Targeting Chronic Pain with Epigenetic Drugs: Focus on mGlu2 Receptors

Abstract: Histone deacetylase (HDAC) enzymes regulate gene expression by affecting chromatin structure and/or the activity of transcription factors. We have recently demonstrated that histone deacetylase inhibitors (HDACIs) behave as epigenetic agents capable of inducing analgesia by up-regulating metabotropic glutamate type 2 (mGlu2) receptors. Specifically, the regulation of mGlu2 receptor expression appears to involve the acetylation of the NF-κB transcription factor. mGlu2 and mGlu3 receptors belong to class II metabotropic glutamate receptors. These receptors are coupled to Gi/o proteins and play an important role in mediating antinociception in a variety of inflammatory and chronic pain models. We have shown that the HDACI-mediated mGlu2 receptor up-regulation occurs in the dorsal horn of the spinal cord and in the dorsal root ganglia, supporting a predominant role for mGlu2 receptors as mediators of analgesia in experimental animal models of chronic pain. We suggest that drugs that increase the expression of mGlu2 receptors, such as HDACIs or acetylating drugs (e.g. L-acetylcarnitine), may be effective in patients with chronic pain that are refractory to conventional analgesics.
mGlu2 metabotropic glutamate receptors restrain inflammatory pain and mediate the analgesic activity of dual mGlu2/mGlu3 receptor agonists

Abstract: In this study we used mGlu2 or mGlu3 knock-out mice to dissect the specific role for these two receptors in the endogenous control of inflammatory pain and their specific contribution to the analgesic activity of mixed mGlu2/3 receptor agonists.Our results showed that mGlu2-/- mice display a significantly greater pain response compared to their wild type littermates. Interestingly the increased pain sensitivity in mGlu2-/- mice occurred only in the second phase of the formalin test. No differences were observed in the first phase. In contrast, mGlu3-/- mice did not significantly differ from their wild type littermates in either phase of the formalin test.When systemically injected, a single administration of the mGlu2/3 agonist, LY379268 (3 mg/kg, ip), showed a significant reduction of both phases in wild-type mice and in mGlu3-/- but not in mGlu2-/- mice. However tolerance to the analgesic effect of LY379268 (3 mg/kg, ip) in mGlu3-/- mice developed following 5 consecutive days of injection.Taken together, these results demonstrate that: (i) mGlu2 receptors play a predominant role over mGlu3 receptors in the control of inflammatory pain in mice; (ii) the analgesic activity of mixed mGlu2/3 agonists is entirely mediated by the activation of the mGlu2 subtype and (iii) the development of tolerance to the analgesic effect of mGlu2/3 agonists develops despite the lack of mGlu3 receptors.Metabotropic glutamate (mGlu) receptors are considered promising targets in the treatment of chronic pain. Abstract: Activation of group II metabotropic glutamate receptors (mGlu2 and mGlu3) induces antinociception in several pain models in rodents [1-5]. Consistent with these studies, we have shown that L-acetylcarnitine (LAC), a drug clinically effective in the treatment of neuropathic pain of various origins [6-9], up-regulates the expression of mGlu2 in the dorsal root ganglia (DRG) and in the dorsal horn (DH) of the spinal cord [10,11]. This has challenged the previous view that LAC increases pain thresholds and relieves neuropathic pain by enhancing brain acetylcholine synthesis [12] or by increasing the trophism of peripheral nerves [13,14]. Consistent with the "mGlu2 hypothesis of LAC-induced analgesia," the mGlu2/3 receptor antagonist, LY341495, prevents LAC-induced analgesia in rodents [10]. Interestingly, LAC selectively enhances the expression of mGlu2 receptors and has no effect on the expression of mGlu3 receptors [10,15], although these two receptor subtypes are highly homologous and share similar functions in the CNS [16]. This suggests that the expression of mGlu2 and mGlu3 is differentially regulated and that unraveling the nature of this difference may lead to the identification of new targets for the treatment of neuropathic pain.Analysis of the 5'-region upstream of the coding sequence of the human GRM2 gene (encoding mGlu2) [GenBank: AB045011], using the Transcription Factor Binding Sites Database TRANSFACT and TFSEARCH, revealed the presence of many potential regulatory elements for transcription factors of the NF-κB family, including p50 and p65/Rel-A, and for the coactivator p300. In contrast, only one binding site for the NF-κB family protein, c-Rel, and no binding sites for p65/RelA and p300 have been described in the putative promoter region of the human GRM3 gene encoding mGlu3 [17]. Hyperalgesic activity of kisspeptin in mice

Abstract: Immunofluorescent staining in the mouse skin showed the presence of GPR54 receptors in PGP9.5-positive sensory fibers. Intraplantar injection of kisspeptin (1 or 3 nmol/5 μl) induced a small nocifensive response in naive mice, and lowered thermal pain threshold in the hot plate test. Both intraplantar and intrathecal (0.5 or 1 nmol/3 μl) injection of kisspeptin caused hyperalgesia in the first and second phases of the formalin test, whereas the GPR54 antagonist, p234 (0.1 or 1 nmol), caused a robust analgesia. Intraplantar injection of kisspeptin combined with formalin enhanced TRPV1 phosphorylation at Ser800 at the injection site, and increased ERK1/2 phosphorylation in the ipsilateral dorsal horn as compared to naive mice and mice treated with formalin alone.These data demonstrate for the first time that kisspeptin regulates pain sensitivity in rodents and suggest that peripheral GPR54 receptors could be targeted by novel drugs in the treatment of inflammatory pain.Kisspeptin is a 54-amino acid peptide originally discovered for its activity as metastasis-suppressor [1]. It is encoded by the Kiss1 gene as a 145-amino acid precursor protein and cleaved to a 54-amino acid protein as well as into shorter products (kisspeptin-10,-13,-14) known to play a critical role in the neuroendocrine regulation of reproduction [2-5].In the brain, kisspeptin is localized not only in areas involved in gonadotropin secretion, but also in other regions such as the amygdala, hippocampus, and the periacqueductal gray [6,7].Its action is mediated by a 7-TM receptor named GPR54, also known as KISS1R, which is coupled to polyphosphoinositide hydrolysis via a Gq/11 GTP binding protein [2,8].Loss-of-function mutations of GPR54 cause a non-Kallman variant of hypogonadotropic/hypogonadism in humans (i.e. hypogonadotropic/hypogonadism without anosmia) [2,9]. Abstract: the preparation of g-lialo2 by coprecipitation and sol-gel synthesis was investigated. ceramic powders obtained by coprecipitation synthesis were prepared from aqueous solutions of aluminum and lithium nitrates using sodium hydroxide as precipitant agent. by sol-gel synthesis, the ceramic powders were prepared from hydrolysis of aluminum isopropoxide. the materials obtained by two routes of synthesis were dried at 80oc and calcined at 550, 750, 950 and 1150oc. the characterization was done by x-ray diffraction, infrared spectroscopy, emission and absorption atomic spectrometry, helium picnometry, specific surface area (bet method) and scanning electronic microscopy. mixtures of crystalline phases were obtained by coprecipitation synthesis: 80oc- lial2(oh)7.2h2o + al(oh)3; 550 and 750oc- a-lialo2 + h-al2o3; 950 and 1150oc- g-lialo2 + lial5o8. chemical analysis showed molar ration al/li @ 3. crystalline single-phases were obtained by sol-gel synthesis above 550oc: 550oc-a-lialo2; 750, 950 and 1150oc-g-lialo2. these powders presented molar ration al/li @ 1. thus, g-lialo2 crystalline phase was obtained at 750oc by sol-gel synthesis while by coprecipitation synthesis, a mixture of crystalline phases was obtained. these results showed the superiority of the sol-gel synthesis for the preparation of pure g-lialo2.

Thus, gamma-LiAlO2 crystalline phase was obtained at 750oC by sol-gel synthesis while by coprecipitation synthesis, a mixture of crystalline phases was obtained. These results showed the superiority of the sol-gel synthesis for the preparation of pure gamma-LiAlO2.
BeppoSAX and XMM-Newton spectral study of 4U 1735-44

Abstract: Low-mass X-ray binary systems consist of a neutron star and a main-sequence companion star. The compact object accretes matter via Roche-lobe overflow, which leads to an accretion disk. In addition to a broad-band continuum emission of a thermal component and a Comptonization part, evidence for a broad iron K{\alpha} line is found in several sources. Some of them show an asymmetric line profile as well, which could originate from relativistic effects. To understand the spectral behavior of the system 4U 1735-44, we study the broad-band spectrum and especially the iron line feature between 6.4 and 6.97 keV. The shape of the line allows one to determine the region where the line is produced. Together with the continuum models, a geometrical model of the source can be proposed. Furthermore, the effects of pile-up in the XMM-Newton observation are investigated. We analyzed data obtained with the X-ray satellites BeppoSAX and XMM-Newton. The XMM-Newton data were analyzed, specifically taking into account pile-up effects. With the help of the data of these two satellites, we performed a detailed spectral study in an energy range from 0.2-24 keV. During the observations, the source was in the so-called banana state of an atoll source. Fitting the BeppoSAX data, we found line features that we were able to model with a reflection model, whereas the continuum was modeled with a combination of a thermal component and a Comptonization part. The analysis of the XMM-Newton data gave evidence for a broad but not asymmetric iron line. We found no broadening or asymmetry of the line because of pile-up.
Ego strength and health: An empiric study in hemodialysis patients  [PDF]

Abstract: Introduction: Ego strength represents an important variable that could be predictable about health and compliance in chronic diseases. In this study we propose a new questionnaire, E.F.E. “Ego Functioning Experience”, able to reveal the psychological functioning profile in Hemodialysis patients. The aim of this work, is to underline the existing relations between emotional profile and compliance, whit E.F.E., that may be predictive of a state of psychologycal health in hemodialysis patients. Methods: Study population included 90 hemodialysis patients. The study protocol was made by three psychological tests: the E.F.E. Questionnaire, the DMI, the Self-Liking and Self-Competence Scale. Results: Factor analysis extracts three factors: 1) “Need of Ego support”; 2) “Ego activity oriented to treatment”; 3) “Ego strength”. The correlation coefficients between the E.F.E. Questionnaire factors (three factors) and other measures, showed a direct correlation with the first factor and “turning against object” TAO of the DMI test (correlation is significant at the 0.001 level) and an inverse relation with set mechanisms of “Principalization”, PRN of the DMI (correlation is significant at the 0.005 level). The correlations of the second and the third factors with other measures, were not statistically significant. No relations were found about other variables. Conclusions: The E.F.E. questionnaire is a simple evaluation to detect hemo-dialysis patients who may need greater attention to the psychological health and therefore the need for treatment such as psychological support. In fact, lower presence of ego strength is indicative of poor compliance to clinical treatment in hemodialysis, but also of worsening of psychiatric symptoms such as demoralization and depressed mood. In conclusion, an increased social support is needed in hemodialysis patient in order to achieve better compliance and achieve a better state of psychological health in chronic hemodialysis patients.

Abstract: The genus Solenopsis includes more than 100 species in the New World. When introduced, some species cause serious harm to the native wildlife and agriculture. Two forms of social organization are known in the genus, namely, monogyne and polygyne. Here, we present the genetic profile of a Solenopsis invicta Buren 1972 colony recorded in the central region of the city of São Paulo, São Paulo state, Brazil (23°;33'37.18\"S; 46°;42'48.66\"W) to describe allele diversity. Eight microsatellite loci were used for the analysis of 30 worker ants. The results show the presence of seven alleles genotyped at the same microsatellite locus. Because this is an unusual finding, all of the alleles were sequenced, and surprisingly, an eighth allele was identified. These data suggest that the intracolonial genetic profile of fire ants must be investigated more frequently because there may be unexpected, albeit unknown, genetic patterns for S. invicta that can help inform better control and management programs.

Abstract: this work constitutes the first survey of fruitful species of rond?nia state , in the brazilian amazon. this started in 2005, until now 22 species had been catalogued, distributed in 13 botanical families. among the families palmae and sterculiaceae have been distinguished, each one with five described species. some species are not part of the popular knowledge, being consumed mainly for the wild fauna, as diospyros brasiliensis mart. (caqui-do-mato), bellucia grossularioides (l.) triana (jambinho) and maclura tinctoria (l.) engl. (amoreira). beyond the nourishing use, some species present new potentials, case of a?aí euterpe oleracea mart., whose seeds are used in the manufacture of ?bio-jewels?.
