Transcriptional activation of the Axl and PDGFR-α by c-Met through a ras- and Src-independent mechanism in human bladder cancer

Abstract: Both NIH-Met5 and T24-Met3 cell lines harboring an inducible human c-Met gene were established. C-Met-related RTKs were screened by RTK microarray analysis. The cross-talk of RTKs was demonstrated by Western blotting and confirmed by small interfering RNA (siRNA) silencing, followed by elucidation of the underlying mechanism. The impact of this cross-talk on biological function was demonstrated by Trans-well migration assay. Finally, the potential clinical importance was examined in a cohort of 65 cases of locally advanced and metastatic bladder cancer patients.A positive association of Axl or platelet-derived growth factor receptor-alpha (PDGFR-α) with c-Met expression was demonstrated at translational level, and confirmed by specific siRNA knock-down. The transactivation of c-Met on Axl or PDGFR-α in vitro was through a ras- and Src-independent activation of mitogen-activated protein kinase/extracellular signal-regulated kinase (MEK/ERK) pathway. In human bladder cancer, co-expression of these RTKs was associated with poor patient survival (p < 0.05), and overexpression of c-Met/Axl/PDGFR-α or c-Met alone showed the most significant correlation with poor survival (p < 0.01).In addition to c-Met, the cross-talk with Axl and/or PDGFR-α also contributes to the progression of human bladder cancer. Evaluation of Axl and PDGFR-α expression status may identify a subset of c-Met-positive bladder cancer patients who may require co-targeting therapy.The RTK c-Met is expressed during normal development and plays a crucial role in many cell regulatory processes [1]. Abstract: In this study, the genetic and antigenic properties of these strains were analyzed and the genetic diversity of EV-71 subgenogroups surfacing in Taiwan was depicted, which includes 3 previously reported subgenogroups of C5, B5, and C4, and one C2-like subgenogroup. Based on the phylogenetic analyses using their complete genome nucleotide sequences and neutralization tests, the C2-like subgenogroup forms a genetically distinct cluster from other subgenogroups, and the antisera show a maximum of 128-fold decrease of neutralization titer against this subgenogroup. In addition, the subgenogroup C4 isolates of 2008 were found quite similar genetically to the Chinese strains that caused outbreaks in recent years and thus they should be carefully watched.Other than to be the first report describing the existence of C2-like subgenogroup of EV-71 in Taiwan, this article also foresees a potential of subgenogroup C4 outbreaks in Taiwan in the near future.Belonging to the genus Enterovirus of the family Picornaviridae, human enterovirus 71 (EV-71) is one of the most causative pathogens infecting humans and may cause outbreaks of hand-foot-mouth disease (HFMD), herpangina, and severe neurological symptoms, especially in young children [1]. GSK3beta-Mediated Drp1 Phosphorylation Induced Elongated Mitochondrial Morphology against Oxidative Stress

Abstract: Multiple phosphorylation sites of Drp1 have been characterized for their functional importance. However, the functional consequence of GSK3beta-mediated phosphorylation of Drp1 remains unclear. In this report, we pinpointed 11 Serine/Threonine sites spanning from residue 634~736 of the GED domain and robustly confirmed Drp1 Ser693 as a novel GSK3beta phosphorylation site. Our results suggest that GSK3beta-mediated phosphorylation at Ser693 does cause a dramatic decrease of GTPase activity; in contrast, GSK3beta-mediated phosphorylation at Ser693 appears not to affect Drp1 inter-/intra-molecular interactions. After identifying Ser693 as a GSK3beta phosphorylation site, we also determined that K679 is crucial for GSK3beta-binding, which strongly suggests that Drp1 is a novel substrate for GSK3beta. Thereafter, we found that overexpressed S693D, but not S693A mutant, caused an elongated mitochondrial morphology which is similar to that of K38A, S637D and K679A mutants. Interestedly, using H89 and LiCl to inhibit PKA and GSK3beta signaling, respectively, it appears that a portion of the elongated mitochondria switched to a fragmented phenotype. In investigating the biofunctionality of phosphorylation sites within the GED domain, cells overexpressing Drp1 S693D and S637D, but not S693A, showed an acquired resistance to H2O2-induced mitochondrial fragmentation and ensuing apoptosis, which affected cytochrome c, capase-3, -7, and PARP, but not LC3B, Atg-5, Beclin-1 and Bcl2 expressions. These results also showed that the S693D group is more effective in protecting both non-neuronal and neuronal cells from apoptotic death than the S637D group. Altogether, our data suggest that GSK3beta-mediated phosphorylation at Ser693 of Drp1 may be associated with mitochondrial elongation via down-regulating apoptosis, but not autophagy upon H2O2 insult.

Abstract: This paper presents a multicriterica decision making method (MCDM) to evaluate the performance of the fourth-party logistics providers. The four indexes of balanced scorecard (BSC) are used as the evaluation indexes. AHP (analytical hierarchy process) is used for rating the weights of criteria and alternatives. MCDM method of SAW (Simple Additive Weighting) is used for ranking the companies. Results show that the approach is applicable for the performance measurement problem.

This paper develops a multi-criteria decision making method (MCDM) method to evaluate the employee performance in a logistics company. Analytic Hierarchy Process (AHP) is used for the weights of criteria and employees. Technique for order preference by similarity to an ideal solution (TOPSIS) is used for ranking the overall performance of the employees. Result shows that the proposed method is a generalized method and is applicable for the performance appraisal problem.

Inhibitory Effects of Resveratrol on the Epstein-Barr Virus Lytic Cycle

Abstract: Reactivation of Epstein-Barr virus (EBV) from latency to the lytic cycle is required for the production of viral particles. Here, we examine the capacity of resveratrol to inhibit the EBV lytic cycle. Our results show that resveratrol inhibits the transcription of EBV immediate early genes, the expression of EBV lytic proteins, including Rta, Zta, and EA-D and reduces viron production, suggesting that this compound may be useful for preventing the proliferation of the virus.
It Takes a Village to Raise a Child: Building Social Capital in Safe And Cohesive Neighbourhood

Abstract: A vibrant and close-knit community is always associated with norms of trust and reciprocity with friendly neighbours in order to produce generalised social capital for the wider society. The researcher argues that the experience of long term interaction among neighbours, neighbourhood safety and cohesion and an active neighbourhood environment all contribute to social capital among neighbours. As a result, young people become more trusting in and reciprocative with people in general. Young Australians and permanent residents aged 16 to 25 (n=283) participated in this study through an online survey. The findings support previous literature, which found cohesive neighbours are more trusting than less cohesive neighbours. The results also indicate that the young people who perceived their neighbourhood as safe were relatively more trusting than those who perceived their neighbourhood as less safe. These suggest that creating neighbourhood ‘togetherness’ and preventions of crime promotes could promote more neighbourhood social capital. In turn, young people enjoy more generalised social capital in the wider society.
Content Analysis of People with Disabilities in Chinese-Language Elementary School Textbooks

Abstract: Textbooks are able to shape the attitude of students. The descriptions of people with disabilities in textbooks would influence students’ attitude to people with disabilities. The purpose of this study was to analyze people with disabilities described in Chinese-language elementary school textbooks, including the prevalence of categories of disabilities, the development of their character, significant others and inclusive education. The results of study include: (1) People with disabilities are significantly underrepresented in Chinese-language elementary school textbooks. The most prevalent disabilities found in textbooks focused on physical disabilities in nature. Four categories of disabilities neglected in textbooks including hearing impairment, autism, emotional disabilities, and developmental retardation. (2) Textbooks tended to emphasize the character development of well-known people with disabilities. (3) Significant others for people with disabilities including parents, teachers, and their peers. (4) Only two compositions regarding to inclusive education in textbooks. The themes focused on the participation of physically and mentally disabled students in class activities and experiencing disabilities.

Abstract: Polygonum cuspidatum is widely used as a medicinal herb in Asia. In this study, we examined the ethyl acetate subfraction F3 obtained from P. cuspidatum root and its major component, emodin, for their capacity to inhibit the Epstein-Barr virus (EBV) lytic cycle. The cell viability was determined by the MTT [3-(4,5-dimethyldiazol-2-yl)-2,5-diphenyltetrazolium bromide] method. The expression of EBV lytic proteins was analyzed by immunoblot, indirect immunofluorescence and flow cytometric assays. Real-time quantitative PCR was used to assess the EBV DNA replication and the transcription of lytic genes, including BRLF1 and BZLF1. Results showed that the F3 and its major component emodin inhibit the transcription of EBV immediate early genes, the expression of EBV lytic proteins, including Rta, Zta, and EA-D and reduces EBV DNA replication, showing that F3 and emodin are potentially useful as an anti-EBV drug.
