Abstract: this paper analyses the inflections of the high school policies in brazil, from 2007 onwards. it questions the context in which they occurred and the contradictions present in the process. it searches to identify the main changes, the discourses that supported them and the measures and governmental programs that followed them. it depicts and analyses these changes and their implications in the states, focusing on minas gerais. based on the gathered arguments, it stresses a few considerations and tendencies in the path traced by the current policies concerning the finalities and the universalization of this stage of the country's basic education.

Abstract: This article compares diffusion models used to describe seedless grape drying at low temperature. The models were analyzed, assuming the following characteristics of the drying process: boundary conditions of the first and the third kind; constant and variable volume, V; constant and variable effective mass diffusivity, D; constant convective mass transfer coefficient, h. Solutions of the diffusion equation (analytical and numerical) were used to determine D and h for experimental data of seedless grape drying. Comparison of simulations of drying kinetics indicates that the best model should consider: 1) shrinkage; 2) convective boundary condition; 3) variable effective mass diffusivity. For the analyzed experimental dataset, the best function to represent the effective mass diffusivity is a hyperbolic cosine. In this case, the statistical indicators of the simulation can be considered excellent (the determination coefficient is R2 = 0.9999 and the chi-square is χ2 = 3.241 × 10–4).

Abstract: The identification of arthropod bloodmeals is important in many epidemiological studies, as, the understanding of the life cycle of vectors and the patogens they transmit, as well as helping to define arthropods' control strategies. The precipitin test has been used for decades, but ELISA is slowly becoming more popular. To compare the two tests for sensitivity, specificity and accuracy to detect small insect bloodmeals, Aedes aegypti or Ae. fluviatilis mosquitoes were fed either on feline, canine or human hosts. Mosquitoes were frozen at 6, 12, 24, 48 or 72 h after feeding. Precipitin test showed better specificity and accuracy and ELISA test showed higher sensitivity. Better results with both tests were achieved when mosquitoes were frozen within 48 h from feeding.

Abstract: A citometria de fluxo é considerada a técnica de sexagem de sêmen mais promissora, em virtude de a acui-dade de separa o estar em torno de 90%. Dessa forma, o presente estudo objetivou determinar o efeito da IA com sêmen de touros Nelores sexados por citometria de fluxo sobre a taxa de prenhez, bem como sua acurácia. Foram inseminadas 427 novilhas da ra a Nelore utilizando uma única dose de sêmen por animal, sendo que em 241 novi-lhas utilizou-se o sêmen sexado por citometria de fluxo com concentra es de 3 x 106 espermatozóides portado-res do cromossomo X/dose e em 186 utilizou-se o sêmen n o sexado com concentra es mínimas de 10 x 106 es-permatozóides viáveis/dose. As insemina es ocorreram depois da detec o de cio natural ou após tratamento para sincroniza o. Os resultados dos índices de prenhez n o indicaram diferen as entre o sêmen sexado e o n o-sexado e a acurácia na determina o do sexo apresentou níveis elevados de confiabilidade (92,4%). The flow-sorting sperm technology is considered as the most promising technique for sexing semen, due to the separation accuracy of around 90%. Thus, the objec-tive of this study was to determinate the effect of Articial Insemination (AI) with semen of Nelore bulls sexed by flow cytometry on the pregnancy rate and its accuracy. Throughout the study, 427 Nelore heifers were inseminated with only one dose of semen as follows: 241 heifers were treated with flow-sexed sperm (3 x 106 total flow-sorting X-chromosome bearing sperm/dose) and 181 were treated with unsexed sperm (10 x 106 total sperm/dose). The heifers were inseminated after detection of natural heat or after syn-chronization treatment. The results obtained for pregnancy rates did not indicate differences between flow-sexed and unsexed sperm and the accuracy in the sex determination presented high levels and reliability (92.4%). Abstract: by taking into account the nature, configurations, and meanings of the teaching work as its analysis object, this article discusses the methodology being developed in a research project. the text addresses the methodological-theoretical choices made, the field work done with teachers and the preliminary results achieved. the conclusions confirm the researchers' initial expectation concerning the ability expressed by teachers of getting involved in the analysis of their own work, the confidence in the collective work and the possibility of changes by means of their organization.

Abstract: despite representing a major advance in rheumatology practice, the use of tumor necrosis factor blockade (anti-tnfs) in the treatment of rheumatoid arthritis (ra) has given rise to a problem that was considered solved in many developed countries, i.e. the heightened risk of reactivation of latent tuberculosis infection (ltbi). the identification of cases of ltbi has thus been made obligatory prior to starting any anti-tnf treatment. the cutaneous tuberculin test (ppd) is not the ideal screening test for this group of patients. low specificity, cross-reactivity with vaccine antigens and other exogenous microorganisms coupled to a defect in the cell-mediated component of the immunological response account for the inadequacy of ppd as a screening tool in this subset of patients. assays using the detection of in vitro ifng production by peripheral blood mononuclear cells stimulated by specific antigens (esat-6 and cfp-10), which are found neither in the bcg vaccine nor in other micro-organisms in the environment should perform better than the ppd, owing to a presumed higher specificity as well as to correlation with indirect measures of exposure to mycobacterium tuberculosis besides decreased cross-reactivity determined by prior bcg vaccination and/or other infections.

Abstract: the authors report a case of a 29 year old woman who has seropositive rheumatoid arthritis for six months, and developed severe granulocytopenia and important splenomegaly, however she didn′t show any joint inflammation. she did not respond either to pulse or oral steroids, or to oral methotrexate and leflunomide. she also developed an adverse reaction to the use of infliximab and did not respond well to adalimumab. although she has had repeated infections, despite the various forms of antibiotics and long-term use of g-csf, with high titers of rheumatoid factor, and high levels of esr and crp, the classic rituximab method for treating rheumatoid arthritis was used. there was a good clinical response with an increase in the number of neutrophils following normalization of them, together with the reduction of rheumatoid factor titers, esr and crp. at the moment, the patient is in remission, according to both clinical and laboratory criteria and taking 5mg of prednisone per day and 10mg of methotrexate per week.

Assessment of TP53 Mutations in Benign and Malignant Salivary Gland Neoplasms

Abstract: Despite advances in the understanding of the pathogenesis of salivary gland neoplasms (SGN), the molecular pathways associated with enhanced tumor growth and cell survival remain to be established. The aim of the present study was to investigate whether TP53 mutations are relevant to SGN pathogenesis and if they impact on p53 protein expression. The study included 18 benign and 18 malignant SGN samples. Two polymorphic microsatellite markers at the TP53 genetic locus were chosen to assess loss of heterozygosity (LOH) in the samples that had matched normal DNA. The TP53 exons 2–11 were amplified by PCR, and all of the products were sequenced. Reverse transcription-PCR of the TP53 open reading frame (ORF) was carried out in the samples that had fresh tissue available, and immunohistochemistry for the p53 protein was performed in all samples. TP53 LOH was only found in two pleomorphic adenomas. We found two missense mutations in exon 7 (one in a pleomorphic adenoma and the other in a polymorphous low grade adenocarcinoma), another in exon 8 (in a carcinoma ex pleomorphic adenoma) and a fourth missense mutation in exon 10 (in a mucoepidermoid carcinoma). In addition, a nonsense mutation was found in exon 8 of an adenoid cystic carcinoma. Several intronic and exonic SNPs were detected. Although almost all of the malignant samples were immunopositive for p53, approximately 37% of the benign samples were positive, including the sample harboring the missense mutation and one of the samples that showed LOH. The complete TP53 ORF could be amplified in all samples analyzed, including the IHC negative samples, the samples showing LOH and one sample displaying a missense mutation. In summary, our results show that TP53 mutations are not a frequent event in SGN and that p53 immunopositivity might not be associated with sequence mutations in SGN.
