Inhibition of glutamate dehydrogenase and insulin secretion by KHG26377 does not involve ADP-ribosylation by SIRT4 or deacetylation by SIRT3

Abstract: We investigated the mechanisms involved in KHG26377 regulationof glutamate dehydrogenase (GDH) activity, focusing onthe roles of SIRT4 and SIRT3. Intraperitoneal injection of micewith KHG26377 reduced GDH activity with concomitant repressionof glucose-induced insulin secretion. Consistent withtheir known functions, SIRT4 ribosylated GDH and reduced itsactivity, and SIRT3 deacetylated GDH, increasing its activity.However, KHG26377 did not affect SIRT4-mediated ADP-ribosylation/inhibition or SIRT3-mediated deacetylation/activationof GDH. KHG26377 had no effect on SIRT4 protein levels,and did not alter total GDH, acetylated GDH, or SIRT3 proteinlevels in pancreatic mitochondrial lysates. These results suggestthat the mechanism by which KHG26377 inhibits GDHactivity and insulin secretion does not involve ADP-ribosylationof GDH by SIRT4 or deacetylation of GDH by SIRT3.

Abstract: We evaluated the expression of the costimulatory moleculesCD80 and CD83 and major histocompatibility (MHC) class IIinduced by 2,4-dinitrofluorobenzene (DNFB) in the RAW264.7 macrophage cell line. In contrast to the previously reportedeffect of DNFB on dendritic cells, CD86 expression didnot change. Furthermore, we observed that the CD83 expressionlevel transiently increased and then decreased.Induction of CD80 and MHC class II molecule expression anda decrease in CD83 expression by DNFB in vitro were alsoconfirmed in splenocytes of BALB/c and NC/Nga mice.However, DNFB did not influence CD83 expression in peritonealCD11b+ cells from BALB/c or NC/Nga mice. Detailedin vivo experiments and further studies on the possible contributionof CD11b+ cells to induce atopic dermatitis (AD)would be helpful to attain a better understanding of ADpathogenesis.

Abstract: Nardostachys jatamansi (NJ) belonging to the Valerianaceae familyhas been used as a remedy for gastrointestinal inflammatory diseasesfor decades. However, the potential for NJ to ameliorate alcoholicchronic pancreatitis (ACP) is unknown. The aim of thisstudy was to examine the inhibitory effects of NJ on ACP.C57black/6 mice received ethanol injections intraperitoneally for 3weeks against a background of cerulein-induced acute pancreatitis.During ACP, NJ was ad libitum administrated orally with water.After 3 weeks of treatment, the pancreas was harvested for histologicalexamination. NJ treatment increased the pancreatic acinarcell survival (confirmed by amylase level testing) and reduced collagendeposition and pancreatic stellate cell (PSC) activation. In addition,NJ treatment reduced the activation but not death of PSC. Inconclusion, our results suggest that NJ attenuated ACP through theinhibition of PSC activation.

Effects of Annealing and Removal of the Water-Soluble Fraction of Dry-Milled Rice Flour on the Texture of Cooked Rice Noodles  [PDF]

Abstract: Rice noodles were prepared using dry-milled rice flours, which were treated by annealing and removing the water-soluble fraction to improve the quality of noodles without using chemical additives. The combined treatment (TC) with annealing and water-soluble fraction removal decreased the cooking losses for Goamibyeo but not for Chenmaai and Milyang260, which had soft kernels and contained less damaged starch than the hard kernel rice after milling. TC significantly reduced the hardness and adhesiveness of cooked noodles, and increased the cohesiveness according to the texture profile analysis. A sensory evaluation detected an increase in mouth feel firmness and elasticity of cooked TC noodles. These results indicate that annealing at room temperature for 3 h followed by the removal of the water-soluble fraction is effective for reducing the cooking losses and improving the textural properties of noodles made from rice flour with high starch damage.
PEP-1-p18 prevents neuronal cell death by inhibiting oxidative stress and Bax expression

Abstract: P18, a member of the INK4 family of cyclin-dependent kinaseinhibitors, is a tumor suppressor protein and plays a key cellsurvival role in a variety of human cancers. Under pathophysiologicalconditions, the INK4 group proteins participate in novelbiological functions associated with neuronal diseases andoxidative stress. Parkinson’s disease (PD) is characterized by loss ofdopaminergic neurons, and oxidative stress is important in itspathogenesis. Therefore, we examined the effects of PEP-1-p18 onoxidative stress-induced SH-SY5Y cells and in a PD mouse model.The transduced PEP-1-p18 markedly inhibited 1-methyl-4-phenylpyridinium-induced SH-SY5Y cell death by inhibiting Baxexpression levels and DNA fragmentation. Additionally, PEP-1-p18prevented dopaminergic neuronal cell death in the substantia nigraof a 1-methyl-4-phenyl-1,2,3,6,-tetrahydropyridine-induced PDmouse model. These results indicate that PEP-1-p18 may be auseful therapeutic agent against various diseases and is a potentialtool for treating PD.
Suppression of 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced skin inflammation in mice by transduced Tat-Annexin protein

Abstract: We examined that the protective effects of ANX1 on 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced skin inflammationin animal models using a Tat-ANX1 protein. Topicalapplication of the Tat-ANX1 protein markedly inhibited TPAinducedear edema and expression levels of cyclooxygenase-2(COX-2) as well as pro-inflammatory cytokines such as interleukin-1 beta (IL-1β), IL-6, and tumor necrosis factor-alpha(TNF-α). Also, application of Tat-ANX1 protein significantlyinhibited nuclear translocation of nuclear factor-kappa B(NF-κB) and phosphorylation of p38 and extracellular signalregulatedkinase (ERK) mitogen-activated protein kinase(MAPK) in TPA-treated mice ears. The results indicate thatTat-ANX1 protein inhibits the inflammatory response byblocking NF-κB and MAPK activation in TPA-induced miceears. Therefore, the Tat-ANX1 protein may be useful as atherapeutic agent against inflammatory skin diseases.
Primordial curvature perturbation during and at the end of multi-field inflation

Abstract: We study the generation of the primordial curvature perturbation in multi-field inflation. Considering both the evolution of the perturbation during inflation and the effects generated at the end of inflation, we present a general formula for the curvature perturbation. We provide the analytic expressions of the power spectrum, spectral tilt and non-Gaussianity for the separable potentials of two inflaton scalars, and apply them to some specific models.
Using Guasha to treat musculoskeletal pain: A systematic review of controlled clinical trials

Abstract: We searched 11 databases (without language restrictions): MEDLINE, Allied and Complementary Medicine (AMED), EMBASE, Cumulative Index to Nursing and Allied Health Literature (CINAHL), Korean Studies Information (KSI), DBPIA, Korea Institute of Science and Technology Information (KISTI), KoreaMed, Research Information Service System (RISS), China National Knowledge Infrastructure (CNKI) and the Cochrane Library. The search strategy was Guasha (OR scraping) AND pain. Risk of bias was assessed with the Cochrane criteria (i.e. sequence generation, blinding, incomplete outcome measures and allocation concealment).Five randomized controlled trials (RCTs) and two controlled clinical trials (CCTs) were included in the present study. Two RCTs compared Guasha with acupuncture in terms of effectiveness, while the other trials compared Guasha with no treatment (1 trial), acupuncture (4 trials), herbal injection (1 trial) and massage or electric current therapy (1 trial). While two RCTs suggested favorable effects of Guasha on pain reduction and response rate, the quality of these RCTs was poor. One CCT reported beneficial effects of Guasha on musculoskeletal pain but had low methodological quality.Current evidence is insufficient to show that Guasha is effective in pain management. Further RCTs are warranted and methodological quality should be improved.Guasha was defined as a therapeutic modality that uses several tools to scrape or rub the surface of the body to relieve blood (Xue) stagnation. Guasha is used for pain relief in Chinese medicine. Tools for Guasha including a Chinese soup spoon, an edge-worn coin, a slice of water-buffalo horn, a cow rib, honed jade and a simple metal cap with a smooth round lip with oil or water are used in Guasha to scrape or rub the skin to relieve blood stagnation at the body surface [1]. Ginseng for Health Care: A Systematic Review of Randomized Controlled Trials in Korean Literature

Abstract: Objective This systematic review was performed to summarise randomised clinical trials (RCTs) assessing the efficacy and safety of ginseng in the Korean literature. Method The study involved systematic searches conducted in eight Korean Medical databases. The methodological quality of all of the included studies was assessed using the Cochrane Risk of Bias tool. We included all RCTs on any type of ginseng compared to placebo, active treatment or no treatment in healthy individuals or patients regardless of conditions. Results In total, 1415 potentially relevant studies were identified, and 30 randomised clinical trials were included. Nine RCTs assessed the effects of ginseng on exercise capacity, cognitive performance, somatic symptoms, quality of life, and sleeping in healthy persons. Six RCTs tested ginseng compared with placebo for erectile dysfunction, while another four studies evaluated the effects of ginseng against no treatment for gastric and colon cancer. Two RCTs compared the effect of red ginseng on diabetes mellitus with no treatment or placebo, and the other nine RCTs assessed the effects of ginseng compared with placebo or no treatment on various conditions. The methodological caveats of the included trials make their contribution to the current clinical evidence of ginseng somewhat limited. However, the 20 newly added trials (66.7% of the 30 trials) may provide useful information for future trials. Ginseng appears to be generally safe, and no serious adverse effects have been reported. Conclusions The clinical effects of ginseng have been tested in a wide range of conditions in Korea. Although the quality of RCTs published in the Korean literature was generally poor, this review is useful for researchers to access studies that were originally published in languages that they would otherwise be unable to read and due to the paucity of evidence on this subject.
