Krüppel-like factor 5 is a crucial mediator of intestinal tumorigenesis in mice harboring combined ApcMin and KRASV12 mutations

Abstract: At 12 weeks of age, ApcMin/KRASV12 mice had three times as many intestinal tumors as ApcMin mice. This increase in tumor number was reduced by 92% in triple transgenic ApcMin/KRASV12/Klf5+/- mice. The reduction in tumor number in ApcMin/KRASV12/Klf5+/- mice was also statistically significant compared to ApcMin mice alone, with a 75% decrease. Compared with ApcMin/KRASV12, tumors from both ApcMin/KRASV12/Klf5+/- and ApcMin mice were smaller. In addition, tumors from ApcMin mice were more distally distributed in the intestine as contrasted by the more proximal distribution in ApcMin/KRASV12 and ApcMin/KRASV12/Klf5+/- mice. Klf5 levels in the normal-appearing intestinal mucosa were higher in both ApcMin and ApcMin/KRASV12 mice but were attenuated in ApcMin/KRASV12/Klf5+/- mice. The levels of β-catenin, cyclin D1 and Ki-67 were also reduced in the normal-appearing intestinal mucosa of ApcMin/KRASV12/Klf5+/- mice when compared to ApcMin/KRASV12 mice. Levels of pMek and pErk1/2 were elevated in the normal-appearing mucosa of ApcMin/KRASV12 mice and modestly reduced in ApcMin/KRASV12/Klf5+/- mice. Tumor tissues displayed higher levels of both Klf5 and β-catenin, irrespective of the mouse genotype from which tumors were derived.Results of the current study confirm the cumulative effect of Apc loss and oncogenic KRAS activation on intestinal tumorigenesis. The drastic reduction in tumor number and size due to Klf5 heterozygosity in ApcMin/KRASV12 mice indicate a critical function of KLF5 in modulating intestinal tumor initiation and progression.Cancer is the result of deregulated cellular homeostasis and is typically characterized by increased proliferation and/or decreased apoptosis [1]. Abstract: The reading and the readings of young people are at the crossroads of social and cultural mediations in which the school institution and the family share the first role. During decades, work on the reading of the young people distinguished the readings for leisure and the readings for school. Since about fifteen years, this cleavage does not correspond any more to reality. Well before the learning of the reading, the desire of reading takes its source in the exercise of the mother tongue and in the family uses of print. Reading skills come to reinforce it. The acquisition of a reading practice and the construction of a reader behaviour take place at home according to the choices of youth literature generally presented by the primary school and the beginning of secondary school. With the age of the secondary school, the reader builds two different universes of reading. One is composed of texts prescribed by the teachers and the other of readings called "for oneself". Whatever the age and skills, the dynamics of the activity of reading remains fragile. It rests on emotional investments.
Adenoma Formation following Limited Ablation of p120-Catenin in the Mouse Intestine

Abstract: p120 loss destabilizes E-cadherin and could therefore result in tumor and/or metastasis-promoting activities similar to those caused by E-cadherin downregulation. Previously, we reported that p120 is essential in the intestine for barrier function, epithelial homeostasis and survival. Conditional p120 ablation in the mouse intestine induced severe inflammatory bowel disease, but long-term cancer-related studies were impossible because none of the animals survived longer than 21 days. Here, we used a tamoxifen-inducible mouse model (Vil-Cre-ERT2;p120fl/fl) to limit the extent of p120 ablation and thereby enable long-term studies. Reducing p120 KO to ~10% of the intestinal epithelium produced long-lived animals outwardly indistinguishable from controls. Effects of prolonged p120 absence were then evaluated at intervals spanning 2 to 18 months. At all time points, immunostaining revealed microdomains of p120-null epithelium interspersed with normal epithelium. Thus, stochastic p120 ablation is compatible with crypt progenitor cell function and permitted lifelong renewal of the p120-null cells. Consistent with previous observations, a barrier defect and frequent infiltration of neutrophils was observed, suggesting that focal p120 loss generates a microenvironment disposed to chronic inflammation. We report that 45% of these animals developed tumors within 18 months of tamoxifen induction. Interestingly, β-catenin was upregulated in the majority, but none of the tumors were p120 null. Although further work is required to directly establish mechanism, we conclude that limited p120 ablation can promote tumorigenesis by an indirect non-cell autonomous mechanism. Given that byproducts of inflammation are known to be highly mutagenic, we suggest that tumorigenesis in this model is ultimately driven by the lifelong inability to heal chronic wounds and the substantially increased rates of stochastic gene mutation in tissue microenvironments subjected to chronic inflammation. Indeed, although technical issues precluded direct identification of mutations, β-catenin upregulation in human colon cancer almost invariably reflects mutations in APC and/or β-catenin.
The POZ-ZF Transcription Factor Kaiso (ZBTB33) Induces Inflammation and Progenitor Cell Differentiation in the Murine Intestine

Abstract: Since its discovery, several studies have implicated the POZ-ZF protein Kaiso in both developmental and tumorigenic processes. However, most of the information regarding Kaiso’s function to date has been gleaned from studies in Xenopus laevis embryos and mammalian cultured cells. To examine Kaiso’s role in a relevant, mammalian organ-specific context, we generated and characterized a Kaiso transgenic mouse expressing a murine Kaiso transgene under the control of the intestine-specific villin promoter. Kaiso transgenic mice were viable and fertile but pathological examination of the small intestine revealed distinct morphological changes. Kaiso transgenics (KaisoTg/+) exhibited a crypt expansion phenotype that was accompanied by increased differentiation of epithelial progenitor cells into secretory cell lineages; this was evidenced by increased cell populations expressing Goblet, Paneth and enteroendocrine markers. Paradoxically however, enhanced differentiation in KaisoTg/+ was accompanied by reduced proliferation, a phenotype reminiscent of Notch inhibition. Indeed, expression of the Notch signalling target HES-1 was decreased in KaisoTg/+ animals. Finally, our Kaiso transgenics exhibited several hallmarks of inflammation, including increased neutrophil infiltration and activation, villi fusion and crypt hyperplasia. Interestingly, the Kaiso binding partner and emerging anti-inflammatory mediator p120ctn is recruited to the nucleus in KaisoTg/+ mice intestinal cells suggesting that Kaiso may elicit inflammation by antagonizing p120ctn function.
From physical mobility to psychological mobilization: The benefits of changing location and caregiver in a mobile team for adolescent psychiatry  [PDF]

We have developed a mobile team for adolescents with psychological difficulties that can intervene within 48 hours, if necessary. Practically, caregivers working in pairs go wherever the adolescents are (home, General Practioner’s office, etc.), travelling in a motorhome designed as a mobile office. This method allows us to approach more closely the adolescents and their families in the here and now that corresponds to our societal evolution. Most of the adolescents seen by the mobile team do not express any explicit demand but the demand is expressed by the professionals in direct contact with these adolescents in difficulty (school professionals, General Practitioners, etc.), who alert the mobile team and solicit the adolescents’ parents to call the mobile team. One characteristic of this mobile team is the mobility of setting (by changing locations, including the use of a mobile office, and caregivers, so that multiple representations are mobilized). Statistical analyses conducted on 520 adolescents followed by the mobile team showed that the first meetings tend to take place in the family’s home, subsequently transferring to the mobile office and finally in the community mental health centres. There is therefore a transition from the living space (a place that is part of the here and now) to the caregiving space (a space of thought and for thinking), with the mobile office serving as an intermediate step in this movement towards accessing care. Furthermore, there was a significant association between changes of locations and continuity of care. Thus, there were significantly more breakdowns in continuity when the consultations all take place in a single location (family home, mobile office, community centre). The results and their therapeutic implications will be discussed in this article, especially with regard to the key role played by physical movement in psychological mobilization, and by plurality of representations associated with different locations and caregivers.

Abstract: Economic, demographic, social, technological and political changes worldwide are putting academic institutions under intense pressures. In response, universities are adopting new managerial approaches to their activities: lean, six sigma and lean six sigma. A portrait of this experience emerges from reviewing the literature published over the past decade using the databases Compendex & INSPEC/Engineering Village and Scopus. These approaches have been applied primarily on a highly localized basis to teaching-related processes or to services such as financing, data processing and building maintenance. Some of the challenges raised are not unknown outside of the university setting. The complexity of universities, the difficulties of interpreting notions such as the client, added value, and the connexions between teaching and research, make the implementation of these approaches difficult. While the few measured results available suggest that they do hold promise, their impact remains to be determined.
Integration of Occupational Health and Safety into Lean Manufacturing: Quebec Aeronautics Case Study  [PDF]

Abstract: The literature remains divided the ideal conditions for integration of occupational health and safety (OHS) into continuous improvement initiatives based on the principles of lean manufacturing. Few significant results and little documentation exist on the effectiveness, sustainability and the degree of integration. This paper outlines some possible explanations for the non-success of this integration and examines the hypothesis that integration is unsuccessful when the lean-based improvements themselves suffer from sustainability issues. This research project addressed the case of a Québec manufacturing company and focused on musculoskeletal injury (MSI). Twenty-four employees participated in the study for two months. Corporate documentation was analyzed. Individual interviews were used to document workers’ perception of their work environment. Continuous improvement interventions were observed to identify possible risks of MSI, and obtain a general portrait of OHS and continuous improvement in this company. The majority of the employees surveyed believed that the continuous improvement initiatives were short-lived. They also reported they experienced MSI-related discomfort following organizational changes. Examination of several initiatives showed that results were not maintained over time. The integration of OHS into continuous improvement is not systematic. When this integration is apparent, OHS goals are not always met and are rarely subject to monitoring. Taking OHS into consideration, implementing lean principles does not directly guarantee a reduction in cases of MSI. The integration of OHS into continuous improvement activities is reflected in the systematic consideration of OHS in driving lean improvements. The results of this project will help to develop a better approach to the implementation of lean to improve this integration in our industries.
Resilience Engineering for Sustainable Prevention in the Manufacturing Sector: A Comparative Study of Two Methods of Risk Analysis  [PDF]

Abstract: Manufacturing businesses today have become complex sociotechnical systems. They must manage a variety of risks, including those related to occupational health and safety and emergent ones. In this article, we examine the possibility of using resilience engineering to provide a framework for the management of OHS and emergent risks in manufacturing businesses. More specifically, we compare failure mode effect and criticality analysis (FMECA) to functional resonance analysis method (FRAM), the former being an established risk analysis method in the manufacturing sector and the latter being a relatively novel method used primarily in high-risk sectors such as air transportation, hospitals and railways. These two methods differ in several respects, but comparison reveals their relative advantages as well as their complementarity. Our principal finding confirms the advantages of the functional resonance analysis method in risk management for complex systems. Although failure mode effect and criticality analysis is more effective for understanding technical risks, functional resonance analysis method provides better understanding of the system as a whole and allows more efficient management of organizational risks as well as risks associated with the variability of human performance.

Abstract: [Fran ais] Le site BiomeTTico vise à répondre aux difficultés soulevées par l’apprentissage de la traduction spécialisée (biomédecine). Il s’agit d’un multi-outil informatique d’aide à l’enseignement de la traduction biomédicale au baccalauréat et aux cycles supérieurs, con u en fonction d’une approche cognitive de l’apprentissage et de la pratique de la traduction. Prenant en compte différentes catégories d’utilisateurs, il se veut également une plate-forme de valorisation des travaux de recherche, notamment au 2e et au 3e cycle, et, dans une certaine mesure, un site de référence pour les traducteurs professionnels. L’organisation du site est fondée sur des principes d’utilisabilité, d’interactivité et de participation collaborative au contenu, adaptés à un contexte pédagogique. Il intègre différents produits de la recherche menée ces dernières années et il est appelé à évoluer dans le temps. Bien que son contenu soit spécifique à la biomédecine, sa structure est réutilisable dans d’autres contextes d’enseignement de la traduction. [English] BiomeTTico is a Web site intended to respond to various problems related to the acquisition of translation competence and knowledge in LSP (languages for specific purposes ; here, biomedicine), as well as to enhance the value of research projects of post-graduate students. Designed from a cognitive perspective, this is a multi-tool helping the teaching of biomedical translation at different levels, from beginners to advanced students at the doctorate level. It takes into account a number of principles such as usability, interactivity and collaborative participation, as well as different types of users. Furthermore, it integrates a number of products resulting from the research activities over the past few years and is meant to evolve with time. Although the content of the site is specific to biomedicine, the structure may be used in other translation teaching contexts.
On the Coupling between Helium Settling and Rotation-Induced Mixing in Stellar Radiative Zones: II- Application to light elements in population I main-sequence stars

Abstract: In the two previous papers of this series, we have discussed the importance of t he $\mu$-gradients due to helium settling on rotation-induced mixing, first in a n approximate analytical way, second in a 2D numerical simulation. We have found that, for slowly rotating low mass stars, a process of ``creeping paralysis" in which the circulation and the diffusion are nearly frozen may take place below the convective zone. Here we apply this theory to the case of lithium and beryll ium in galactic clusters and specially the Hyades. We take into account the rota tional braking with rotation velocities adjusted to the present observations. We find that two different cells of meridional circulation appear on the hot side of the "lithium dip" and that the "creeping paralysis" process occurs, not dir ectly below the convective zone, but deeper inside the radiative zone, at the to p of the second cell. As a consequence, the two cells are disconnected, which ma y be the basic reason for the lithium increase with effective temperature on thi s side of the dip. On the cool side, there is just one cell of circulation and t he paralysis has not yet set down at the age of the Hyades; the same modelisatio n accounts nicely for the beryllium observations as well as for the lithium ones .
