Effect of Variable Sulfur Supply on Thiol Compounds and Arsenic Accumulation in Kandelia Obovata (S., L.) Yong Seedlings  [PDF]

Abstract: In the present study, Kandelia obovata (S., L.) Yong seedlings were exposed to As V(0, 30, 60 and 150 mg kg-1) under different levels of sulfur (0, 0.1, 0.2 and 0.4% (w: w)) supply. A highly significant negative correlation between arsenic concentration in leaves with sulfur treatment (r=-0.554, P<0.01) is established. Concentration of arsenic in roots proves much higher than that in leaves. The GSH concentrations in K. obovata seedlings show significantly increases (rleave=0.648, rroot=0.602; P<0.01) with increasing sulfur supply in the growth media. Arsenic induced GSH formation in leaves of K. obovata seedlings, and clearly restrained GSH formation in roots. Arsenic exposure significantly increased the synthesis of non-protein thiols (NPT). Supply of sulfur at 0.1% and 0.2% promoted the content of NPT (P<0.05), but at 0.4% sulfur there was little effect on the NPT content (P>0.05). In conclusion, an increase in sulfur supply to plants may improve their accumulation capacity for arsenic through enhanced tolerance caused by a positive effect on the thiol metabolism of the plants.
A Survey on English Learning Motivation of Students in Qingdao Agricultural University

Abstract: A survey from students of both Advanced and Ordinary English classes in Qingdao Agricultural University shows that most of the students are motivated by both intrinsic and extrinsic motivations. However, intrinsic motivation is more important than extrinsic motivation in contributing to English learning. There is no difference in anxiety between students in Advanced English class and in Ordinary English class and the attitude of students in Advanced English class is more proper than that of students in Ordinary English class. Therefore, they are more active in English learning than the students in Ordinary English Class are. The higher autonomous learning ability they have, the better the achievement they get.
Gene cloning and expression of cadherin in midgut of Helicoverpa armigera and its Cry1A binding region

Abstract: Cadherins belong to one of the families of animal glycoproteins responsible for calcium-dependent cell-cell adhesion. Recent literatures showed that the cadherin-like in midgut of several insects served as the receptor of Bt toxin Cry1A and the variation of cadherin-like is related to insect’s resistance to Cry1A. The full-length cDNA encoding cadherin-like of Helicoverpa armigera is cloned by degenerate PCR and RACE techniques and the gene was designated as BtR-harm, which is 5581 bp in full-length, encoding 1730 amino acid residues (BtR-harm was deposited in GenBank and the accession number is AF519180). Its predicted molecular weight and isoelectric point were 195.39 kDa and 4.23, respectively. The inferred amino acid sequence includes a signal sequence, 11 cadherin repeats, a membrane-proximal region, a transmembrane region and a cytoplasmic region. Sequence analysis indicated that the deduced protein sequence was most similar to the cadherin-like from Heliothis virescens with 84.2% identity and highly similar to three other lepidopteran cadherin from Bombyx mori, Manduca sexta and Pectinophora gossypiella, with the sequence identities of 60.3.6%, 57.5% and 51.0%, respectively. The cDNA encoding cadherin gene was expressed successfully in E. coli and the recombinant proteins can bind with Cry1Ac. Truncation analysis and binding experiment of BtR-harm revealed that the Cry1A binding region was a contiguous 244-amino acid sequence, which located between amino acid 1217 and 1461. Semi-quantitative RT-PCR analysis showed that BtR-harm was highly expressed in midgut of H. armigera, very low expressed in foregut and hindgut and was not expressed in other tissues. After H. armigera producing resistance to Cry1Ac, the expression quantity of BtR-harm significantly decreased in midgut of H. armigera. It is the first confirmation that BtR-harm can function as receptor of Cry1Ac in H. armigera and the binding region was located on a contiguous 244 amino acid sequence, suggesting that the decrease of expression quantity of BtR-harm is one of the main reasons for H. armigera resistance to Cry1Ac.
“Climate effect” of the northeast cold vortex and its influences on Meiyu

Abstract: The Northeast Cold Vortex (NECV) is an important weather system in the middle and high latitudes in East Asia. Its time scale is synoptic, yet the frequent activities of NECV have significant “climate effect” which influences not only the monthly temperature in the lower troposphere in Northeast China but also the Meiyu rainfall in East Asia. On the basis of ERA-40 reanalysis data provided by ECMWF, the “climate effect” of NECV and its relationship with Meiyu in East Asia are studied. It is shown that there is significant correlation between NECV during the Meiyu period and rainfall amount: strong NECV corresponds to more Meiyu rainfall and weak NECV corresponds to less rainfall. In strong NECV years, the dry and cold air from the north is led to the south by NECV, converges with the lower-level warm and wet southwesterly on the north verge of Meiyu region, thus forms an unstable stratification of “upper dryness and lower wetness”. Triggered by ascending motion, the Meiyu rainfall amount is more than usual. It is on the contrary in weak NECV years. The anomalous SST in north Pacific in the previous year may be a factor that results in the anomalous NECV at Meiyu period. The land-sea thermal contrast in summer facilitates NECV, while that in winter inhibits NECV. All of the above provide a meaningful result for the short-term climate prediction of NECV and Meiyu.

Abstract: The survey conducted in Qingdao Agricultural University reveals the relationship between motivation and achievement as follows: instrumental motivation influences both high achievers and low achiever; while high achievers have greater integrative motivation than lower ones; Interest plays an extremely important role in study and high achievers have a strong sense of achievement.
PAEs in composts of municipal sludge and rice straw

Abstract: Four composting treatments for municipal sludge and rice straw were carried out, which included turning composting, inoculation-turning composting, continuous aeration, and intermittent aeration. GC/MS determination of phthalic acid esters (PAEs) in different composted composts showed that the total contents of six PAE compounds (sigma PAEs) ranged from 0.980-17.832 mg.kg-1, decreasing in the order of turning composting (17.832 mg.kg-1) > inoculation-turning composting (13.927 mg.kg-1) > intermittent aeration (10.765 mg.kg-1) > continuous aeration (9.815 mg.kg-1). DnOP was dominated in the composts. The degradation rate of sigma PAEs after composting was 1.37-45.71% (mean 27.63%), and that of diethyl phthalate (DEP), di-n-butyl phthalate (DnBP) and butylbenzyl phthalate (BBP) was 95.76-98.68%, 79.56-99.46% and 87.42-98.42%, respectively. The concentration of di-n-octyl phthalate (DnOP) was increased in the composts. Aeration-composting, and continuous aeration composting in particular, was more efficient than other treatments to degrade PAEs in municipal sludge.
