Finasteride Inhibits Human Prostate Cancer Cell Invasion through MMP2 and MMP9 Downregulation

Abstract: Introduction The use of the 5-alpha reductase inhibitors (5-ARIs) finasteride and dutasteride for prostate cancer prevention is still under debate. The FDA recently concluded that the increased prevalence of high-grade tumors among 5-ARI-treated patients must not be neglected, and they decided to disallow the use of 5-ARIs for prostate cancer prevention. This study was conducted to verify the effects of finasteride on prostate cell migration and invasion and the related enzymes/proteins in normal human and tumoral prostatic cell lines. Materials and Methods RWPE-1, LNCaP, PC3 and DU145 cells were cultivated to 60% confluence and exposed for different periods to either 10 μM or 50 μM finasteride that was diluted in culture medium. The conditioned media were collected and concentrated, and MMP2 and MMP9 activities and TIMP-1 and TIMP-2 protein expression were determined. Cell viability, migration and invasion were analyzed, and the remaining cell extracts were submitted to androgen receptor (AR) detection by western blotting techniques. Experiments were carried out in triplicate. Results Cell viability was not significantly affected by finasteride exposure. Finasteride significantly downregulated MMP2 and MMP9 activities in RWPE-1 and PC3 cells and MMP2 in DU145 cells. TIMP-2 expression in RWPE-1 cells was upregulated after exposure. The cell invasion of all four tested cell lines was inhibited by exposure to 50 μM of finasteride, and migration inhibition only occurred for RWPE-1 and LNCaP cells. AR was expressed by LNCaP, RWPE-1 and PC3 cells. Conclusions Although the debate on the higher incidence of high-grade prostate cancer among 5-ARI-treated patients remains, our findings indicate that finasteride may attenuate tumor aggressiveness and invasion, which could vary depending on the androgen responsiveness of a patient’s prostate cells.
Importancia do ligamento femoropatelar medial no deslocamento e na inclina??o lateral da patela: estudo radiográfico em cadáveres

Abstract: objective: acute patellar luxation is a complex disease that mainly affects young patients. its physiopathology is little known and the understanding of it and its therapeutic conduct are controversial. the medial femoropatellar ligament (mfpl) is the main static stabilizer for preventing lateral displacement of the patella. in order to assess the stability of the femoropatellar joint, the authors radiographically assessed the presence, or absence of lateral displacement and inclination of the patella before and after the mfpl section in the knees of cadavers. methods: thirty knees of cadavers were radiographed on the axial incidence of the patella by means of the technique described by merchant before and after mfpl section. the merchant angle of congruence and laurin lateral femoropatellar angle were measured. results: the medial femoropatellar ligament presented a mean length of 4.8cm, and width of 1.6 cm. in six anatomical pieces there was no change in the laurin lateral femoropatellar angle (20%), in three anatomical pieces the change was one degree (10%), in 20 (67%) two degrees and in one anatomical piece it was four degrees (3%). changes occurred between zero and two degrees in 97% of the knees of cadavers. in six anatomical pieces there was no change in the merchant angle of congruence (17%); in six anatomical pieces the change was one degree (20%), in 17 (57%) two degrees, in one anatomical piece it was three degrees (3%) and in one it was six degrees (3%). these changes occurred between zero and two degrees in 93% of the knees of cadavers. conclusion: analysis of the results obtained in this study allowed us to conclude that the medial femoropatellar ligament is important in the lateral inclination and displacement of the patella with the knee flexed at 45o.
Molecular Assessment of Non-Muscle Invasive and Muscle Invasive Bladder Tumors: Mapping of Putative Urothelial Stem Cells and Toll-Like Receptors (TLR) Signaling  [PDF]

Abstract:

Purpose: The main objectives of this study were to characterize and compare the urothelial stem cells (healthy and cancer cells) and TLRs features in the urinary bladder of men without lesionsand with non-muscle-invasive and muscle invasive urothelial tumors. Materials and Methods: Thirty samples of the urinary bladder of 50 to 80-year-old men, with and without diagnosis of malignant urothelial lesions were used. The 30 samples were divided into 3 groups (n = 10 per group): Normal Group; Non-Muscle Invasive Bladder Cancer Group; Muscle Invasive Bladder Cancer Group. The samples were histopathologically and immunohistochemically analyzed. The study was conducted at teaching Hospital of the University of Campinas (UNICAMP). Results: The CD44 and CD133 immunoreactivities were significantly intense in the muscle-invasive cancer group when compared to the other groups. The ABCG2 biomarker demonstrated intense immunoreactivities in both non-muscle and muscle invasive groups, and absent immunoreactivity in the normal group. All groups showed weak CD117 immunoreactivity. Putative Healthy Stem Cells (CD44/CD133/ CD117+) occurred in all groups. Putative Cancer Stem Cells (CD44/CD133/ABCG2+) only occurred in the non-muscle and muscle invasive cancer groups. TLR2 immunoreactivity was significantly lower in the non-muscle invasive cancer group and absent in the muscle invasive cancer group. TLR4 immunoreactivity was significantly lower in both cancer groups. Conclusions: This study leads us to the conclusion that putative cancer stem cell occurrence was sensitive to the decreased in TLR2 and TLR4 immunoreactivities. Also, TLR2 and TLR4 demonstrated their involvement in the regulation of the different biomarkers for putative healthy and cancer urothelial stem cells, probably acting as negative regulators of urothelial carcinogenesis. Taken together data obtained suggest that use of TLRs agonists could be a promising alternative for the treatment of non-muscle and muscle invasive bladder tumors.

Avalia o dos pacientes submetidos à fixa o de fraturas diafisárias do úmero pela técnica minimamente invasiva - Placa-ponte Evaluation of patients undergoing fixation of diaphyseal humeral fractures using the minimally invasive bridge - Plate technique

Abstract: OBJETIVO: O objetivo deste trabalho é relatar a experiência de nosso grupo no tratamento da fratura diafisária de úmero, através da técnica de placa-ponte por via anterior. MéTODOS: Foram avaliados 17 pacientes com fratura diafisária do úmero aguda com indica o de tratamento cirúrgico operados no período 2006-2010. Foram utilizadas as classifica es AO e Gustilo e Anderson. Todos os pacientes foram operados pela técnica de placa-ponte anterior e cumpriram seguimento pós-operatório mínimo de 12 meses. RESULTADOS: Foram tratados 16 homens e uma mulher. A idade média foi de 31,8 anos (18-52). Dentre os mecanismos de les o, encontramos: cinco acidentes motociclísticos, quatro acidentes automobilísticos, três fraturas por projétil de arma de fogo, duas quedas ao solo e, por último, agress o, esmagamento e atropelamento, com um paciente cada. Oito pacientes apresentavam fratura exposta, sendo duas grau I, uma grau II, quatro grau IIIa e uma grau IIIb, de acordo com a classifica o de Gustilo e Anderson. Com rela o à classifica o AO, encontramos: uma 12A1, três 12A2, quatro 12A3, uma 12B1, quatro 12B2, três 12B3 e uma 12C2. A média de seguimento pós-operatório foi de 25 meses (12-48). Como complica es, dois pacientes apresentaram dor no cotovelo e déficit de ADM e um teve infec o. O tempo médio de consolida o foi de 17,5 semanas. N o houve perda de redu o, pseudartrose ou consolida o viciosa nesta série de pacientes. CONCLUS O: Os autores acreditam que a técnica descrita apresenta baixos índices de complica es e morbidade, demonstrando bons resultados iniciais, embora a série seja limitada pela pequena amostra. OBJECTIVE: The aim was to describe the experience of our group in treating humeral shaft fractures using the bridge-plate technique via an anterior approach. METHODS: Seventeen patients with acute diaphyseal humeral fractures with an indication for surgical treatment who were operated in 2006-2010 were evaluated. The AO and Gustilo & Anderson classifications were used. All the patients were operated using the anterior bridge-plate technique and completed a follow-up period of at least twelve months. RESULTS: Sixteen men and one woman were treated. Their mean age was 31.8 years (18-52). Among the injury mechanisms found were: five motorcycle accidents, four car accidents, three fractures due to firearm projectiles, two falls to the ground and finally, with one case each, assault, crushing and being run over. Eight patients had open fractures: two grade I, one grade II, four grade Ilia and one grade Illb, according to the Gustilo-Anderson classif

Abstract: the authors clinically assessed 19 patients (19 knees - 15 medial meniscus and 5 lateral meniscus) submitted to meniscus suture using 2 kinds of absorbable implants (polylactic acid) arrow? and clear fix?. the study is composed of 15 males and 4 females, ages ranging 16 - 44 years old (average = 26.8 years old). the mean follow-up time was 24 months, ranging from 18 to 32 months, at most. the surgical technique was constituted of a suture in one or both meniscus (1 case), through arthroscopy with arrow? in 15 patients and clear fix? in 4. from the 19 individuals, 6 were submitted to isolated meniscal suture, 11 combined to acl reconstruction and 2 to acl reconstruction with tibial valgusing osteotomy. the results were assessed according to physical examinations including the maneuvers of appley and mc murray. pre- and postoperative functional evaluation of the knee was performed using a modified lysholm scale. all patients had negative meniscal maneuvers postoperatively. the average preoperative score according to lysholm scale was 39.8 reaching 91.5 postoperatively. the authors conclude that the meniscus suture using absorbable implants has shown to be efficient so far, and that, technically, it is simpler than conventional suture.

Abstract: the authors present a series of seven case reports of localized pigmented villonodular synovitis (lpvns) treated by arthroscopy resection between june of 1994 and october of 2001. at the baseline evaluation, symptoms ranged from diffuse pain to localized swelling of the knee, with or without mechanical blockage of the joint. magnetic resonance helped in localizing the lesion and in the follow up evaluation. diagnosis was confirmed through anatomicopathological examination. by the end point evaluation, none of the patients presented recurrence of the pre-operative symptoms referred before. magnetic resonance imaging also showed absence of symptoms. despite of the small number of patients, typical of this affection, the authors believe that arthroscopic excision of the lpvns is an effective method, with low morbidity, providing remission of the symptoms and a low recurrence potential.

Abstract: the authors studied 100 normal children from the general population of both genders with ages ranging from 5 to 9 years old in order to evaluate the plantar arch index and the flat-feet prevalence. the flat-feet evaluation was obtained by means of the footprint and the plantar arch index (ip), which establishes the ratio between central and posterior regions of this footprint, determining a mean ip and a limit to the flat-foot. they conclude that the plantar arch index is easy to obtain from footprints and that there are no differences in terms of gender or age. the mean values of the plantar arch index within this age group are stable and range from 0.61 to 0.67, with plantar arch indexes greater than 1.15 being regarded as flat feet.

Abstract: the color in vegetables is one of the main quality attributes observed by the consumer and it is influenced by intrinsic characteristics and canning process. this research meant to investigate the effect of increasing calcium concentrations applied to a nutrient solution on physical measurements of the color of snap bean pods.this experiment was carried out wtih the cultivar uel 1 grown in coarse sand with nutrient solution supplied at the following levels of calcium: 0, 75, 150 and 300 ml l-1. fresh pods were analysed for calcium content. the fresh and canned pods were evaluated for color with tristimulus colorimetry and data were expressed according to the hunter system. the fresh pods were lighter and yellowisher when calcium was added to the nutrient solution. the heat used for the canning process had a significant effect on reducing lightness and greeness, even with the greater amounts of calcium in the pods.

Abstract: A cor em vegetais é um dos principais atributos de qualidade observado pelo consumidor e é influenciada por características intrínsecas e de processamento. O objetivo desta investiga o foi avaliar as altera es de cor de feij o vagem fresco e processado após aplica o de concentra es crescentes de cálcio via absor o radicular. O feij o vagem cultivar UEL 1 foi cultivado com aplica o de concentra es crescentes de cálcio via solu o nutritiva contendo 0, 75 , 150 e 300 mL L-1 de cálcio em substrato de areia grossa. A cor das vagens ao natural e enlatadas foram avaliadas por colorimetria de três estímulos, e os resultados foram expressos no sistema Hunter. As vagens ao natural apresentaram-se mais claras e amarelas quando foi adicionado cálcio à solu o nutritiva. O calor empregado no processo de enlatamento teve efeito pronunciado na redu o da luminosidade e na cor verde, mesmo com o aumento na quantidade de cálcio nas vagens.
