DNA immunoadsorption for childhood-onset systemic lupus erythematosus  [PDF]

Abstract: We present a retrospective review of DNA immunoadsorption (DNA-IA) therapy on clinical symptoms as well as indicators in pediatric cases with systemic lupus erythematosus (SLE), and follow up the short-term curative effects. 16 SLE cases were treated by DNA-IA for 3 times every other day. We observed the changes on clinical manifestations and immunological indicators, in order to compare the alteration of these indicators including clinical manifestations, Systemic Lupus Erythematosus Disease Active Index (SLEDAI) scores, 24 hurinary protein excretion, autoantibodies, serum IgG and complement C3. 13 cases were followed up regularly, within 3 months after DNA-IA therapy, 12 cases of clinical manifestations improved (92.3%). SLEDAI scores in 10 cases decreased from (16.20 ± 12.54) to less than 5 (76.9%), 8 cases of ANA, anti-DNA antibodies were negative (61.5%), 13 cases with IgG level in serum recovered to normal (10.39 ± 4.38) g/L, C3 level rose to normal (1.06 ± 0.23) g/L. 3 to 6 months after IA, clinical manifestations and laboratory examinations in all cases got maximum improved. 9 months after IA, SLEDAI score in 2 cases (15.4%) rose to more than 5, anti-DNA antibody in 2 cases (15.4%) became positive, and 1case (7.7%) with serum C3 decreased again. 2 cases died from multiple organs dysfunction within 3 to 6 months after IA. No serious complications were found during DNA-IA. We recommend that DNA immunoadsorption is a safe and effective therapy for active childhood-onset SLE, which could improve clinical symptoms, eliminate ANA and anti-DNA antibodies. Combining with corticosteroids and immunosuppressive drugs, DNA-IA could significantly reduce the activity of disease and protect vital organs function in the short term.

Abstract: 利用模式生物秀丽隐杆线虫(Caenorhabditis elegans)研究了电晕法空气负离子(Electrically generated negative ai ions,ENIs)暴露的生物学效应.将成年野生型和mev-1突变型线虫及新鲜虫卵持续暴露于空气负离子中(空气负离子剂量1×105~1.5×105ions·cm-3),观察空气负离子暴露对线虫发育周期、寿命、繁殖性能和性腺细胞凋亡的影响.结果表明:空气负离子暴露可显著加速野生型和mev-1突变型线虫发育(p<0.05),缩短寿命(p<0.05),与野生型相比,空气负离子暴露对mev-1突变线虫寿命影响更大(p<0.05);空气负离子暴露可显著降低线虫后代数量(p<0.05),原因可能在于空气负离子暴露显著诱导性腺细胞凋亡的增加(p<0.05).以上结果表明,持续空气负离子暴露对线虫具有损伤作用,mev-1基因突变导致线虫对空气负离子暴露更加敏感.

Abstract: The tandem arginine biosynthetic genes argCJBDFR from Corynebacterium crenatum were amplified by PCR.Sequence analysis indicates that there are five open reading frames,including argJ,argB,argD,argF and argR in the 6080bp stretch.The amino acid sequences coded by argJBDFR genes show 92%,95%,96%,97.5% and 100% identity to those of Corynebacterium glutacimum,respectively.
Study of the Performance Sensitivity to SNR Mismatch in BICM-ID System

Abstract: The iterative decoding schemes such as BICM-ID and other concatenated coding systems require the knowledge of channel Signal-to-Noise Ratio (SNR). The effect of SNR estimation errors is studied for BICM-ID in the paper and the performance sensitivity of SNR mismatch for the log-APP decoding algorithm and its simplified implementations is analyzed. Analysis and simulation results show that the max-log-APP algorithm is robust against any SNR mismatch problem, while the log-APP algorithm and other simplified implementations are more tolerant of overestimation of SNR than underestimation, which is because they use different correction functions. The required SNR accuracy to prevent large performance degradation is increased with the increase of channel SNR, generally from -2dB to +6dB. Therefore, when there is no perfect channel state information, the max-log-APP algorithm is the optimum decoding algorithm for BICM-ID.
Relationship of polymorphism of megsin C2093T and C2180T with IgA nephrology:a Meta-analysis

Abstract: Objective To analyze the relation between the polymorphism of megsin C2093T and C2180T gene and IgA nephropathy.Methods Studies related to the relation of polymorphism of megsin C2093T and C2180T to IgA nephropathy were authenticated from databases including Medline,Embase and Cochrane Library from Jan.1980 to Aug.2010.The cited literature of obtained articles and some core English and Chinese medical journals relevant to nephropathy were also searched by hand(2000 to Aug.2010).The data were analyzed by RevMan 5.0 software.Results Five papers were finally included,meta-analysis results showed that there was statistical difference between megsin 2093C allele and risk for IgA nephropathy [odds ratio(OR) = 1.20,95% CI 1.04 to 1.40].No statistical difference existed between megsin 2093CC genotype and risk for IgA nephropathy(OR=1.18,95% CI 0.90 to 1.55).In Asian subgroups,there was significant difference between megsin 2093C allele and risk for IgA nephropathy(OR=1.19,95% CI 1.01 to 1.40),while no significant association existed between megsin 2093CC genotype and risk for IgA nephropathy(OR=1.19,95% CI 0.88 to 1.61).Also,no significant difference was revealed in the total meta-analysis between the megsin 2180C allele and risk for IgA nephropathy(OR=0.89,95%CI 0.72 to 1.10) or CC genotype and risk for IgA nephropathy(OR=1.04,95%CI 0.69 to 1.56).Moreover,no significant difference was found between megsin 2093 and 2180C allele and IgA nephropathy progression(OR=0.89,95%CI 0.69 to 1.15;OR=0.84,95%CI 0.48 to 1.46,respectively).There was also no significant difference between megsin 2093 and 2180CC genotype and IgA nephropathy progression(OR=0.80,95%CI 0.42 to 1.49;OR=0.79,95%CI 0.32 to 1.93,respectively).Significant difference was found between the 2093T-2180T haplotype and IgA nephropathy progression(OR=0.54,95%CI 0.33 to 0.87).Conclusion Megsin 2093C allele is the susceptible gene of IgA nephropathy,while megsin 2093 genotype,2180 allele or genotype are not.The 2093T-2180T haplotype plays a protective effect on the progression of IgA nephropathy.

A Meta Analysis on Risks of Adverse Pregnancy Outcomes in Toxoplasma gondii Infection

Abstract: Objective Quantified risks of congenital Toxoplasma gondii infection and abnormal pregnancy outcomes following primary maternal infection were evaluated with meta- analysis based on published studies. Methods The related literatures were searched in multiple literature databases regardless of languages. Odds ratio (OR) and 95% confidence interval (CI) were used to evaluate the risks of vertical transmission of Toxoplasma gondii and abnormal pregnancy outcomes following primary maternal infection with meta-analysis. Results 53 of the 2632 searched literatures were included in our analysis. The incidence of abnormal pregnancy outcomes in T. gondii infected pregnant women (infected group) was significantly higher than that in the uninfected pregnant women (control group) (OR = 5.10; 95% CI, 3.85–6.75). Toxoplasma gondii infection rate in the abnormal-pregnancy-outcome group was significantly higher than in the normal-pregnancy group (OR = 3.71; 95% CI, 3.31–4.15). The pooled rate of vertical transmission was 20% (95% CI, 15%–26%) in maternal infection of T. gondii. The incidences of vertical transmission in women who were infected in the first, second or third trimester of pregnancy were 5% (95%CI, 2%–16%), 13% (95%CI, 7%–23%), and 32% (95%CI, 24%–41%), respectively. The rates of vertical transmission in women who were treated with spiramycin-only, PSF (pyrimethamine + sulfadiazine + folinic acid) or PS (pyrimethamine + sulfadiazine) combined with spiramycin, or other untypical treatments were 13% (95%CI, 7%–22%), 13%(95%CI, 7%–25%), and 24%(95%CI, 18%–32%), respectively. Conclusions Toxoplasma gondii infection can result in adverse pregnancy outcomes in pregnant women. The pooled rate of vertical transmission was 20% in maternal infection and the incidences of vertical transmission increased in the first, second or third trimester of pregnancy. The pooled rates of transmission in groups treated with spiramycin-only, PSF or PS combined with spiramycin, or other untypical treatments were not significantly different.
