O FóRUM DE APOIO à FORMA O DOCENTE DE MATO GROSSO COMO LóCUS DE PRODU O DE POLíTICAS

Abstract: O objetivo central do presente estudo é analisar o papel do Fórum Permanente de Apoio à Forma o Docente no estado de Mato Grosso (Fórum/MT) como lócus de produ o de políticas educacionais para a forma o de professores. Parte‐se do pressuposto de que a formula o de políticas é um processo que ocorre em arenas de luta nas quais há sempre possibilidades de resiliências, de ressignifica o e de recontextualiza o. Entende‐se que o Fórum/MT, constitui‐se numa destas arenas de lutas. Este fato abre a possibilidade de caracterizá‐lo como uma comunidade epistêmica. O caminho metodológico adotado no estudofoi definido a partir de análises de textos políticos, que motivaram a cria o do Fórum/MT e de atas das reuni es. No Mato Grosso os dados indicam que as institui es públicas de educa o superior do Estado têm papel preponderante na oferta de cursos de forma o de professores. Foi também definido que o levantamento da demanda será feito pelos Centros de Forma o e Atualiza o dos Profissionais da Educa o Básica de Mato Grosso (Cefapro/MT), assim como a tarefa de forma o continuada.Conclui‐se que a cria o e atua o do Fórum/MT permite reiterar a ideia de política como um processo complexo de recontextualiza o de sentidos e proposi es, uma vez que há influencias de diversos setores e, de forma expressiva, pelo contexto da prática.
Transcriptional Analysis of Drought-Induced Genes in the Roots of a Tolerant Genotype of the Common Bean (Phaseolus vulgaris L.)

Abstract: In Brazil, common bean ( Phaseolus vulgaris L.) productivity is severely affected by drought stress due to low technology cultivation systems. Our purpose was to identify differentially expressed genes in roots of a genotype tolerant to water deficit (BAT 477) when submitted to an interruption of irrigation during its development. A SSH library was constructed taking as “driver” the genotype Carioca 80SH (susceptible to drought). After clustering and data mining, 1572 valid reads were obtained, resulting in 1120 ESTs (expressed sequence tags). We found sequences for transcription factors, carbohydrates metabolism, proline-rich proteins, aquaporins, chaperones and ubiquitins, all of them organized according to their biological processes. Our suppressive subtractive hybridization (SSH) library was validated through RT-qPCR experiment by assessing the expression patterns of 10 selected genes in both genotypes under stressed and control conditions. Finally, the expression patterns of 31 ESTs, putatively related to drought responses, were analyzed in a time-course experiment. Our results confirmed that such genes are more expressed in the tolerant genotype during stress; however, they are not exclusive, since different levels of these transcripts were also detected in the susceptible genotype. In addition, we observed a fluctuation in gene regulation over time for both the genotypes, which seem to adopt and adapt different strategies in order to develop tolerance against this stress.
Protein Folding Modulates the Swapped Dimerization Mechanism of Methyl-Accepting Chemotaxis Heme Sensors

Abstract: The periplasmic sensor domains GSU0582 and GSU0935 are part of methyl accepting chemotaxis proteins in the bacterium Geobacter sulfurreducens. Both contain one c-type heme group and their crystal structures revealed that these domains form swapped dimers with a PAS fold formed from the two protein chains. The swapped dimerization of these sensors is related to the mechanism of signal transduction and the formation of the swapped dimer involves significant folding changes and conformational rearrangements within each monomeric component. However, the structural changes occurring during this process are poorly understood and lack a mechanistic framework. To address this issue, we have studied the folding and stability properties of two distinct heme-sensor PAS domains, using biophysical spectroscopies. We observed substantial differences in the thermodynamic stability (ΔG = 14.6 kJ.mol?1 for GSU0935 and ΔG = 26.3 kJ.mol?1 for GSU0582), and demonstrated that the heme moiety undergoes conformational changes that match those occurring at the global protein structure. This indicates that sensing by the heme cofactor induces conformational changes that rapidly propagate to the protein structure, an effect which is directly linked to the signal transduction mechanism. Interestingly, the two analyzed proteins have distinct levels of intrinsic disorder (25% for GSU0935 and 13% for GSU0582), which correlate with conformational stability differences. This provides evidence that the sensing threshold and intensity of the propagated allosteric effect is linked to the stability of the PAS-fold, as this property modulates domain swapping and dimerization. Analysis of the PAS-domain shows that disorder segments are found either at the hinge region that controls helix motions or in connecting segments of the β-sheet interface. The latter is known to be widely involved in both intra- and intermolecular interactions, supporting the view that it's folding and stability are at the basis of the specificity and regulation of many types of PAS-containing signaling proteins.

Abstract: the enteropathogenic role of cytotoxic necrotizing factor (cnf)-producing escherichia coli was investigated by searching cnf genes among 2074 isolates from 200 children with and 200 without acute diarrhea in brazil. fourteen (7%) cases versus 10 (5%) control children carried at least one cnf positive isolate (p = 0.50) and most isolates expressed cnf type 1. dna sequences of virulence factors of extraintestinal pathogenic e. coli (expec) were detected in 78.6% of cnf1-producing isolates. besides not being associated with human acute diarrhea, the cnf1-producing isolates here identified may represent potential expec transitorily composing the normal intestinal flora.

Abstract: The occurrence of fim, pap and sfa operons in Escherichia coli isolated from breeders with salpingitis and chicks with omphalitis was evaluated. Analysis of 100 E. coli isolates, by colony hybridization tests, showed that 78 (78%) were fim+, one (1%) was sfa+, seven (7%) were fim+ associated with pap+, eigth (8%) were fim+ and sfa+, one (1%) was fim+pap+sfa+ and five (5%) isolates did not hybridize with any probe. These results suggest that fim adhesion-encoding operon plays an important role in pathogenesis of E. coli infection in chickens with salpingitis and omphalitis.

Abstract: Introduction: Diarrheagenic Escherichia coli (DEC) is a relevant cause of diarrhea, particularly among infants and young children in developing countries. Methodology: We compared the frequency, antimicrobial resistance, adherence, enterovirulence and genetic diversity of DEC isolates from Guarani indigenous population under five living in distinct villages in Brazil. Results: Of the 314 E. coli isolates from 57 children, with and without diarrhea, 15% (48/314) were classified in DEC categories: aEPEC (56%, 27/48), EAEC (35%, 17/48) and ETEC (8%, 4/48). ETEC belonged to plylogroup A, EAEC to groups A, B1, B2 and D, and aEPEC to phylogroups A, B1, and B2. EAEC exhibited the aggregative adherence phenotype while ETEC and aEPEC the aggregative and undefined patterns. Multidrug-resistance was detected in aEPEC, ETEC and EAEC while extensive drug-resistance was found in EAEC and aEPEC. RAPD typing revealed a genetically diverse bacterial population. Conclusion: This is the first report regarding aspects of DEC in an indigenous Brazilian population, showing that Guarani children are DEC carriers and that antimicrobial resistance at high levels is widely disseminated among these enteropathogens.
