Fatty acids, cholesterol, oxidative rancidity, and color of irradiated shrimp

Abstract: the effect of gamma irradiation (0, 2, 4, and 6 kgy doses) applied to frozen and packed headed shrimp on the fatty acid profile, cholesterol content, and lipid and color stability was evaluated. myristic acid was higher in shrimp irradiated with 4 and 6 kgy and palmitic acid was higher in samples irradiated with 2 and 6 kgy compared to non-irradiated samples. stearic and behenic acids were lower in shrimp irradiated with 6 kgy compared to non-irradiated shrimp. with regard to non-irradiated shrimp, palmitoleic, oleic, and linoleic acids and total monounsaturated fatty acids were higher in shrimp irradiated with 6 kgy. saturated fatty acid and cholesterol contents in irradiated samples were not different from those in non-irradiated shrimp. lipid oxidation was higher in samples irradiated with 2, 4, and 6 kgy. redness and yellowness of cooked shrimp were higher in samples irradiated with 6 kgy than in those in non-irradiated samples. the application of irradiation in doses up to 6 kgy on frozen and packed headed shrimp does not affect negatively the fatty acid profile, cholesterol content, and lipid and color stability.
Type of dietary lipids and storing time on egg stability

Abstract: the objective of this work was to evaluate the influence of diets containing different lipid sources on eggs quality during refrigerated storage, on yolk fatty acid composition, and on cholesterol in the yolk. four diets were used containing soy oil (so), sunflower seed (ss), and meat and bone meal + soy oil (mbm + so) or meat and bone meal + tallow (mbm + ta). the experiment followed a factorial design 4 × 3 with four dietary treatments and three storage times. the eggs were stored at 4 °c for 0, 30, and 60 days. the collected eggs were analyzed for egg weight loss, haugh units, yolk moisture, yolk lipid oxidation, and cooked yolk firmness. refrigerated storage reduced haugh units, and increased yolk moisture. sixty days of storage time reduced the firmness of hard-cooked yolk. there was an interaction between dietary treatment and storage time for egg weight loss and lipid oxidation. with regard to yolk fatty acid profile, mbm + ta diet increased the contents of palmitic and palmitoleic acids. the levels of oleic and arachidonic acids were higher in yolks from birds fed with ss diet. linoleic acid level was higher in the yolk from treatment with so diet. diets containing mbm + so induced higher levels of docosahexaenoic acid. yolk cholesterol content was reduced with the inclusion of ss in the diet. therefore, the type of lipid present in the diet and refrigerated storage for 60 days at 4 °c can affect the egg quality.

Abstract: the objective of this work was to assess the effect of feeding rabbits diets containing coconut meal (cm) on meat proximal composition, the fatty acid profile, and the polyunsaturated to saturated (p/s) fatty acids ratio in the meat. the experiment utilized 60 rabbits fed diets containing five levels of cm (0.00, 6.25, 12.50, 18.75, and 25.00%) and 12 animals per treatment. increasing levels of cm in the diet did not affect (p > 0.05) meat centesimal composition. the levels of palmitoleic, stearic, and linolenic acids in the meat were linearly affected by dietary cm levels. when compared to the levels of fatty acids in the meat from the 0.00% cm diet, the palmitic acid was lower (p < 0.05) in the meat from all diets containing cm, the myristic and stearic acids were higher (p < 0.05), and the palmitoleic acid was lower (p < 0.05) in the meat from the diet containing 25.00% cm. the linolenic acid level was lower (p < 0.05) in meats from diets containing either 18.75 or 25.00% cm. the p/s relation in the meat was not affected (p > 0.05) by the levels of cm in the diets. this suggested that the inclusion of this by-product in the diets is feasible even at the 25.00% level.
Antioxidant effect of mango seed extract and butylated hydroxytoluene in bologna-type mortadella during storage

Abstract: the effects of mango seed extract (mse) and butylated hydroxytoluene (bht) on ph, lipid oxidation, and color of bologna-type mortadella during refrigerated storage for 21 days were studied. bologna-type mortadella samples were formulated to contain 0.1% mse, 0.2% mse, or 0.01% bht. after 14 days of storage, the products containing mse 0.1 or 0.2% had higher ph values than those containing bht 0.01%. lipid oxidation values increased with storage time but were not affected by the type of antioxidant. the highest values for color parameter l* were observed for mortadella containing bht 0.01% after 7, 14, and 21 days of storage. values for the color parameters a* and b* tended to decrease during mortadella storage. products containing 0.1 or 0.2% mse showed higher values for color parameter a* and lower values for color parameter b* compared to those containing 0.01% bht. it can be concluded that mse can be used in 0.1 or 0.2% levels in bologna-type mortadella with similar or better antioxidant effects than those of bht 0.01%.

Abstract: abstract introduction: various methodologies have been reported for long term specimen preservation, which protect the dna from degradation allowing its posterior analysis in molecular studies. however, the effectiveness of the preservation methods may vary among insect groups; therefore, it is convenient to determine the preservation method to be used with a particular group before performing molecular studies with a large number of specimens. objetives: an experimental comparative study was conducted to evaluate different preservation treatments for anopheles albimanus, to know their utility in conserving the quantity and quality of dna to be used in subsequent molecular studies. methods: dna was extracted from each specimen, it was then quantified by spectrophotometer and the internal transcribed spacer 2 (its2) was amplified by pcr. results: its2 amplification was obtained in all specimens without previous preservation, in 60% of those preserved in sílica gel and in 20% of those preserved at -80°c and in 70% ethanol. a significant difference (p<0.05) was found between amplification proportions. amplification was not obtained in specimens preserved frozen at -20°c and not correlation by logistic regression was observed between the do260/do280 index and dna concentration of specimens presenting its2 amplification. conclusions: the data suggest that keeping specimens in silica gel or frozen at -80°c may be the best conditions to preserve anopheles specimens for subsequent molecular studies.

Comparación de métodos de preservación de especímenes Anopheles (Diptera: Culicidae) para la extracción de ADN

Abstract: Objetivos: Evaluar diferentes tratamientos de preservación de especímenes Anopheles albimanus para conocer su utilidad para conservar la cantidad y calidad del ADN que permita su análisis en estudios moleculares posteriores. Materiales y métodos: Se realizó un estudio comparativo experimental con tres bloques de tratamientos de preservación: sílica gel, etanol y congelación; los dos últimos bloques se dividieron en dos niveles cada uno: etanol absoluto, etanol al 70% y congelación a -20°C, congelación a -80°C respectivamente. El bloque control estuvo conformado por especímenes frescos sin tratamiento de preservación. Se extrajo el ADN de cada espécimen, se cuantificó por espectrofotometría y se amplificó el espaciador interno transcrito 2 (ITS2) mediante PCR. Se realizó un análisis comparativo entre bloques de tratamiento usando la frecuencia de amplificación de ITS2. Resultados: Se obtuvo la amplificación de ITS2 de todos los especímenes sin preservación previa, del 60% de los preservados en sílica gel y del 20% de los preservados en congelación a -80°C y en etanol al 70%. Se halló diferencia estadísticamente significativa (p<0,05) entre las proporciones de amplificación observadas. No se obtuvo amplificación de los especímenes preservados en congelación a -20°C y no se observó una correlación por regresión logística (p>0,05) entre el índice DO260/DO280 y la concentración de ADN de los especímenes que presentaron amplificación de ITS2. Conclusiones: Los datos sugieren que la conservación en sílica gel o la congelación que se vayan a -80°C pueden ser las mejores condiciones para la preservación de especímenes Anopheles a utilizar en estudios moleculares posteriores.

Abstract: Los envenenamientos causados por serpientes del género Bothrops constituyen un problema de salud en algunos países tropicales, no solamente por la mortalidad sino por el alto porcentaje de incapacidades físicas que ocasionan. Las fosfolipasas A2 (PLA2) son componentes abundantes en los venenos bothrópicos y crotálicos, y se caracterizan por inducir da os irreversibles en el músculo. Considerando el aporte de las plantas en el tratamiento ofídico, este trabajo se enfocó en la búsqueda de inhibidores de las fosfolipasas, bien sea como alternativa terapéutica o como coadyuvante del tratamiento ofídico. De un tamizaje de 37 especies de plantas vasculares y briofitos contra las PLA2, se seleccionó la especie Murraya paniculata por su promisoria actividad preliminar. Se siguió con el fraccionamiento biodirigido por la evaluación de la actividad inhibitoria de la PLA2 y de la fracción con mayor actividad inhibitoria, por medio de cromatografía de gases acoplada a masas se identificaron metabolitos responsables de la actividad, para proponer un modelo de inhibición enzima-sustrato. Los resultados de este proyecto pretenden contribuir en la búsqueda de moléculas inhibitorias de las PLA2 en plantas vasculares, como la especie M. paniculata, que podrían llegar a ser una herramienta en el desarrollo de nuevos productos para mejorar el pronóstico del accidente ofídico. Envenomation by snakes of the genus Bothrops are a health problem in some tropical countries, not only because of the mortality but given the high percentage of physical disabilities it causes. Phospholipases A2 (PLA2) are abundant constituents in bothropic and crotalic venoms, characterized by inducing irreversible muscle damage. Due to the contribution of plants in the snakebite treatment, we focus on the search of phospholipases inhibitors, either as an alternative treatment or as a therapeutic adjuvant of the current treatments. From a screening of 37 vascular plants and bryophytes in the search for such inhibitors, the species Murraya paniculata was selected due to its promising preliminary activity. Starting from this point further work consisting of bioassay-guided fractionation followed by the evaluation of the inhibitory activity of PLA2, the metabolites responsible for the activity were detected through a tandem of gas chromatography - mass spectrometry in the most active fraction, to propose substrate-enzyme inhibition model. Abstract: aiming to contribute with the valorization of agricultural wastes that present disposal problems and the use of renewable energy sources, was researched the rice husk combustion in atmospheric bubbling fluidized bed reactor. the methodology was to evaluate the air excess influence in the flue gases and the ash characteristics. it was determined the gases composition (co, co2 and nox) and characterized the ash generated (xrd, xrf). 40% excess air promotes temperatures inside the reactor around 700oc with higher conversion efficiencies, but declining the amorphous silica potential of the ash. contrary behavior was evidenced at 128% excess air. the flue gases showed relative variability for each operating condition. in the co case, an average concentration of 200 ppm was found (on a dry basis of 11% o2) for air excess between 40% and 82,5%.

Abstract: This article discusses the causes of formation of micron-size protein structures in liquid plasma or serum of the patients with different diseases, which are accompanied by inflammatory reac-tions. Self-organizing processes in sessile dry-ing drops of natural and model biological liq-uids are used for study of possible mechanisms of development the protein phase instability in serum. There was shown that violation of opti-mal ratio between albumin and osmotic active components could lead to loss of albumin ag-gregative stability and albumin coagulation structures formation. Possible role of these structures in pathogenesis of inflammation is discussed.
