Regulatory effects of the JAK3/STAT1 pathway on the release of secreted phospholipase A2-IIA in microvascular endothelial cells of the injured brain

Abstract: Supernatants were collected from primary cultures of BMVECs. The release of sPLA2-IIA, and the expression of inducible nitric oxide synthase (iNOS), phospho-JAK3, phospho-STAT1, total JAK3 and STAT1, β-actin, and bovine serum albumin (BSA) were analyzed by Western blot or ELISA. NO production was calculated by the Griess reaction. sPLA2 enzyme activity was measured with a fluorometric assay. Specific inhibitors of NO (L-NAME and aminoguanidine, AG), JAK3 (WHI-P154,WHI), STAT1 (fludarabine, Flu), and STAT1 siRNA were used to determine the involvement of these molecules in the LPS-induced release of sPLA2-IIA from BMVECs. Nuclear STAT1 activation was tested with the EMSA method. The monolayer permeability of BMVECs was measured with a diffusion assay using biotinylated BSA.Treatment of BMVECs with LPS increased the release of sPLA2-IIA and nitrite into the cell culture medium up to 24?h. Pretreatment with an NO donor, sodium nitroprusside, decreased LPS-induced sPLA2-IIA release and sPLA2 enzyme activity, and enhanced the expression of iNOS and nitrite generation after LPS treatment. Pretreatment with L-NAME, AG, WHI-P154, or Flu notably reduced the expression of iNOS and nitrite, but increased sPLA2-IIA protein levels and sPLA2 enzyme activity. In addition, pretreatment of the cells with STAT1 siRNA inhibited the phosphorylation of STAT1, iNOS expression, and nitrite production, and enhanced the release of sPLA2-IIA. Pretreatment with the specific inhibitors of NOS, JAK2, and STAT3 decreased the permeability of BMVECs. In contrast, inhibition of sPLA2-IIA release increased cell permeability. These results suggest that sPLA2-IIA expression is regulated by the NO-JAK3-STAT1 pathway. Importantly, sPLA2-IIA augmentation could protect the LPS-induced permeability of BMVECs.Our results demonstrate the important action of sPLA2-IIA in the permeability of microvascular endothelial cells during brain inflammatory events. NELF Potentiates Gene Transcription in the Drosophila Embryo

Abstract: A hallmark of genes that are subject to developmental regulation of transcriptional elongation is association of the negative elongation factor NELF with the paused RNA polymerase complex. Here we use a combination of biochemical and genetic experiments to investigate the in vivo function of NELF in the Drosophila embryo. NELF associates with different gene promoter regions in correlation with the association of RNA polymerase II (Pol II) and the initial activation of gene expression during the early stages of embryogenesis. Genetic experiments reveal that maternally provided NELF is required for the activation, rather than the repression of reporter genes that emulate the expression of key developmental control genes. Furthermore, the relative requirement for NELF is dictated by attributes of the flanking cis-regulatory information. We propose that NELF-associated paused Pol II complexes provide a platform for high fidelity integration of the combinatorial spatial and temporal information that is central to the regulation of gene expression during animal development.
Asymptotics of Landau constants with optimal error bounds

Abstract: We study the asymptotic expansion for the Landau constants $G_n$ $$\pi G_n\sim \ln N + \gamma+4\ln 2 + \sum_{s=1}^\infty \frac {\beta_{2s}}{N^{2s}},~~n\rightarrow \infty, $$ where $N=n+3/4$, $\gamma=0.5772\cdots$ is Euler's constant, and $(-1)^{s+1}\beta_{2s}$ are positive rational numbers, given explicitly in an iterative manner. We show that the error due to truncation is bounded in absolute value by, and of the same sign as, the first neglected term for all nonnegative $n$. Consequently, we obtain optimal sharp bounds up to arbitrary orders of the form $$ \ln N+\gamma+4\ln 2+\sum_{s=1}^{2m}\frac{\beta_{2s}}{N^{2s}}< \pi G_n < \ln N+\gamma+4\ln 2+\sum_{s=1}^{2k-1}\frac{\beta_{2s}}{N^{2s}}$$ for all $n=0,1,2,\cdots$, $m=1,2,\cdots$, and $k=1,2,\cdots$. The results are proved by approximating the coefficients $\beta_{2s}$ with the Gauss hypergeometric functions involved, and by using the second order difference equation satisfied by $G_n$, as well as an integral representation of the constants $\rho_k=(-1)^{k+1}\beta_{2k}/(2k-1)!$.
Regulation of Electroacupuncture on Gastric Myoelectrial Activities: Monitered by Electrogastrograms  [PDF]

Abstract:

It is reported that acupuncture on some acupoints can enhance the regularity of gastric myoelectrical activities, which can be observed from the electrogastrograms (EEGs). In this paper, the electroacupuncture (EA) treatments were performed on subjects with slight gastric disorders and the EEGs were monitored in the meanwhile. The EEGs features of baseline, during and after EA treatments were analyzed. The results show that the EA treatments do enhance the regularity of EEGs. And a highly possible pattern for EA to attain the enhancement is to regulate the pacemaker quantity and propagation of slow waves rather than regulating the amplitude of slow waves.

Abstract:

Volatility is an important variable in the financial market. We propose a model-free implied volatility method to measure the volatility and test the volatility risk premium. The model-free implied volatility does not depend on the option pricing model, and extracts information from all the option contracts. We provide empirical evidence from the S & P 500 index option that model-free implied volatility is more accurate to forecast the future volatility and the volatility risk premium does not exist.

Small and Medium Sized Entities Management’s Perspective on Principles-Based Accounting Standards on Lease Accounting  [PDF]

Abstract: Lease accounting is viewed as one of the top priorities for the International Financial Reporting Standards (IFRS) convergence. Small and medium sized entities are an important part of the economy, and this research investigates the management’s perspective on the adoption of principles-based IFRS about lease accounting. This researcher interviewed four managers from three different small and medium sized entities, and found the management to be more concerned about their long-term business success than the change of accounting standards. Only when the entities have a loan with the bank, then the management focuses on the lease classification. The interview also suggests that the managers and business owners in the small and medium sized entities have limited knowledge and skills in accounting reporting standards. These firms outsource their accounting needs to local accountants rather than having their own in-house departments. The other aspect of focus for management of these firms is tax consequence of IFRS adoption. The research suggests other regulatory agencies, i.e., Internal Revenue Service, should also be involved in enhancing financial statement transparency and usefulness after the adoption of accounting standards.
