A proteomic analysis of the functional effects of fatty acids in NIH 3T3 fibroblasts

Abstract: Found in the majority of tissues of the body, fibroblasts are responsible for the synthesis and secretion of most extracellular matrix (ECM) components, such as proteoglycans, collagens, laminin and fibronectin, which bind to proteins expressed on cell surfaces thus modulating physiologic responses. Fibroblasts can also secrete proteinases, including matrix metalloproteinases and plasminogen, hence playing an important role in ECM degradation and tissue remodeling [1-3].The dysregulation of fibroblast biology is associated with several diseases and pathological states, including deficient wound healing [4-6], pulmonary diseases [7,8], cardiovascular diseases [9,10] and cancer [11]. Therefore, the discovery of new therapies modulating fibroblast biology could be a powerful target to develop treatment against such diseases. Oleic (18:1n-9), linoleic (18:2n-6) and palmitic (16:0) acids are the most abundant fatty acids in the western diets [12]. The latter is a saturated fatty acid found in palm oil, butter, milk, cheese and meats, whereas oleic acid is a monounsaturated fatty acid found in olive oil, meat, eggs and milk and linoleic acid is a polyunsaturated fatty acid found in soybean, sunflower, safflower and corn oils. The effects of prostaglandin derived from n-3 and n-6 fatty acids on specific protein expression (e.g. COX-2 and IL-6) in NIH 3T3 fibroblasts [13] and some aspects of the impact of fatty acids on process involving fibroblast function [14-16] have been reported. However, no systematic study has assessed the pleiotropic effects of fatty acids on fibroblast protein expression. Therefore, the aim of this study was to assess the effects of oleic (OLA), linoleic (LNA) or palmitic acids (PAM) on protein expression of NIH 3T3 fibroblasts, as determined by 2D-DIGE, i.e., separation of proteins by isoelectric focusing in the first dimension followed by SDS-PAGE in the second. Abstract: Este texto procura mostrar que a metodologia através da resolu o de problemas pode ser trabalhada sem grandes rupturas no trabalho docente dos professores que ensinam Matemática. Entretanto, para que isso aconte a, os professores devem, em um primeiro momento, analisar e discutir suas concep es e seus conhecimentos sobre educa o, conhecimento matemático, ensino, aprendizagem, avalia o, entre outros elementos presentes no trabalho docente para verificar se s o consistentes diante dessa perspectiva de ensinar e de aprender Matemática. Assim, com concep es e conhecimentos coerentes com a metodologia da resolu o de problemas, os professores podem enfrentar com sucesso as situa es inesperadas e n o pensadas que, muitas vezes, ocorrem quando utilizam a resolu o de problemas n o só como uma metodologia, mas também como uma filosofia de ensino. A experiência do professor como resolvedor de problemas e a reflex o, com especialistas, sobre os resultados de uma aula de Matemática focada na resolu o de problemas s o condi es essenciais para potencializar esse caminho metodológico bastante promissor para a educa o matemática. This text is intended to show that problem solving methodology can be worked with no big ruptures in Mathematics teaching. However, this is possible only if teachers, in a first moment, analyze and discuss their conceptions and knowledge about education, mathematical knowledge, teaching and learning, evaluation, among other elements in teaching work, to verify if they are consistent in this perspective of teaching and learning Mathematics. So, with conceptions and knowledge that match problem solving methodology, teachers can successfully face unexpected and unpredictable situations that, many times, happen when problem solving is seen not only as a methodology but also as a teaching philosophy. The teacher's experience as a problem solver and consideration, with experts, about results in a Mathematics class based on problem solving are essential conditions to support that way of teaching, which is very promising to Mathematics education. A problem that explores fractions properties shows a possible teaching work on that methodology that intends to make mathematical learning more meaningful for students.
High-Fat Diet Induces Apoptosis of Hypothalamic Neurons

Abstract: Consumption of dietary fats is amongst the most important environmental factors leading to obesity. In rodents, the consumption of fat-rich diets blunts leptin and insulin anorexigenic signaling in the hypothalamus by a mechanism dependent on the in situ activation of inflammation. Since inflammatory signal transduction can lead to the activation of apoptotic signaling pathways, we evaluated the effect of high-fat feeding on the induction of apoptosis of hypothalamic cells. Here, we show that consumption of dietary fats induce apoptosis of neurons and a reduction of synaptic inputs in the arcuate nucleus and lateral hypothalamus. This effect is dependent upon diet composition, and not on caloric intake, since pair-feeding is not sufficient to reduce the expression of apoptotic markers. The presence of an intact TLR4 receptor, protects cells from further apoptotic signals. In diet-induced inflammation of the hypothalamus, TLR4 exerts a dual function, on one side activating pro-inflammatory pathways that play a central role in the development of resistance to leptin and insulin, and on the other side restraining further damage by controlling the apoptotic activity.

Abstract: this paper discusses the interrelations between real wages, the real exchange rate and productivity in the brazilian economy during the nineties. this period is particularly interesting as it combined trade liberalization, the valorization of the real and relatively high rates of growth of labor productivity. with this objective, a vec model was used to test a theoretical model based on monopolisitic competition. the model predicted correctly the association between real wages and productivity, but it did not perform well as regards real wages and unemployment.

Abstract: in this article i analyze the hegemonic discourse articulated by the disciplinary communities that research the science teaching, with the goal of identifying the meanings that are being produced in the initial years of elementary school. i take as reference the productions of the two first and the two last "encontros nacionais de pesquisa em educa??o em ciências" (enpec), that presented reflections directed towards this school level, in an effort to understand the constitution of the process of a speech that intends to establish what is and what should be science teaching in the elementary levels of school, and also to establish which the formative needs are so that teaching can be exerted in that direction. using laclau and mouffe's (2004) theory of discourse, which enables me to perceive it as a hybrid discourse that pervades the different curricular contexts of production.
Habitat utilization and CAM occurrence among epiphytic bromeliads in a dry forest from southeastern Brazil

Abstract: we studied the community and habitat occupation of epiphytes to understand how these plants cope with a supposedly stressful habitat: i) how general epiphytes occupy tree trunks, ii) how epiphytic bromeliads, occupy their supportive trees, iii) how cam bromeliads are spatially distributed. the study was done in the dry forest of jacarepiá, state of rio de janeiro. data collection on epiphytes, phorophytes, and trees was based on the point-center quarter method. the photosynthetic pathway of the bromeliad species was determined using isotope ratio mass spectrometry. the presence of gesneriaceae, araceae, and cactaceae indicates that some humidity is present in the area allowing the presence of supposedly less-specialized epiphytes. there was no correlation between epiphyte abundance and phorophyte diameter, and phorophytes had larger sizes than trees that do not host epiphytes. there was correlation between tree diameter and bromeliad abundance, and lack of correlation between diameter and bromeliad richness. only one species was typical of the understorey and one was typical of the canopy, while intermediate heights were occupied by different species. the only c3 bromeliad species (vriesea procera (mart. ex schult.f.) wittm.) was significantly more exposed than the other species. if cam occurrence is related to water economy, the fact that a c3 species is subjected to more exposed conditions is remarkable. further comments are presented on the proportion between cam bromeliad species and abundance in dry forest. regarding life forms, holoepiphytes, as opposed to hemiepiphytes, showed not to be restricted by the phorophyte's diameter suggesting a more successful establishment of this life form.

Abstract: about 1 million cases of transplantation were registered around the world. the first pancreas transplantation in brazil was in 1996, proceeded by a medical team of s?o paulo. the objetctive of this study was to characterize the socio- demographic and epidemiological profile of the receivers in list and those submitted to this procedure. it is an inter-relational retrospective study considereing the period from 1996 to 2004. results demonstrated the predominance of women, from white race, and, with an average of 35 years. the origin of the patients was the states of s?o paulo and minas gerais.
Conceptual metaphor and comprehension in business writing

Abstract: Recent research studies in cognitive linguistics (Gibbs, 1999; K vecses, 2005; Lakoff, 1987; Lakoff & Johnson, 1980; Lakoff & Johnson, 1999) have demonstrated that metaphor is not merely a figure of speech. The findings of these studies have shown that metaphor influences a good deal of how people think as it comprises a specific mental, cross-domain mapping in the conceptual system (Balaban, 1999; Ibarretxe-Antuňanu, 1999; Lakoff, 2006; Lakoff & Johnson, 1980). This article looks at whether employing different conceptual metaphors in different versions of the same business report will have an effect on reader comprehension. A small-scale research study was conducted with a group of second-year university students, in which they were given one of two texts concerning the recovery of the economy. Both texts were adapted from a newspaper report and seeded with metaphors and metaphoric expressions. The multiple-choice questions that followed aimed to determine to what extent the readers’ comprehension and interpretation of the report were influenced by the different conceptual metaphors used. The findings indicate that language that served to introduce the sources or targets directly into the content did not necessarily have an effect on the processing of subsequent metaphors involving these concepts; however, it appeared that the surface patterns of metaphorical discourse did affect the inferences drawn about the different conceptual metaphors. It can thus be concluded that a specific metaphoric framework in written discourse does influence the interpretation of the content.

Abstract: Neste trabalho procuro identificar o cientificismo como marca constituinte do discurso educacional, que favorece e sustenta o processo de naturaliza o dos conteúdos de ensino. Argumento que esta naturaliza o constituí e expressa uma dada hegemonia que privilegia um determinado modelo cultural em detrimento de outros possíveis. O trabalho se estrutura em três momentos. No primeiro procuro caracterizar, em linhas gerais, o pós-estruturalismo como modo de pensamento que questiona aspectos centrais do Humanismo e do Renascimento, que se expressam no projeto de modernidade. Depois busco identificar o processo de escolariza o, que se desenvolve a partir do século XIX, e se consolida no século XX, como um processo a servi o da propaga o do projeto cultural moderno homogeneizador. Neste exercício reflexivo lan o m o da análise dos relatos colhidos durante as entrevistas que realizei com professoras da rede municipal de ensino da cidade do Rio de Janeiro no período em que acontecia a implementa o da proposta curricular da rede, a Multieduca o (1996). Destes relatos emergiram as quest es que motivam minhas reflex es no doutorado, e que têm encontrado no paradigma pós-estruturalista um referencial produtivo para análise.
