Synthesis and Characterization of New Chiral Monoanionic [ON] Ancillary Phenolate Ligands  [PDF]

Abstract: Three new chiral monoanionic [ON] ancillary phenolate ligands with varying pendant arms have been synthesized in moderate to high yields (50% - 85%) via Mannich-type condensation reaction of chiral substituted phenol, formaldehyde and (+)-bis-[(R)-1-phenylethyl]amine. These new organic compounds were fully characterized via nuclear magnetic resonance spectroscopy (1H and 13C) and elemental analysis. The newly synthesized ligands are suitable candidates for metal-catalyzed ring-opening of lactones and asymmetric catalysis.
External quality assessment of cytomegalovirus DNA detection on dried blood spots

Abstract: The 27 responding laboratories from 14 countries submitted 33 datasets obtained by means of conventional PCR (n = 5) or real-time PCR (n = 28) technologies. A correct positive result was reported in at least 91% of datasets in samples with a viral load of 8.8 × 104 copies/ml or higher. However only 59% and 12% identified the 9.4 × 103 and 7.3 × 102 copies/ml samples, respectively, correctly as positive. False positive results were reported by 9% of laboratories and in 11% of datasets.These results indicate a clear need for improvement of methods as sensitivity and false-positivity still appear to be a major problem in a considerable number of laboratories.Congenital CMV infection is the most widespread congenital infection in humans and is a major cause of neurological damages such as hearing loss, visual impairment and mental retardation in children. Diagnosis of congenital CMV infection requires laboratory testing done on samples collected in the first three weeks of life. Testing for CMV-DNA in neonatal blood collected on filter-paper (dried blood spot, DBS) has proved a valid means of diagnosis with both clinical and epidemiological relevance. Applications of this assay range from diagnosis in the neonatal period as an alternative to the conventional urine culture method to the unique quality of ascertaining whether damages arising during infancy are due to congenital infection [1,2].Most laboratories use an in-house developed assay for detection of CMV-DNA in DBS. However, no international standard is available and previous external quality assessment studies have shown that the quality of nucleic acid amplification methods such as PCR varies considerably between laboratories [3,4]. Abstract: In the present study, we elaborated the results obtained on blood samples collected in the first two weeks of life from 22 symptomatic and 48 asymptomatic newborns with cCMV diagnosed through urine testing. We evaluated the performances of two quantitative methods (pp65 antigenemia test and plasma Real-time PCR) and the semi-quantitative results of dried blood sample (DBS) test in the aim of identifying a valid method for measuring viral load.Plasma qPCR and DBS tests were positive in 100% of cases, antigenemia in 81%. Only the latter test gave quantitatively different results in symptomatic versus asymptomatic children. qPCR values of 103 copies/ml were found in 52% of newborn. "Strong" DBS test positivity cases had higher median values of both pp65 positive PBL and DNA copies/ml than cases with a "weak" positivity.As expected antigenemia test was less sensitive than molecular tests and DBS test performed better on samples with higher rates of pp65 positive PBL and higher numbers of DNA copies/ml. The prognostic significance of the results of these tests will be evaluated on completion of the ongoing collection of follow-up data of these children.Congenital cytomegalovirus infection (cCMV) is present in 0.7% of neonates [1] and represents one of the most feared events during pregnancy, causing not only immediate but also delayed sequelae to the infant. Abstract: In the contemporary postcolonial discourse, multiculturalism is an oft used term which has been variously defined by different scholars. Analyzing the different facets of multiculturalism, the paper attempts to associate it with diasporic writings. For the purpose I have chosen the works of Jhumpa Lahiri and Margaret Wilson; Lahiri is a post colonial American writer of Indian origin while Wilson is a colonial American writer who had lived in India and also written on India. The thrust of the paper is to contrast their multicultural concerns considering their respective social ambience.
Zero cycles with modulus and zero cycles on singular varieties

Abstract: Given a smooth variety $X$ and an effective Cartier divisor $D \subset X$, we show that the cohomological Chow group of 0-cycles on the double of $X$ along $D$ has (under some conditions) a canonical decomposition in terms of the Chow group of 0-cycles ${\rm CH}_0(X)$ and the Chow group of 0-cycles with modulus ${\rm CH}_0(X|D)$ on $X$. When $X$ is projective, we construct an Albanese variety with modulus and show that this is the universal regular quotient of ${\rm CH}_0(X|D)$. As a consequence of the above decomposition, we prove the Roitman torsion theorem for the 0-cycles with modulus. We show that, if $X$ is affine, ${\rm CH}_0(X|D)$ is torsion-free and that there is an injective cycle class map ${\rm CH}_0(X|D) \hookrightarrow K_0(X,D)$, where $K_0(X,D)$ denotes the relative $K_0$-group for the inclusion $D\subset X$. For a smooth affine surface $X$, this is strengthened to show that $K_0(X,D)$ is an extension of ${\rm CH}_1(X|D)$ by ${\rm CH}_0(X|D)$. We derive from this Bloch's formula for 0-cycles with modulus on surfaces.
Relative cycles with moduli and regulator maps

Abstract: Let X be a separated scheme of finite type over a field k and D a non-reduced effective Cartier divisor on it. We attach to the pair (X, D) a cycle complex with modulus, whose homotopy groups - called higher Chow groups with modulus - generalize additive higher Chow groups of Bloch-Esnault, R\"ulling, Park and Krishna-Levine, and that sheafified on $X_{Zar}$ gives a candidate definition for a relative motivic complex of the pair, that we compute in weight 1. When X is smooth over k and D is such that $D_{red}$ is a normal crossing divisor, we construct a fundamental class in the cohomology of relative differentials for a cycle satisfying the modulus condition, refining El-Zein's explicit construction. This is used to define a natural regulator map from the relative motivic complex of (X,D) to the relative de Rham complex. When X is defined over $\mathbb{C}$, the same method leads to the construction of a regulator map to a relative version of Deligne cohomology, generalizing Bloch's regulator from higher Chow groups. Finally, when X is moreover connected and proper over $\mathbb{C}$, we use relative Deligne cohomology to define relative intermediate Jacobians with modulus $J^r_{X|D}$ of the pair (X,D). For r= dim X, we show that $J^r_{X|D}$ is the universal regular quotient of the Chow group of 0-cycles with modulus.
Types of Human Stem Cells and Their Therapeutic Applications  [PDF]

The present review examines in the first place various kinds of naturally occurring stem cells, including germ cells and embryonic stem cells (ES cells), as well as haemopoietic stem cells, which are historically the first to be used for medical treatment. Attention is also given to cancer stem cells, as a source of perseverant malignant disease. The main interest is now represented by the variety of somatic cells, which can be re-programmed to different types of differentiated cells, the so-called induced pluripotent stem cells (IPSC’s). Focus is now directed not only to the factors which make such events possible like de-differentiation and reconversion but also to the stages involved in this process. It is actually postulated that the transition from differentiated cells to pluripotent cells follows a definite sequence with evidence of two waves of gene regulations. Main applications of stem cell therapy are reviewed, from the established use of haemopoietic stem cells for clinical transplantation in a variety of haematological disorders to the initial attempts to employ stem cells for the treatment of other disparate conditions. Problems related to stem cell treatment with both ES and IPS cells, like the necessity of a large in vitro expansion to provide sufficient amounts of cells and the related risk of genomic abnormalities are illustrated. The necessity of safe procedures for the development of this venture is also outlined.
