Voltage profile generation for simultaneous multi-protein detection in western blot analysis  [PDF]

Abstract: Western blotting is a popular technique for examining expression levels of proteins using gel-based electrophoretic fractionation followed by blotting and antibody reactions. Although this is a mature technique, one of the major limitations is the need to prepare an individual electrophoretic gel for each of the protein species to be analyzed. Since most analyses require the detection of multiple protein species, a procedure that allows utilization of a single gel for detecting multiple protein species should significantly save time and resources. In this paper, we developed a novel multiprotein detection device, which enabled simultaneous detection of several proteins species from a single electrophoretic gel. In this device, a protein transfer unit utilized a multi-anode plate that generated a non-uniform voltage profile. This voltage profile enabled uniform transfer regardless of molecular mass of proteins. In vitro experiments using samples, isolated from boneforming osteoblast cells, showed that the expression levels of 5 - 7 different proteins were detectable in the presence and absence of mechanical stimulation that activated genes necessary for bone formation. The result supports the notion that through simultaneous detection of multiple protein species, the described device contributes to reduction in procedural time and sample amounts, as well as a removal of variations among multiple gels.

Abstract: This study presents the development of an innovative artificial finger-like device that provides position specific mechanical loads at the end of the long bone and induces mechanotransduction in bone. Bone cells such as osteoblasts are the mechanosensitive cells that regulate bone remodelling. When they receive gentle, periodic mechanical loads, new bone formation is promoted. The proposed device is an under-actuated multi-fingered artificial hand with 4 fingers, each having two phalanges. These fingers are connected by mechanical linkages and operated by a worm gearing mechanism. With the help of 3D printing technology, a prototype device was built mostly using plastic materials. The experimental validation results show that the device is capable of generating necessary forces at the desired frequencies, which are suitable for the stimulation of bone cells and the promotion of bone formation. It is recommended that the device be tested in a clinical study for confirming its safety and efficacy with patients.

Abstract: Blotting is a common technique widely used for molecular analysis in life sciences. The Western blot, in particular, is a process of transferring protein samples from a polyacrylamide gel to a blotting membrane and detecting the levels of specific proteins through reactions with primary and secondary antibodies. The state-of-the-art of Western blotting usually generates one blotting membrane per gel. However, multiple copies of blots are useful in many applications. Two blotting copies from a single protein gel, for instance, can be used for identifying a total amount of proteins of interest as well as its specific subpopulation level such as a phosphorylated isoform. To achieve this multi-blotting operation from a single gel, we modified a blotting procedure and developed a novel blotting device. The device consisted of a multi-anode plate and a microcontroller. It was designed to generate a well-controlled electrophoretic voltage profile, which allowed a quasi-uniform transfer of proteins of any size. The prototype device was built and its operation procedure was described. The experimental results clearly supported the notion that the described device was able to achieve multiple blotting from a single gel and reduce time and cost for protein analysis.
A New Multiscale Representation for Shapes and Its Application to Blood Vessel Recovery

Abstract: In this paper, we will first introduce a novel multiscale representation (MSR) for shapes. Based on the MSR, we will then design a surface inpainting algorithm to recover 3D geometry of blood vessels. Because of the nature of irregular morphology in vessels and organs, both phantom and real inpainting scenarios were tested using our new algorithm. Successful vessel recoveries are demonstrated with numerical estimation of the degree of arteriosclerosis and vessel occlusion.
Technology Development via Human Resources Exploitation: Evidence from Southeast Asian Countries  [PDF]

Abstract: The purpose of this article is to summarize and integrate the articles appearing in this special issue within the unique technology development (TD) and human resources (HR) of Southeast Asian countries. This paper examines how the synergy between TD and HR can lead to improved national economic performance. We proposed that better HR exploitation can sustain the capabilities of TD. Thus, the detail indexes are first evaluated with each category, which constructs the basic meanings of TD and HR. Consequently, the contexts of TD are clustered by different patterns with similar attributes in each nation. Each pattern can monitor the performance of HR in specific TD pattern. In this empirical study, we discover that the leading three countries (Singapore, Malaysia, and Thailand) have a strong positive rank correlation between TD and HR; In contrast, the rest of countries are poor rank correlation with regard to the two categories. These results mention that the level of TD, which depends on HR exploitation, is an important indicator of technological capability and even a proof of economic growth. We also examine the TD policies with strategic grid and offer a better road path of TD to follow, not only for those type countries to retune their policy, but also to parallel development in HR areas—such as the level of education upgrading, labor force environment improving, man power utilizing. Finally, Southeast Asian countries should calibrate feasible HR when use it to elaborate TD.
Left Atrial Volume Index in Patients with Dilated Cardiomyopathy—Correlation with Left Ventricular Function  [PDF]

Abstract: Background: The aim of this study was to determine the relation of left atrial (LA) volume and LA volume index with left ventricular function and to determine the association of duration of symptoms and left atrial volume index in patients with dilated cardiomyopathy. Materials and Methods: This was an observational, single centre study conducted in India. A total of 50 patients who were admitted to department of cardiology from July, 2008 to February, 2009 with diagnosis of dilated cardiomyopathy and an ejection fraction of <40% were included. Results: Of the 50 patients, 34 (68%) were males. 27 (54%) patients were in NYHA class II and 23 (46%) patients were in NYHA class III. LA volume was found to be ≥40 ml in all patients. LV function and LA volume were found to be correlated (r = -0.789, p < 0.01). Similarly, there was a correlation between LV function and LA volume index (r = -0.826, p < 0.01). There was no correlation between LA volume index and duration of symptoms (r = 0.04). Conclusion: It can be concluded that there is a strong inverse correlation between LA volume and left ventricular function and also between LA volume index and left ventricular function. The patients with NYHA class III were having larger left atrial volume than those with NYHA class II. Moreover, the duration of symptoms has no correlation with left atrial volume index.
Angiographic Profile and Treadmill Test Relationship of Women with Chest Pain Suggestive of Coronary Artery Disease  [PDF]

Abstract: Aim: The aim of present study was to analyze the prevalence and pattern of coronary artery disease (CAD) among women who underwent coronary angiography (CAG) and Treadmill Test (TMT). Methods: A total of 100 consecutive women patients for coronary angiogram were studied at Department of Cardiology, Government Medical College, Kottayam, Kerala, India on over a period of 6 months from February 2008. Total 65 women completed TMT stress test. The patients were subjected to CAG and TMT. Data were analyzed by using SPSS 20.0 software. Results: A total of 100 women patients were included. Among that 66% patients had hypertension and 47% patients had diabetes mellitus. The CAG revealed the presence of 17%, 13% and 15% of single vessel disease, double vessel disease, and triple vessel disease respectively. Vessel involvements were notably higher in positive TMT values during stress test. The sensitivity and specificity of TMT for detecting coronary artery disease in women were 61% and 69% respectively. Conclusion: It is concluded that, the findings of the present study effectively demonstrated higher predictive value of TMT in women with chest pain suggestive of CAD.
Respiratory rehabilitation with abdominal weights: a prospective case study  [PDF]

Abstract: Objective: Abdominal weights was used to strengthen the diaphragm of a C6 ASIA (A) tetraplegic subject with the aim of studying the long term effect of the technique as a part of respiratory rehabilitation. Setting: Department of Physical Medicine and Rehabilitation, Christian Medical College, Vellore, Tamil Nadu, India. Study Design: Prospective case study. Material and methods: The peak EMG amplitude of the diaphragm (DIA), intercostals (INT) and sternoclidomastiod (SCM) were assessed using a surface EMG and inspired lung volume (ILV) was assessed using an adjustable portable spirometer. The measurements were repeated after 3, 6, 9 and 12 months of inspiratory muscle training for a period of 15 minutes daily, 6 days a week for 12 months. Results and discussion: Peak amplitudes recorded by the EMG of DIA and SCM muscles showed a progressive increase, INT muscle did not show a consistent change. INV showed a gradual rise from 1772ml to 2760 ml over the study period. These values have the following significance: 1) Use of abdominal weights as a part of respiratory rehabilitation has beneficial long term effects; 2) In patients with tetraplegia, respiratory muscles in particular the diaphragm, are trainable in terms of muscle efficiency; 3) The improvement in the muscle efficiency obtained during the early rehabilitation can be maintained or improved using simple non sophisticated exercises like abdominal weights post discharge. Conclusions: Abdominal weights can be used as an effective adjunct to pulmonary rehabilitation in improving the efficiency of diaphragm on a long term basis, thereby reducing the risks associated with pulmonary complications.
Sub-Optimal Generation Portfolio Variance with Rate of Return Regulation  [PDF]

Abstract: This paper demonstrates that continuation of traditional rate-of-return electric utility regulation of transmission and distribution assets will impede the ability of customers to optimize their generation portfolios. Under linear price regulation, with increasing (decreasing) returns to scale customers will choose a more (less) risky generation portfolio than they would with no transmission and distribution asset rate-of-return regulation. Similar problems arise under non-linear (two-part) pricing of transmission and distribution assets. When the per-unit price is set at marginal cost, with increasing (decreasing) marginal cost, customers will choose a more (less) risky generation portfolio than they would with no transmission and distribution asset regulation. With price caps the optimal generation portfolio is chosen.

Abstract: The nexus of neo-liberalist influences in our current risk society has produced a crisis for both New Zealand sociology and Social Work, playing out in practice domains and in the academy. This paper argues that by co-habituating and co-operating, we may have a tangible way forward. One of the biggest challenges for Social Work practitioners is to come to terms with the role of theory in the practice of their discipline—a discipline that is often fast-paced, but increasingly focused on dealing with one client at a time, and often reduced to a dyad emphasis in practise: that of client and worker. One of the biggest challenges for the sociologist embarking on a career in research is to come to terms with sociology as methodological toolkit for social activism where knowledge of theory can be applied toward sustained societal change. Both offer a methodological approach to understanding the human condition in context. Both disciplines are at risk because of neo-liberalisation, and this, we argue must be avoided by a move toward each other.
