Cysteine Dioxygenase 1 Is a Tumor Suppressor Gene Silenced by Promoter Methylation in Multiple Human Cancers

Abstract: The human cysteine dioxygenase 1 (CDO1) gene is a non-heme structured, iron-containing metalloenzyme involved in the conversion of cysteine to cysteine sulfinate, and plays a key role in taurine biosynthesis. In our search for novel methylated gene promoters, we have analyzed differential RNA expression profiles of colorectal cancer (CRC) cell lines with or without treatment of 5-aza-2′-deoxycytidine. Among the genes identified, the CDO1 promoter was found to be differentially methylated in primary CRC tissues with high frequency compared to normal colon tissues. In addition, a statistically significant difference in the frequency of CDO1 promoter methylation was observed between primary normal and tumor tissues derived from breast, esophagus, lung, bladder and stomach. Downregulation of CDO1 mRNA and protein levels were observed in cancer cell lines and tumors derived from these tissue types. Expression of CDO1 was tightly controlled by promoter methylation, suggesting that promoter methylation and silencing of CDO1 may be a common event in human carcinogenesis. Moreover, forced expression of full-length CDO1 in human cancer cells markedly decreased the tumor cell growth in an in vitro cell culture and/or an in vivo mouse model, whereas knockdown of CDO1 increased cell growth in culture. Our data implicate CDO1 as a novel tumor suppressor gene and a potentially valuable molecular marker for human cancer.
The Relative Expression of Mig6 and EGFR Is Associated with Resistance to EGFR Kinase Inhibitors

Abstract: The sensitivity of only a few tumors to anti-epidermal growth factor receptor EGFR tyrosine kinase inhibitors (TKIs) can be explained by the presence of EGFR tyrosine kinase (TK) domain mutations. In addition, such mutations were rarely found in tumor types other than lung, such as pancreatic and head and neck cancer. In this study we sought to elucidate mechanisms of resistance to EGFR-targeted therapies in tumors that do not harbor TK sensitizing mutations in order to identify markers capable of guiding the decision to incorporate these drugs into chemotherapeutic regimens. Here we show that EGFR activity was markedly decreased during the evolution of resistance to the EGFR tyrosine kinase inhibitor (TKI) erlotinib, with a concomitant increase of mitogen-inducible gene 6 (Mig6), a negative regulator of EGFR through the upregulation of the PI3K-AKT pathway. EGFR activity, which was more accurately predicted by the ratio of Mig6/EGFR, highly correlated with erlotinib sensitivity in panels of cancer cell lines of different tissue origins. Blinded testing and analysis in a prospectively followed cohort of lung cancer patients treated with gefitinib alone demonstrated higher response rates and a marked increased in progression free survival for patients with a low Mig6/EGFR ratio (approximately 100 days, P = 0.01).
Promoter DNA Methylation of Oncostatin M receptor-β as a Novel Diagnostic and Therapeutic Marker in Colon Cancer

Abstract: In addition to genetic changes, the occurrence of epigenetic alterations is associated with accumulation of both genetic and epigenetic events that promote the development and progression of human cancer. Previously, we reported a set of candidate genes that comprise part of the emerging “cancer methylome”. In the present study, we first tested 23 candidate genes for promoter methylation in a small number of primary colon tumor tissues and controls. Based on these results, we then examined the methylation frequency of Oncostatin M receptor-β (OSMR) in a larger number of tissue and stool DNA samples collected from colon cancer patients and controls. We found that OSMR was frequently methylated in primary colon cancer tissues (80%, 80/100), but not in normal tissues (4%, 4/100). Methylation of OSMR was also detected in stool DNA from colorectal cancer patients (38%, 26/69) (cut-off in TaqMan-MSP, 4). Detection of other methylated markers in stool DNA improved sensitivity with little effect on specificity. Promoter methylation mediated silencing of OSMR in cell lines, and CRC cells with low OSMR expression were resistant to growth inhibition by Oncostatin M. Our data provide a biologic rationale for silencing of OSMR in colon cancer progression and highlight a new therapeutic target in this disease. Moreover, detection and quantification of OSMR promoter methylation in fecal DNA is a highly specific diagnostic biomarker for CRC.
Molecular cloning and characterization of a new peroxidase gene (OvRCI) from Orychophragmus violaceus

Abstract: A new peroxidase gene from Orychophragmus violaceus was cloned. The full-length cDNA of O.violaceus peroxidase gene (OvRCI, GenBank. Acc. No. AY428037) was 1220 bp and contained an 1128 bp open reading frame encoding a protein of 375 amino acids. Homology analysis and molecular modeling revealed that OvRCI strongly resembled other peroxidase genes. Quantitative real-time PCR analysis revealed that it was a constitutively salt-inducible gene and its transcript level was most abundant after 24 h treatment with 200 mmol.L-1 sodium chloride. Our studies suggested that OvRCI was a new member of the family of recently cloned peroxidase genes.

Abstract: Carbon nanotubes (CNTs)/mesostructured silica core-shell nanowires with a carbon nanotube core and controllable highly ordered periodic mesoporous silica shell are syntheiszed via the interfacial surfactant template. The core-shell nanowires are characterized by transmission electron microscope (TEM), X-ray diffraction pattern (XRD) and nitrogen sorption/desorption. The results indicate that the core-shell nanowires have highly ordered periodic mesoporous silica shell (space group p6mm), high BET surface area and narrow pore size distribution. Moreover, the morphology of core-shell nanowires can be controlled by the pH value. The core-shell nanowires have promising applications in biosensors, nanoprobes and energy storage due to their good dispersibility in polar solvents.

Abstract: 为了提高单幅线图的三维物体识别重建效率,并保证优化算法的收敛性,基于划整为零、分而治之的策略,首先把单幅线图划分为若干封闭线框,然后按其连接关系进行分组,随后针对各组线框所表征的面片进行三维重建计算,把一个整体优化问题转化为若干局部优化计算问题.在此基础上,分析构造了适用于局部优化识别重建的计算准则,提出了基于旋转变换的分步求解方法.通过子面片绕其父面作旋转变换,利用局部优化准则完成同级子面片的重建计算,根据线图中各线框表征的面片之间的级联关系逐级向下传递,实现全部面片三维重建.算例测试表明,本文方法适于多面体的三维重建,且平均计算效率较全局优化方法高出20%.
: The divide-and-conquer strategy is adopted to improve the efficiency of 3D reconstruction of a single line drawing and guarantee the convergence of the optimization algorithm for polyhedral objects. First, the single line drawing is divided into a lot of closed polygons, then all closed polygons are grouped according to their connection relationships, and finally all faces represented by the closed polygons in one group are reconstructed into 3D. In this way, a normal global optimization problem is resolved in a step-by-step fashion. Based on this strategy, several local optimization criteria are discussed, and a step resolving algorithm for 3D reconstruction by use of the rotation transformation are proposed. By rotating the child face against its parent face until some local optimization criteria are met, all child faces in the same level can be resolved in 3D space. Utilizing the top-down linkage relationships among faces represented by the loops on the drawing, all the child faces will be reconstructed step by step. Case studies indicate that this method is suitable for 3D reconstruction of multi-face objects and the efficiency can be improved by 20% compared with typical global optimization methods

Abstract: Different types of knowledge such as R&D, marketing, and manufacturing knowledge have been focused in the past researches, while few scholars paid attention to strategic management knowledge. This paper focuses on the attainment of strategic objective of MNCs’ Chinese subsidiaries, by analyzing the strategic management knowledge transfer from MNC headquarters to the Chinese subsidiaries. The influence of organizational culture, trust and absorptive capacity on the attainment of strategic objective was examined. 115 Chinese subsidiaries were surveyed in the empirical research, showing different results under the Chinese market background. This paper offers a new perspective for the further researches of knowledge transfer and the strategic management practices.

Abstract:

Most of the tourism products were sold by middlemen travel agencies. There is a very complex relationship between travel agencies and stakeholders. The phenomena including zero fee, debt fee and guides cheating are mess as cluster, which greatly damage the interests of consumers and destroy the image of the industry and market reputation. This thesis aims to analyze the root causes of travel agencies’ interest demands and the game of chaos that is the low threshold set by tourism agencies, the error location in tour guide career, un-standard tourist products, and so on. And then it explores practical solutions and technical methods: 1) taking more specializers and professionals in tour guides; 2) making the unemployment relationship between tourism agencies and tour guides; and 3) setting up the standardization in tourist products, and so on.
