Engineering the Expression and Characterization of Two Novel Laccase Isoenzymes from Coprinus comatus in Pichia pastoris by Fusing an Additional Ten Amino Acids Tag at N-Terminus

Abstract: The detail understanding of physiological/biochemical characteristics of individual laccase isoenzymes in fungi is necessary for fundamental and application purposes, but our knowledge is still limited for most of fungi due to difficult to express laccases heterologously. In this study, two novel laccase genes, named lac3 and lac4, encoding proteins of 547 and 532-amino acids preceded by 28 and 16-residue signal peptides, respectively, were cloned from the edible basidiomycete Coprinus comatus. They showed 70% identity but much lower homology with other fungal laccases at protein level (less than 58%). Two novel laccase isoenzymes were successfully expressed in Pichia pastoris by fusing an additional 10 amino acids (Thr-Pro-Phe-Pro-Pro-Phe-Asn-Thr-Asn-Ser?)tag at N-terminus, and the volumetric activities could be dramatically enhanced from undetectable level to 689 and 1465 IU/l for Lac3 and Lac4, respectively. Both laccases possessed the lowest Km and highest kcat/Km value towards syringaldazine, followed by ABTS, guaiacol and 2,6-dimethylphenol similar as the low redox potential laccases from other microorganisms. Lac3 and Lac4 showed resistant to SDS, and retained 31.86% and 43.08% activity in the presence of 100 mM SDS, respectively. Lac3 exhibited higher decolorization efficiency than Lac4 for eleven out of thirteen different dyes, which may attribute to the relatively higher catalytic efficiency of Lac3 than Lac4 (in terms of kcat/Km) towards syringaldazine and ABTS. The mild synergistic decolorization by two laccases was observed for triphenylmethane dyes but not for anthraquinone and azo dyes.
A Novel Attack Graph Posterior Inference Model Based on Bayesian Network  [PDF]

Abstract: Network attack graphs are originally used to evaluate what the worst security state is when a concerned net-work is under attack. Combined with intrusion evidence such like IDS alerts, attack graphs can be further used to perform security state posterior inference (i.e. inference based on observation experience). In this area, Bayesian network is an ideal mathematic tool, however it can not be directly applied for the following three reasons: 1) in a network attack graph, there may exist directed cycles which are never permitted in a Bayesian network, 2) there may exist temporal partial ordering relations among intrusion evidence that can-not be easily modeled in a Bayesian network, and 3) just one Bayesian network cannot be used to infer both the current and the future security state of a network. In this work, we improve an approximate Bayesian posterior inference algorithm–the likelihood-weighting algorithm to resolve the above obstacles. We give out all the pseudocodes of the algorithm and use several examples to demonstrate its benefit. Based on this, we further propose a network security assessment and enhancement method along with a small network scenario to exemplify its usage.

The Potential Biomarker Panels for Identification of Major Depressive Disorder (MDD) Patients with and without Early Life Stress (ELS) by Metabonomic Analysis

Abstract: Objective The lack of the disease biomarker to support objective laboratory tests still constitutes a bottleneck in the clinical diagnosis and evaluation of major depressive disorder (MDD) and its subtypes. We used metabonomic techniques to screen the diagnostic biomarker panels from the plasma of MDD patients with and without early life stress (ELS) experience. Methods Plasma samples were collected from 25 healthy adults and 46 patients with MDD, including 23 patients with ELS and 23 patients without ELS. Furthermore, gas chromatography/mass spectrometry (GC/MS) coupled with multivariate statistical analysis was used to identify the differences in global plasma metabolites among the 3 groups. Results The distinctive metabolic profiles exist either between healthy subjects and MDD patients or between the MDD patients with ELS experience (ELS/MDD patients) and the MDD patients without it (non-ELS/MDD patients), and some diagnostic panels of feature metabolites' combination have higher predictive potential than the diagnostic panels of differential metabolites. Conclusions These findings in this study have high potential of being used as novel laboratory diagnostic tool for MDD patients and it with ELS or not in clinical application.
Distant hybridization leads to different ploidy fishes

Abstract: Distant hybridization makes it possible to transfer the genome of one species to another, which results in changes in phenotypes and genotypes of the progenies. This study shows that distant hybridization or the combination of this method with gynogenesis or androgenesis lead to different ploidy fishes with genetic variation, including fertile tetraploid hybrids, sterile triploid hybrids, fertile diploid hybrids, fertile diploid gynogenetic fish, and their derived progenies. The formations of the different ploidy fishes depend on the genetic relationship between the parents. In this study, several types of distant hybridization, including red crucian carp (Carassius auratus red var.) (2n=100, abbreviated as RCC) (♀)×common carp (Cyprinus carpio L.) (2n=100, abbreviated as CC) (♂), and RCC (2n=100) (♀)×blunt snout bream (Megalobrama amblycephala) (2n=48, abbreviated as BSB) (♂) are described. In the distant hybridization of RCC (♀)×CC (♂), bisexual fertile F3–F18 allotetraploid hybrids (4n=200, abbreviated as 4nAT) were formed. The diploid hybrid eggs and diploid sperm generated by the females and males of 4nAT developed into diploid gynogenetic hybrids and diploid androgenetic hybrids, respectively, by gynogenesis and androgenesis, without treatment for doubling the chromosome. Improved tetraploid hybrids and improved diploid fishes with genetic variation were derived from the gynogenetic hybrid line. The improved diploid fishes included the high-body RCC and high-body goldfish. The formation of the tetraploid hybrids was related to the occurrence of unreduced gametes generated from the diploid hybrids, which involved in premeiotic endoreduplication, endomitosis, or fusion of germ cells. The sterile triploid hybrids (3n=150) were produced on a large scale by crossing the males of tetraploid hybrids with females of diploid fish (2n=100). In another distant hybridization of RCC (♀)×BSB (♂), different ploidy fishes were obtained, including diploid bisexual fertile natural gynogenetic fish (2n=100), sterile triploid hybrids (3n=124), and bisexual fertile tetraploid hybrids (4n=148). Furthermore, two kinds of pentaploid hybrids (5n=172 and 5n=198) were formed. The biological characteristics and the mechanisms of formation of the different ploidy fish were compared and discussed at the cellular and molecular level. The results indicated distant hybridization or the combination of this method with gynogenesis or androgenesis affects the formation of different ploidy fish with genetic variation.
Comparison of hadron shower data with simulations

Abstract: An analog hadron calorimeter (AHCAL) prototype of 5.3 nuclear interaction lengths thickness has been designed and constructed by members of the CALICE Collaboration. The AHCAL prototype consists of a 38-layer sandwich structure of steel plates and 7608 scintillator tiles that are read out by wavelength-shifting fibres coupled to SiPMs. The signal is amplified and shaped with a custom-designed ASIC. A calibration/monitoring system based on LED light was developed to monitor the SiPM gain and to measure the full SiPM response curve in order to correct for non-linearity. Ultimately, the physics goals are the study of hadronic shower shapes and testing the concept of particle flow. The technical goal consists of measuring the performance and reliability of 7608 SiPMs. The AHCAL prototype was commissioned in test beams at DESY, CERN and FNAL, and recorded hadronic showers, electron showers and muons at different energies and incident angles.

Abstract: The Merrifield-Simmons index () of a graph is defined as the number of subsets of the vertex set, in which any two vertices are nonadjacent, that is, the number of independent vertex sets of The Hosoya index () of a graph is defined as the total number of independent edge subsets, that is, the total number of its matchings. Abstract: In the title complex, [Sn2(C7H7)4(C10H8N2O3)2(C2H5OH)2], the Sn(IV) atom is seven-coordinated in a distorted pentagonal–bipyramidal geometry by three O atoms and one N atom from the pyruvate benzoyl hydrazone ligand, one ethanol O atom and two axial C atoms from trans-benzyl groups, thus forming a dimeric molecule (overline{1} symmetry) via weak Sn—O interactions. The C atoms of one phenyl ring and the ethanol molecule are disordered over two sets of sites with site-occupancy factors of 0.57 (5):0.43 (5) and 0.79 (2):0.21 (2), respectively. Intermolecular O—H...O hydrogen bonds stabilize the crystal structure.
