from air conditioners. Then, the PHOENICS CFD software was used to analyze how the green space layout influences outdoor thermal environment based on the heat dissipated from air conditioners and the solar heat reemitted from the built surfaces. The results show that a green space located in the center of this investigated area and at the far side of the downstream of a summer monsoon is the recommended layout. The layouts, with green space in the center, can decrease the highest temperature by 0.36 °C.

Abstract: Time,cost and quality are the three main control objects in construction project.The objects are undeterministic because of the undeterministic construction conditions,systematic analysis and fuzzy tradeoff optimization for all objects are very important during engineering management.This paper firstly presents definition of the fuzzy object membership,then presents the fuzzy tradeoff optimization model for time-cost-quality using fuzzy multi-attribute group decision utility function theory.Applying multi-model network planning techniques and self-adaptive genetic algorithms,the most satisfied decision results can be obtained.Finally,a case study is given to verify the effectness of the model and the feasibility of solution method.
