Quantitative Analysis of ATP Sulfurylase and Selenocysteine Methyltransferase Gene Expression in Different Organs of Tea Plant (Camellia sinensis)  [PDF]

Abstract: Tea plant (Camellia sinensis) has unique biological features for the study of cellular and molecular mechanisms, an evergreen broad-leaved woody plant which can accumulate selenium in soil abundant of Selenium. Expression of the genes related to Selenium (Se) metabolism is an adaptation to the soil environment for a long period. The purpose of the present study was to explore if there exist differences of expression about these genes in tea plant between growing in Selenium-abundant and normal soil. A quantitative real-time reverse transcription polymerase chain reaction (Q-RT-PCR) assay was done for quantification of ATP sulfurylase (APS) and selenocysteine methyltransferase (SMT) mRNA normalized to Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) gene in tea plant. Young leaves, mature leaves and tender roots from tea plants growing in soil abundant of Selenium were respectively obtained from Shitai County, Anhui Province, and also the relevant materials of the selenium un-enriched tea plant planted at agricultural garden of Ahui Agriculture University were taken as control for real-time PCR analysis. The results showed that APS1, APS2 and SMT expression levels for either young or mature leaves in selenium-enriched tea plant were lower than that in ordinary (selenium un-enriched) tea plant. In contrast, the APS1, APS2 and SMT expression level of roots in selenium-enriched tea plant were all higher than that in ordinary tea plant. APS1 gene expression level of roots in selenium-enriched tea plant was about 1.6 times higher than that in the ordinary tea plant, APS2 gene expression level was about 4.8-fold higher than that in the ordinary tea plant, SMT gene expression level was about 3.3 times higher than that in the ordinary tea plant. Among various tissues of selenium-enriched tea plant, APS1 gene relative expression level of young leaves was similar to or slightly higher than mature leaves, and the one of roots was the lowest among them; APS2 gene relative expression level of young leaves was similar to or slightly higher than the roots, and the one of mature leaves was the lowest among them; SMT gene relative expression level of young leaves was similar to or slightly higher than mature leaves, and the one of roots was the highest among them. Our results suggest that there existed correlation between selenium and expression levels of these genes.

Simultaneous Distillation Extraction of Some Volatile Flavor Components from Pu-erh Tea Samples—Comparison with Steam Distillation-Liquid/Liquid Extraction and Soxhlet Extraction

Abstract: A simutaneous distillation extraction (SDE) combined GC method was constructed for determination of volatile flavor components in Pu-erh tea samples. Dichloromethane and ethyl decylate was employed as organic phase in SDE and internal standard in determination, respectively. Weakly polar DB-5 column was used to separate the volatile flavor components in GC, 10 of the components were quantitatively analyzed, and further confirmed by GC-MS. The recovery covered from 66.4%–109%, and repeatability expressed as RSD was in range of 1.44%–12.6%. SDE was most suitable for the extraction of the anlytes by comparing with steam distillation-liquid/liquid extraction and Soxhlet extraction. Commercially available Pu-erh tea samples, including Pu-erh raw tea and ripe tea, were analyzed by the constructed method. the high-volatile components, such as benzyl alcohol, linalool oxide, and linalool, were greatly rich in Pu-erh raw teas, while the contents of 1,2,3-Trimethoxylbenzene and 1,2,4-Trimethoxylbenzene were much high in Pu-erh ripe teas.

Abstract: Analysis on land use coordination degree is instructive to land intensive use and land sustainable use. The coordination is established to analyze the land use coordination degree of Nanjing from 1999 to 2008; on the basis of this, coordination factors are analyzed by the multiple stepwise regression. The result shows that land use coordination degree in Nanjing has been in a rising trend in the recent ten years, but there is still a distance from the full coordination; we find that the main factors of land coordination use are population growth, economic development and the level of afforesting.

Abstract: While there is increasingly strategic importance in knowledge, it is facing new challenges to manage knowledge effectively. The paper studies the motivational construct model for knowledge sharing from the perspective of content theories and social motivation theories. The data collected by using the questionnaire survey from a variety of industries was analyzed on the basis of interviews and pretest. The results of exploratory factor analysis (EFA) and confirmatory factor analysis (CFA) showed the motivational construct model for knowledge sharing was comprised of existence motivation, relatedness motivation, growth motivation and norm motivation. The results extended the perspectives for knowledge sharing motivation and provided theoretical evidences for facilitating knowledge sharing behavior.
Multi-objective Optimization of Multi-Agent Elevator Group Control System Based on Real-time Particle Swarm Optimization Algorithm  [PDF]

Abstract: In order to get a globally optimized solution for the Elevator Group Control System (EGCS) scheduling problem, an algorithm with an overall optimization function is needed. In this study, Real-time Particle Swarm Optimization (RPSO) is proposed to find an optimal solution to the EGCS scheduling problem. Different traffic patterns and controller mechanisms for EGCS are analyzed. This study focuses on up-peak traffic because of its critical importance to modern office buildings. Simulation results show that EGCS based on Multi-Agent Systems (MAS) using RPSO gives good results for up-peak EGCS scheduling problem. Besides, the elevator real-time scheduling and reallocation functions are realized based on RPSO in case new information is available or the elevator becomes busy because it is unavailable or full. This study contributes a new scheduling algorithm for EGCS, and expands the application of PSO.

Then according to the target of “managerial” accounting talents cultivation mode, it proposes several suggestions for the university about how to cultivate the “managerial” accounting talents’ IT quality.
