Chronic Heat Stress Weakened the Innate Immunity and Increased the Virulence of Highly Pathogenic Avian Influenza Virus H5N1 in Mice

Abstract: Chronic heat stress (CHS) can negatively affect immune response in animals. In this study we assessed the effects of CHS on host innate immunity and avian influenza virus H5N1 infection in mice. Mice were divided into two groups: CHS and thermally neutral (TN). The CHS treatment group exhibited reduced local immunity in the respiratory tract, including the number of pulmonary alveolar macrophages and lesions in the nasal mucosa, trachea, and lungs. Meanwhile, CHS retarded dendritic cells (DCs) maturation and reduced the mRNA levels of IL-6 and IFN- significantly (<.05). After the CHS treatment, mice were infected with H5N1 virus. The mortality rate and viral load in the lungs of CHS group were higher than those of TN group. The results suggest that the CHS treatment could suppress local immunity in the respiratory tract and innate host immunity in mice significantly and moderately increased the virulence in H5N1-infected mice.
A Routing Strategy with Link Disruption Tolerance for Multilayered Satellite Networks  [PDF]

Abstract: Link disruption has a considerable impact on routing in multilayered satellite networks, which includes predictable disruption from the periodic satellite motion and unpredictable disruption from communication faults. Based on the analysis on the predictability of satellite links, a link disruption routing strategy is proposed for multilayered satellite networks, where, a topology period is divided into non-uniform slots, and a routing table in each slot is calculated by the topology predictability of satellite networks, and a congestion control mechanism is proposed to ensure the reliable transmission of packets, and a flooding mechanism is given to deal with the routes selection in the case of unpredictable link disruption. This routing strategy is implemented on the satellite network simulation platform, the simulation results show that the strategy has less delay and higher link utilization, and can meet the routing requirements of multilayered satellite networks.

Abstract: Promotions are commonly used marketing tools for enterprises; the effectiveness of promotional posters is related to the success or failure of promotional activities. This paper embarks from two major elements of poster-promotional information and background color, and studies the effects of different promotional frames and background colors on the perceived value of consumer activity. This study divides the promotional framework into positive promotional framework and negative promotion framework. Through two experiments, the paper discusses how the matching of promotional frame and background color affects consumers value perception of promotional activities, and studies the mediating effect of perceptual diagnosis on the relationship, and the moderating effect of individual control sense on the relationship. The conclusion of the experiment is that the match between the promotional frame and the background color has an interactive effect on the value perception of the promotional activities. The experiment second verifies the existence of this interaction effect and verifies the mediating effect of perceptual diagnostics.
Acute, Multiple-Dose Dermal and Genetic Toxicity of Nu-3: A Novel Antimicrobial Agent

Abstract: Nu-3 [butyl-phosphate-5-thymidine-3-phosphate-butyl] is a modified nucleotide that has been shown to have antimicrobial activity against a range of bacteria including Pseudomonas aeruginosa. However, data on the toxicological profile of Nu-3 are still lacking. In the present study, the toxicity of Nu-3 was evaluated by the following studies: acute oral toxicity, dermal and mucous membrane irritation, multiple-dose toxicity and genotoxicity in vivo and vitro. The acute oral toxicity test in mice showed that Nu-3 had an LD50 of 2001mg/kg body weight. The irritation tests on rats revealed that Nu-3 was not irritant, with an irritation scoring of 0. The multiple-dose toxicity study in rats showed that Nu-3 did not cause significant changes in histology, selected serum chemistry, and hematological parameters compared to the controls. Rats administrated with multiple-doses of Nu-3 showed no visible toxic symptoms. Both in vitro and in vivo, Nu-3 exhibited no notable genetic toxicity. Overall, the data suggest that Nu-3 is hypotoxic or nontoxic antimicrobial compound that warrants being further developed for treating Pseudomonas aeruginosa infection.
Phylogenetic and Pathotypical Analysis of Two Virulent Newcastle Disease Viruses Isolated from Domestic Ducks in China

Abstract: Two velogenic Newcastle disease viruses (NDV) obtained from outbreaks in domestic ducks in China were characterized in this study. Phylogenetic analysis revealed that both strains clustered with the class II viruses, with one phylogenetically close to the genotype VII NDVs and the other closer to genotype IX. The deduced amino acid sequence of the cleavage site of the fusion (F) protein confirmed that both isolates contained the virulent motif 112RRQK/RRF117 at the cleavage site. The two NDVs had severe pathogenicity in fully susceptible chickens, resulting in 100% mortality. One of the isolates also demonstrated some pathogenicity in domestic ducks. The present study suggests that more than one genotype of NDV circulates in domestic ducks in China and viral transmission may occur among chickens and domestic ducks.
Apoptosis and Pro-inflammatory Cytokine Response of Mast Cells Induced by Influenza A Viruses

Abstract: The pathogenesis of the influenza A virus has been investigated heavily, and both the inflammatory response and apoptosis have been found to have a definitive role in this process. The results of studies performed by the present and other groups have indicated that mast cells may play a role in the severity of the disease. To further investigate cellular responses to influenza A virus infection, apoptosis and inflammatory response were studied in mouse mastocytoma cell line P815. This is the first study to demonstrate that H1N1 (A/WSN/33), H5N1 (A/Chicken/Henan/1/04), and H7N2 (A/Chicken/Hebei/2/02) influenza viruses can induce mast cell apoptosis. They were found to do this mainly through the mitochondria/cytochrome c-mediated intrinsic pathway, and the activation of caspase 8-mediated extrinsic pathway was here found to be weak. Two pro-apoptotic Bcl-2 homology domain 3 (BH3) -only molecules Bim and Puma appeared to be involved in the apoptotic pathways. When virus-induced apoptosis was inhibited in P815 cells using pan-caspase (Z-VAD-fmk) and caspase-9 (Z-LEHD-fmk) inhibitors, the replication of these three subtypes of viruses was suppressed and the secretions of pro-inflammatory cytokines and chemokines, including IL-6, IL-18, TNF-α, and MCP-1, decreased. The results of this study may further understanding of the role of mast cells in host defense and pathogenesis of influenza virus. They may also facilitate the development of novel therapeutic aids against influenza virus infection.
Using One-Dimensional Variational Assimilation Algorithm to Obtain Atmospheric Refractive Index from Ground-Based GPS Phase Delay

Abstract: GPS信号相位延迟中包含很多大气信息,通过一定的方法可以从中获取有价值的大气参数。基于地基GPS相位延迟数据,提出了一种结合经验模式的一维变分同化获取大气折射率的方法。利用GPS相位延迟模拟数据进行同化试验,讨论了背景误差的设置对同化结果的影响,采用实测个例对该方法进行了验证。结果表明,该一维变分同化方法可行,并可获取高精度的0~60 km大气折射率。低层大气背景误差设置的准确程度会对整个高度范围内的同化效果产生影响。首次将同化获取的大气折射率应用于无线电波折射修正,取得了很好的修正效果,修正精度可达1 mm量级
Efficient induction of CD25- iTreg by co-immunization requires strongly antigenic epitopes for T cells

Abstract: In the present study, we demonstrated the requirement of highly antigenic epitopes for CD25- iTreg induction. Firstly, we showed that the induction of CD25- iTreg by tolerogenic DC can be blocked by anti-MHC-II antibody. Next, both the number and the suppressive activity of CD25- iTreg correlated positively with the overt antigenicity of an epitope to activate T cells. Finally, in a mouse model of dermatitis, highly antigenic epitopes derived from a flea allergen not only induced more CD25- iTreg, but also more effectively prevented allergenic reaction to the allergen than did weakly antigenic epitopes.Our data thus indicate that efficient induction of CD25- iTreg requires highly antigenic peptide epitopes. This finding suggests that highly antigenic epitopes should be used for efficient induction of CD25- iTreg for clinical applications such as flea allergic dermatitis.The inducible regulatory T cells, or iTreg, differ from the naturally regulatory T cells (nTreg) in that the former are generated in the periphery through encounter with environmental antigens. It is also believed that iTreg play non-overlapping roles, relative to nTreg, in regulating peripheral tolerance [1-3]. Most iTreg reported to date have been CD25+ cells (CD4+CD25+Foxp3+), and it is well established that their induction requires suboptimal stimulation of the T cell receptor (TCR) and cytokines TGF-β and IL-2 [3]. The CD25+ iTreg thus appear to derive primarily from weakly stimulated CD4+ T cells.We previously identified a different subset of iTreg in mice that is CD25- (CD4+CD25-Foxp3+ and IL-10+TGF-beta+IFN-γ-). The CD25- iTreg were induced after co-immunization using a protein antigen and a DNA vaccine encoding the same antigen [4-7]. Abstract: This paper analyzes and compares the purchasing power parity (PPP) and exchange rate worldwide based on the datum of International Comparison Program (ICP) in the year 2005 and 2011. The results indicate that developing countries have been experiencing more substantial currency devaluation, while currency appreciation occurs mainly in high-income countries. After analyzing the relative price levels for 2011 and per capita income, we find that the relative price level is above 1.5 in most countries worldwide, while the relative price level is between 0.5 and 1 in a few high-income countries. When the per capita income is around $3000, the peering relationships of purchasing power parity and exchange rates are the most obvious.

Abstract: This paper tests the effect of per capita income, exchange rate, foreign direct investment inflows, net trade condition index and the final consumption expenditure growth rate on the fluctuation of purchasing power parity basing on panel fixed effects model during the period of 2000-2013 of 62 countries (religions). Empirical results show that the fluctuation of per capita income and exchange rate are the key factors to explain the fluctuation of purchasing power parity, however, purchasing power parity (PPP) fluctuation is influenced by the five variables has some differences in terms of magnitude and direction between different economies. The effect of the foreign investment on purchasing power parity is negative in low income economies, but other economies are the opposite, at the same time, the estimated coefficient of high income is lower than upper & middle income economies. The growth of final consumption expenditure is helpful to improve the purchasing power parity of low income and high income economies. It is surprising that the increase in per capita income will reduce the purchasing power of some developed economies. In China, purchasing power parity will continue a growth trend in future.
