MRN1 Implicates Chromatin Remodeling Complexes and Architectural Factors in mRNA Maturation

Abstract: A functional relationship between chromatin structure and mRNA processing events has been suggested, however, so far only a few involved factors have been characterized. Here we show that rsc nhp6ΔΔ mutants, deficient for the function of the chromatin remodeling factor RSC and the chromatin architectural proteins Nhp6A/Nhp6B, accumulate intron-containing pre-mRNA at the restrictive temperature. In addition, we demonstrate that rsc8-ts16 nhp6ΔΔ cells contain low levels of U6 snRNA and U4/U6 di-snRNA that is further exacerbated after two hours growth at the restrictive temperature. This change in U6 snRNA and U4/U6 di-snRNA levels in rsc8-ts16 nhp6ΔΔ cells is indicative of splicing deficient conditions. We identify MRN1 (multi-copy suppressor of rsc nhp6ΔΔ) as a growth suppressor of rsc nhp6ΔΔ synthetic sickness. Mrn1 is an RNA binding protein that localizes both to the nucleus and cytoplasm. Genetic interactions are observed between 2 μm-MRN1 and the splicing deficient mutants snt309Δ, prp3, prp4, and prp22, and additional genetic analyses link MRN1, SNT309, NHP6A/B, SWI/SNF, and RSC supporting the notion of a role of chromatin structure in mRNA processing.

Abstract: Dieser Aufsatz geht dem Ph nomen des Erlebens der Zukunft anhand einer Fallstudie nach, in der ein skandinavisches New-Economy-Unternehmen eine Reihe von Wirtschaftsjournalisten davon überzeugte, es verk rpere "die Zukunft". Hierzu werden die Verfahrensweisen und rhetorischen Strategien beschrieben, die die Manager anwendeten, um die Journalisten in "Zeugen" zu verwandeln. Die Strategien des Managements werden mit anderen F llen von Zeugenschaft in der Wissenschaft und im Gerichtssaal in Beziehung gesetzt. Der Aufsatz argumentiert, dass die überzeugungskraft des Managements sich aus seiner F higkeit herleitet, eine Reihe von scharfen Kontrasten zwischen dem attraktiven Arbeitsleben innerhalb und dem problematischen Arbeitsleben au erhalb des Unternehmens herauszustellen. Schlie lich wird herausgearbeitet, dass die Strategie des Managements eine Zeit-Welt aufruft, die von dramatischen epochalen Brüchen gepr gt ist und sich radikal von der stabileren und erfassbareren Zeit-Welt unterscheidet, wie sie in normalen wissenschaftlichen Diskursen inszeniert wird. URN: urn:nbn:de:0114-fqs070119 The paper explores the phenomenon of witnessing the future through a case study of how a Scandinavian new economy firm managed to persuade a number of business journalists that it was "the future". It describes the procedures and rhetorical strategies that the manager deployed to turn the journalists into witnesses. It compares the manager's strategy to other cases of effective witnessing in courtrooms and in science. It concludes that the manager's persuasiveness is derived from his ability to articulate a series of pointed contrasts between the attractive working life within the firm and the problematic work life elsewhere. Finally, it notes that the manager's strategy enacts a time-world characterised by dramatic epochal changes, which is radically different from the more stable and knowable time-world that is enacted in ordinary scientific discourses. Abstract: Drawing on actor-network theory, this paper challenges the traditional analytical separation of the socalled social and the so-called technical. First, observational data of an interactional event between a social worker and a client is introduced. Second, the techno-social heterogeneity of the event is elucidated through an analysis based on the concept of translation. Third, the precarious and temporary natures of the techno-social hybrids are discussed through the concept of performance. Finally, the techno-social is proposed as a new object for social science.

Abstract:

In everyday life young people with ethnic minority background, or “the new youth” in Europe, face both a social and cultural integration challenge. On the one hand, they are placed as other young people in an individualisation and identity process of late modern youth life which is demanding and important for future social success. On the other hand, they are challenged by a new cultural lifestyle which may be different from traditions and values of parents. This cultural integration process therefore may make it difficult to engage and become successful in late modern life. So, their cultural identity is challenged and in a change. This way of contrasting two different perspectives in young people’s lives seems difficult because of the different theoretical views embedded in these perspectives. What however both approaches seem to overlook is, that ethnic minority social integration is a process which has in its centre the individual and individualised agent. This focus combines the two perspectives. The integration process is mostly about the agent as an engaged player in his or her life. An agent is one who finds and uses energy in a process of becoming integrated in some kind of social and cultural life. In our paper, we use experiences from

Abstract: We have studied possible effects of the PhiC31 integrase expression in human adult fibroblasts by karyotyping. All control cells were cytogenetically normal, whereas cells expressing PhiC31 integrase show chromosomal abnormalities confirming our previous results using primary embryonic fibroblasts. In order to study the early mechanisms involved we measured H2AX phosphorylation – a primary event in the response to DNA double-strand-breaks. Transient transfection with PhiC31 integrase encoding plasmids lead to an elevated number of cells positive for H2AX phosphorylation detected by immunofluorescence. Western blot analysis confirmed the upregulated H2AX phosphorylation, whereas markers for apoptosis as well as p53 and p21 were not induced. Cells transfected with plasmids encoding the Sleeping Beauty transposase remained cytogenetically normal, and in these cells less upregulation of H2AX phosphorylation could be detected.In primary human fibroblasts expression of PhiC31 integrase leads to a DNA damage response and chromosomal aberrations.PhiC31 integrase, originally isolated from Streptomyces lividans, is widely used for non-viral gene delivery and vector integration [1]. PhiC31 integrase mediated integration leads to prolonged gene expression and has been used for correction of disease models [2,3]. However, it has been reported that PhiC31 integrase can lead to genomic deletions, chromosomal rearrangements and chromosomal instability [4-8]. In mouse cells it was recently shown that the integrase does lead to imprecise deletion of self excision cassettes [9]. In cell lines, PhiC31-mediated integration of plasmid DNA may be accompanied by chromosomal rearrangements in the mammalian host genome with a frequency up to 15% [10]. The mechanisms involved are unknown, but it has been speculated that cryptic PhiC31 attachment sites recombine leading to chromosomal translocations [10]. Danish universities mandatorily obtain student feedback through survey instruments. To understand the construct of study environment a theoretical model is proposed. Based on survey data from students at the University of Aarhus the most important factors predicting student well-being are identified through statistical analysis.
