Cyclosporine A and bromocriptine attenuate cell death mediated by intracellular calcium mobilization

Abstract: To identify the novel inhibitors of endoplasmic reticulumstress-induced cell death, we performed a high throughput assaywith a chemical library containing a total of 3280 bioactive smallmolecules. Cyclosporine A and bromocriptine were identified aspotent inhibitors of thapsigargiin-induced cell death (cut-off at4σ standard score) . However, U74389G, the potent inhibitor oflipid peroxidation had lower activity in inhibiting cell death. Theinhibition effect of cyclosporine A and bromocriptine wasspecific for only thapsigargin-induced cell death. The mechanismof inhibition by these compounds was identified as modificationof the expression of glucose regulated protein-78 (GRP-78/Bip)and inhibition of phosphorylation of p38 mitogen activatedprotein kinase (MAPK). However, these compounds did notinhibit the same events triggered by tunicamycin, which was inagreement with the cell survival data. We suggest that theinduction of protective unfolded protein response by thesecompounds confers resistance to cell death. In summary, weidentified compounds that may provide insights on cell deathmechanisms stimulated by ER stress.
Comparative Study of Various Preparation Methods of Colloidal Silica  [PDF]

Abstract: Colloidal silica can be prepared by various methods and starting materials including ion exchange of aqueous silicates, hydrolysis and condensation of silicon compounds, direct oxidation of silicon, and milling and peptization of silica powder. Various silica sols having particle sizes of 10-60 nm prepared by these methods and the preparation methods have been compared on the basis of their shape, size uniformity, sphericity, stability against pH variation, cation concentration, and price, etc. Silica sol prepared from tetraethoxysilane affords uniform size control and growth, and high purity, despite the relatively high costs. Silica sol prepared from liquid silicates affords relatively easy size and shape control; however, it is difficult to lower the alkali content to a level that is appropriate for carrying out semiconductor chemical mechanical polishing processes; in addition, the waste water treatment carried out for recovering the ion exchange resin gives rise environmental consideration. The properties of colloidal silica prepared from fumed silica powder by milling and dispersion depend on the starting silica source and it is relatively difficult to obtain monodispersed particles using this method. Colloidal silica prepared from silicon by direct oxidation has a monodispersed spherical shape and purity control with reasonable prices. It generates less waste water because it can be directly produced in relatively high concentrations. The cation fraction located in the particle relative to the free cation in the fluid is relatively lower in the silica sol prepared by the direct oxidation than others. A careful comparison of colloidal silica and the preparation methods may help in choosing the proper colloidal silica that is the most appropriate for the application being considered.

Abstract: In this article, we have attempted to analyze current situation and the problem of domestic and overseas harbor container delivery & cargo work automation centered on major harbors and to suggest effective way to deal with the issue in order to improve the productivity of container cargo work per crane, the major index of productivity of high value-added shipbuilding industry. In particular, we have suggested the way to realize effective automation system that can improve the efficiency of harbor container delivery & cargo work through the development of high-tech measuring automation technology using microwave radar and applied design that have broken away from traditional automation system and traditional problems such as dependence on manual work and the problem of laser method in which workers cannot identify laser beam under sunlight and workers’ eyesight can be weakened by being exposed to laser beam.

Abstract: technology transfer (or sharing) is a common way to share external sources. there have been many studies on the relationship of self-r&d and technology transfer regarding its theoretical background and its efficiency. this present research has been developed by examining how a concrete measure of a company or a firm's real performance (increased sales ratio; growth rate) is related to the variables of resources and capabilities using the resource-based theory and open innovation model. the approach of this research is unique in that it examines a sample comprising of licensing-in and technological cooperation variables, categorizes forms according to industry, and looks at such unique variables as a "process" (the ratio of ceo's and related-person's stocks). the data on 361 korean firms was gathered from korea's data analysis, retrieval, and transfer system and worldwide intellectual property search. findings show that human, technology, and fixed assets are related positively to financial performance, and searching, absorbing, and openness capabilities as a control effect is related positively to a firm's increased sales ratio. strategic plans for technology transfer companies are also included in this research.
Prognostic value of expression of molecular markers in adenoid cystic cancer of the salivary glands compared with lymph node metastasis: a retrospective study

Abstract: Background Adenoid cystic cancer arising in the salivary glands has distinctive features such as perineural invasion, distant metastasis, and a variable prognosis. In salivary gland cancer, c-kit, EGFR, and VEGF are representative molecular markers that may predict remnant and recurrent tumors. In this study, the expression of c-kit, EGFR, and VEGF in adenoid cystic cancer was evaluated, and the relationships between the expression of these markers and the clinical findings were investigated. Methods The medical records of 48 patients who were treated for parotid adenoid cystic cancer from January 1990 to January 2006 were reviewed. The tumor location, size, histological subtypes, perineural invasion, the resected margin status, and lymph node metastasis were assessed. Immunohistochemical staining and semiquantitative analysis of c-kit, EGFR and VEGF were performed. The relationship between the expression of each marker and the clinicopathological factors were analyzed. Results Positive c-kit immunostaining was present in 45 patients (94%), with weak positivity (+1) in 23, moderate positivity (+2) in 19 and strong positivity (+3) in three. Positive EGFR immunostaining was observed in 27 (56%), with weak positivity (+1) in 19 and moderate positivity (+2) in eight with no strong positive staining. Positive VEGF immunostaining was present in 42 patients (88%) with weak positivity (+1) in 12, moderate positivity (+2) in 17, and strong positivity (+3) in 13. Only the expression of VEGF was significantly higher in parotid gland tumors than in any other gland (P = 0.032). Marginal involvement was associated with strong VEGF expression (P = 0.02). No marker was significantly correlated with recurrence or the survival rate. Lymph node status was related to the survival rate. Conclusions The expression of c-kit, EGRF, and VEGF had no predictive value for recurrence or the prognosis of adenoid cystic cancer. Only the lymph node status was related to the prognosis.
Cellulosic ethanol production using a yeast consortium displaying a minicellulosome and β-glucosidase

Abstract: Background Cellulosic biomass is considered as a promising alternative to fossil fuels, but its recalcitrant nature and high cost of cellulase are the major obstacles to utilize this material. Consolidated bioprocessing (CBP), combining cellulase production, saccharification, and fermentation into one step, has been proposed as the most efficient way to reduce the production cost of cellulosic bioethanol. In this study, we developed a cellulolytic yeast consortium for CBP, based on the surface display of cellulosome structure, mimicking the cellulolytic bacterium, Clostridium thermocellum. Results We designed a cellulolytic yeast consortium composed of four different yeast strains capable of either displaying a scaffoldin (mini CipA) containing three cohesin domains derived from C. thermocellum, or secreting one of the three types of cellulases, C. thermocellum CelA (endoglucanase) containing its own dockerin, Trichoderma reesei CBHII (exoglucanase) fused with an exogenous dockerin from C. thermocellum, or Aspergillus aculeatus BGLI (β-glucosidase). The secreted dockerin-containing enzymes, CelA and CBHI, were randomly assembled to the surface-displayed mini CipA via cohesin-dockerin interactions. On the other hand, BGLI was independently assembled to the cell surface since we newly found that it already has a cell adhesion characteristic. We optimized the cellulosome activity and ethanol production by controlling the combination ratio among the four yeast strains. A mixture of cells with the optimized mini CipA:CelA:CBHII:BGLI ratio of 2:3:3:0.53 produced 1.80 g/l ethanol after 94 h, indicating about 20% increase compared with a consortium composed of an equal amount of each cell type (1.48 g/l). Conclusions We produced cellulosic ethanol using a cellulolytic yeast consortium, which is composed of cells displaying mini cellulosomes generated via random assembly of CelA and CBHII to a mini CipA, and cells displaying BGLI independently. One of the advantages of this system is that ethanol production can be easily optimized by simply changing the combination ratio of the different populations. In addition, there is no limitation on the number of enzymes to be incorporated into this cellulosome structure. Not only cellulases used in this study, but also any other enzymes, including cellulases and hemicellulases, could be applied just by fusing dockerin domains to the enzymes.
Development of an Electrogenerated Chemiluminescence Biosensor using Carboxylic acid-functionalized MWCNT and Au Nanoparticles

Abstract: A COOH-F-MWCNT-Nafion-Ru(bpy)32+-Au-ADH electrogenerated chemi-luminescence (ECL)electrode using COOH-functionalized MWCNT (COOH-F-MWCNT) and Au nanoparticles synthesized by the radiation method was fabricated for ethanol sensing. A higher sensing efficiency for ethanol for the ECL biosensor prepared by PAAc-g-MWCNT was measured compared to that of the ECL biosensor prepared by PMAc-g-MWCNT, and purified MWCNT. Experimental parameters affecting ethanol detection were also examined in terms of pH and the content of PAAc-g-MWCNT in Nafion. Little interference of other compounds was observed for the assay of ethanol. Results suggest this ECL biosensor could be applied for ethanol detection in real samples.
Multidirectional instability of the shoulder – current concept

Abstract: The multidirectional instability of the shoulder is a complex problem in terms of diagnosis and treatment. Distinct from multidirectional hyperlaxity, multidirectional instability has symptoms related with increased translations in more than one direction. Increased translation by the increased capsular ligamentous laxity is underlying pathology of the posterior and multidirectional instability. This increased capsular laxity can be in-borne or developmental and asymptomatic or minimally symptomatic initially. In this stage, attempted translation does not produce symptoms. Jerk and Kim tests reveal posterior clunk without shoulder pain [1,2].However, repetitive subluxation overloads the posteroinferior glenoid labrum by the excessive rim-loading of the humeral head. This excessive rim-loading eventually develops posteroinferior labral lesion varying from simple retroversion to incomplete detachment (Figure 1). In this stage, patient's symptom which is shoulder pain, originates from the labral lesion when the humeral head glides over the pathologic labrum. The compressive force on the torn labrum in the jerk and Kim tests generates shoulder pain. Therefore, intact labrum does not produce shoulder pain no matter how lax the glenohumeral joint is. Increased translation alone produces asymptomatic posterior clunk until the repetitive rim-loading eventually develops posteroinferior labral lesion [3].Four type of the labral lesion have been reported. The Kim lesion is a concealed and incomplete tear of the posteroinferior labrum which is characterized by loss of labral height and retroversion, marginal crack, and loose inside. The lesion is similar to the intratendinous tear of the rotator cuff in that it is not evident in the initial observation. Dormancy Breaking and Effects on Tuber Yield of Potato Subjected to Various Chemicals and Growth Regulators under Greenhouse Conditions

Abstract: This study was conducted to check the effect of various chemicals and growth regulators was checked on dormancy breaking of potato tubers, and also to check the subsequent effects on tuber yield under greenhouse conditions. The chemicals were tested on three potato cultivars having different dormancy release characters. Among the chemicals the tubers treated with rindite exhibited the highest sprouting ratio in all the cultivars. Cultivar Diamont took the least period of time for 50% of tubers to sprout followed by cultivar Atlantic. Highest decay ratio of tubers was observed for cultivar Atlantic and Diamont treated with thio-urea. The tubers treated with thio-urea induced the highest number of sprouts as compare to all other treatments. Significant differences were observed among the treatments and cultivars for various yield characters
