Effect of policosanol on cerebral ischemia in Mongolian gerbils

Abstract: Policosanol is a mixture of higher aliphatic primary alcohols isolated from sugar cane wax, whose main component is octacosanol. An inhibitory effect of policosanol on platelet aggregation and cerebral ischemia in animal models has been reported. Thus, the objective of the present study was to evaluate the effect of policosanol on cerebral ischemia induced by unilateral carotid ligation and bilateral clamping and recirculation in Mongolian gerbils. Policosanol (200 mg/kg) administered immediately after unilateral carotid ligation and at 12- or 24-h intervals for 48 h significantly inhibited mortality and clinical symptoms when compared with controls, whereas lower doses (100 mg/kg) were not effective. Control animals showed swelling (tissue vacuolization) and necrosis of neurons in all areas of the brain studied (frontal cortex, hippocampus, striatum and olfactory tubercle), showing a similar injury profile. In the group treated with 200 mg/kg policosanol swelling and necrosis were significantly reduced when compared with the control group. In another experimental model, comparison between groups showed that the brain water content of control gerbils (N = 15) was significantly higher after 15 min of clamping and 4 h of recirculation than in sham-operated animals (N = 13), whereas policosanol (200 mg/kg) (N = 19) significantly reduced the edema compared with the control group, with a cerebral water content identical to that of the sham-operated animals. cAMP levels in the brain of control-ligated Mongolian gerbils (N = 8) were significantly lower than those of sham-operated animals (N = 10). The policosanol-treated group (N = 10) showed significantly higher cAMP levels (2.68 pmol/g of tissue) than the positive control (1.91 pmol/g of tissue) and similar to those of non-ligated gerbils (2.97 pmol/g of tissue). In conclusion, our results show an anti-ischemic effect of policosanol administered after induction of cerebral ischemia, in two different experimental models in Mongolian gerbils, suggesting a possible therapeutic effect in cerebral vascular disorders.

Abstract: policosanol is a mixture of higher aliphatic alcohols purified from sugar cane wax, with cholesterol-lowering effects demonstrable in experimental models and in patients with type ii hypercholesterolemia. the protective effects of policosanol on atherosclerotic lesions experimentally induced by lipofundin in rabbits and rats and spontaneously developed in stumptail monkeys have been described. the present study was conducted to determine whether policosanol administered orally to rabbits with exogenous hypercholesterolemia also protects against the development of atherosclerotic lesions. male new zealand rabbits weighing 1.5 to 2 kg were randomly divided into three experimental groups which received 25 or 200 mg/kg policosanol (n = 7) orally for 60 days with acacia gum as vehicle or acacia gum alone (control group, n = 9). all animals received a cholesterol-rich diet (0.5%) during the entire period. control animals developed marked hypercholesterolemia, macroscopic lesions and arterial intimal thickening. intima thickness was significantly less (32.5 ± 7 and 25.4 ± 4 μm) in hypercholesterolemic rabbits treated with policosanol than in controls (57.6 ± 9 μm). in most policosanol-treated animals, atherosclerotic lesions were not present, and in others, thickness of fatty streaks had less foam cell layers than in controls. we conclude that policosanol has a protective effect on the atherosclerotic lesions occurring in this experimental model.

Abstract: Los eventos coronarios constituyen la primera causa de muerte en sujetos con diabetes mellitus tipo 2. Un incremento en la agregación plaquetaria y altos niveles de colesterol asociado a la lipoproteína de baja densidad (C-LDL) contribuyen al riesgo coronario en diabéticos. El D-003, una mezcla de ácidos grasos obtenida de la ca a de azúcar, ha mostrado reducir la agregación plaquetaria y los niveles séricos de C-LDL en sujetos normo e hipercolesterolémicos. Este estudio a doble ciego, controlado con placebo, investigó los efectos del D-003 sobre la agregación plaquetaria y el perfil lipídico en 50 diabéticos tipo 2, los que fueron aleatorizados para recibir después de un periodo inicial, D-003 (10 mg/día) o placebo por 10 semanas. Todos los sujetos completaron el estudio. El D-003 redujo significativamente la agregación plaquetaria inducida por ácido araquidónico (52,9%) y por colágeno (54,4%) y los niveles séricos de C-LDL (26,7%), colesterol total (CT) (19,6%) y triglicéridos (23,9%), mientras que incrementó el C-HDL (12,4%) en relación a los niveles básales y al grupo placebo. El D-003 fue seguro y bien tolerado. Se concluye que el D-003 redujo significativamente la agregación plaquetaria y los niveles séricos de C-LDL en pacientes con diabetes tipo 2, pero otros estudios deben confirmar estos resultados. D-003, a mixture of sugarcane wax acids, has shown to reduce platelet aggregation and serum C-LDL in normocholesterolemic and hypercholesterolemic subjects. This doubleblinded, placebo-controlled study investigated the effects of D-003 on platelet aggregation and lipid profile in 50 type 2 diabetes patients who were randomized, after a baseline phase, to D-003 (10 mg/d) or placebo for 10 weeks. Al I the subjects completed the study. D-003 significantly lowered arachidonic acid- (52.9%) and collagen-induced (54.4%) platelet aggregation, C-LDL (26-7%), total cholesterol (TC) (19.6%) and triglycerides (23.9%), while increased high-density lipoprotein-cholesterol (C-HDL) (12.4%) vs baseline and placebo. D-003 was safe and well tolerated. To conclude with, D-003 significantly reduced platelet aggregation and serum C-LDL in type 2 diabetes, but further studies should confirm these results.

Abstract: D-004, un extracto lipídico de las frutas de Roystonea regia, inhibe la contracción provocada por la norepinefrina en el vas deferens de las ratas. No obstante, el efecto de D-004 sobre las respuestas contráctiles adrenérgicas a1 en las preparaciones aisladas de la próstata no ha sido estudiado. Por lo tanto, este estudio investigó los efectos de D-004 sobre las contracciones provocadas por la fenilefrina en las extracciones aisladas de la próstata de la rata. D-004 (125-500 mg/mL) fue adicionado a las preparaciones a partir de la próstata en la que las contracciones fueron provocadas por la fenilefrina (10-6-10-4 M) o por CIK (50 mM). D-004 (250-500 mg/mL) en forma significativa y la dosis en forma dependiente inhibieron las contracciones provocadas por la fenilefrina mediante un mecanismo no competetitivo, reduciendo así (12-60 %) la contracción máxima por la fenilefrina con respecto a las preparaciones controles. D-004 a 500 μg/mL inhibieron de la misma forma tanto CIK como las respuestas contráctiles-fenilefrina. Tamsulosin (0,01 mg/mL) eliminó la acción de la fenilefrina (100 % de inhibición), pero no afectó las contracciones provocadas por CIK. D-004 (250-500 mg/mL) significantly (p < 0,01).and dose-dependently inhibited the phenylephrine-induced contractions through a non-competitive mechanism, since it reduced (12-60 %) the maximal contraction to phenyle phrine with respect to control preparations. D-004 at 500 mg/mL inhibited similarly both ClK and phenylephrine-contractile responses. Tamsulosin (0.01 mg/mL) abolished (100 % inhibition) the phenylephrine, but unaffected the ClK-induced contractions.

Abstract: depression is among the major causes of morbidity and mortality of adult population and its pharmacologic management includes among other alternative options, the use of the complementary medicine, e.g. the fish oil rich in omega-3 fatty acids (agw3). the d-004, a lipid extract of real palm fruits (roystonea regia) consisting of a fatty acid mixture, has a moderate antidepressant action. the aim of present paper was to compare the effects of oral treatment using d-004 with the agw3 and with its combined therapy in forces swimming in mice. thus, the male mice were distributed into 8 groups: one as control treated with vehicle, two with d-004 (250 and 500 mg.kg), two with agw3 (250 and 500 mg/kg), two with the combination, and another with imipramine (10 mg/kg i.p.). all treatments decreased significantly the immobility time regarding the control, without differences among similar dose of respective treatments. we conclude that d-004, the agw3 and its combined therapy were similary effective to reduce the immobility time in mices without additive or synergic effects with the combined administration of both substances.

Abstract: Mathematical statement of elastodynamic contact problem for cracked body with considering unilateral restrictions and friction of the crack faces is done in classical and weak forms. Different variational formulations of unilateral contact problems with friction based on boundary variational principle are considered. Nonsmooth optimization algorithms of Udzawa’s type for solution of unilateral contact problem with friction have been developed. Convergence of the proposed algorithms has been studied numerically.
