Evaluation of gene amplification and protein expression of HER-2/neu in esophageal squamous cell carcinoma using Fluorescence in situ Hybridization (FISH) and immunohistochemistry

Abstract: One hundred and ninety-nine ESCC cases were evaluated using the Tissue Microarray (TMA) technique. A polyclonal antibody against c-erbB-2 was used for immunohistochemistry. Analyses were based on the membrane staining pattern. The results were classified according to the Herceptest criteria (DAKO): negative (0/1+), potential positive (2+) and positive (3+). The FISH reactions were performed according to the FISH HER2 PharmDx (DAKO) protocol. In each case, 100 tumor nuclei were evaluated. Cases showing a gene/CEN17 fluorescence ratio ≥ 2 were considered positive for gene amplification.The c-erbB-2 expression was negative in 117/185 cases (63.2%) and positive in 68 (36.8%), of which 56 (30.3%) were 2+ and 12 (6.5%) were 3+. No significant associations were found among protein expression, clinicopathological data and overall survival. Among the 47 cases analyzed, 38 (80.9%) showed no gene amplification while 9 (19.1%) showed amplification, as demonstrated by FISH. Cases that were negative (0/1+) and potential positive (2+) for c-erbB-2 expression by immunohistochemistry showed no gene amplification. However, all cases with gene amplification were positive (3+) by immunohistochemistry. According to univariate analysis, there was a significant difference (p = 0.003) in survival rates when cases with and without HER-2/neu amplification were compared.Our data demonstrate the correspondence between gene amplification and protein expression of HER-2/neu. Abstract: the objective was to evaluate the application of phosphorus doses in parcels in the cotton crop in the systems of cultivation under irrigation and rainfed, being aimed the higher efficiency of phosphate fertilization, in comparison to the traditional application, only at sowing. the experiment was carried out in quartzanic neossoil, in the north region of minas gerais. the adopted design was randomized blocks, with three replicates. the treatments were the doses of 50 and 120 kg ha-1 of p2o5 applied in four ways: ps, p1, p2 and p3 [100/0; 75/25; 50/50; 25/75% of the dose of phosphate fertilizer applied, respectively at the sowing and at 35 days after the emergence (dae)], in the irrigated and rainfed crops. two additional treatments, without p, in the crop without and with irrigation, completed the eighteen studied treatments. the increase of the p doses increased the dry mass of aerial part and the number of reproductive structures at 80 dae, the relative efficiency of the phosphate fertilization and the productivity of cotton in bull. the response to p doses occurred only under irrigation. the parceled application of the phosphate fertilizer did not increase the efficiency of the fertilization in the cotton farm in quartzanic neossoil.
Sleep disturbances in 50 children with attention-deficit hyperactivity disorder

Abstract: objective: this study assesses the relationship between sleep disturbances (sd) and attention-deficit and hyperactivity disorder (adhd) to characterize clinical features and associated problems. method: the medical records of 50 children and adolescents ranging in age from 4 to 17 years with adhd without the diagnosis of mental retardation or pervasive developmental disorders were reviewed. results: significant relationships were found between sd and drug therapy (p<0.01), co-morbidity (p<0.01) and greater adherence to treatment prescribed for adhd disorders (p<0.05). conclusion: the results of this study suggest that sd are an important problem in children with adhd and may be linked to increased symptoms.

Abstract: Las políticas culturales son un campo de problemas construido internacionalmente en la segunda mitad del siglo veinte, con el fin de las guerras mundiales y la conformación del Sistema de las Naciones Unidas, en el marco de la afirmación de los derechos humanos. Las políticas culturales han nacido como políticas de los Estados nacionales, orientadas a su consolidación y legitimación dentro del espacio de su territorio y de las relaciones internacionales. Estas políticas han partido de nociones elitistas y patrimonialistas de la cultura, que siendo practicada y concebida como extra a a las grandes mayorías, debía ser objeto de intervenciones que la difundieran y facilitaran el acceso y la participación de todos los hombres en la vida cultural. Las políticas culturales se han extendido hacia el dominio de las industrias culturales, y más tarde se han vinculado con la problemática del desarrollo. En un contexto de transnacionalización, de expansión del mercado y de reformulación del Estado, su implementación combinó el elitismo con el populismo, y las perspectivas tecnocráticas debilitaron su sentido político. Las asimetrías del libre comercio, el peso creciente de las mega corporaciones y las reivindicaciones de identidad, contribuyeron para que recientemente UNESCO aprobara la Convención sobre la protección y promoción de la diversidad de las expresiones culturales. Esta última reforzó la presencia internacional de la organización y revitaliza el debate sobre las políticas culturales desde nuevas perspectivas.
Histomorphometric study of role of lactoferrin in atrophy of the intestinal mucosa of rats  [PDF]

Abstract: The purpose of this work was to study the effects of oral administration of lactoferrin (Lf) in rats subjected to atrophy of the small intestine induced by a diet based on soy protein concentrate as the main protein source. We used 24 male Wistar rats aged 40 days, kept in individual cages under appropriate conditions of temperature, light and humidity. The animals were divided into four groups (n = 6); 1) group SL received soy-based food and, once a day, a supplement of 200mg/kg of Lf administered by gavage; 2) group Si received the soy feed without supplement of Lf; 3) group CL received a diet based on casein plus Lf; 4) group Ci received the casein diet without supplement of Lf. At the end of fifteen days, a 10 mm segment of the initial portion of the small intestine was sectioned and subjected to morphometry of the intestinal crypts and villi and assessment of the number and size of myofibroblasts. Comparison between groups showed that the length of the villi was similar in groups Ci and CL and higher in CL than in SL; SL than in Si, in Ci than in SL, and in Ci than in Si to Ci. The crypt depth was similar in SL and CL, SL and Ci and Ci and CL and was higher in Si than in Ci and in Si than in SL. The number of myofibroblasts was higher in SL than in CL, in SL than in Si, in CL than in Ci, and in SL than in and Ci; between Ci and Si there was no difference. The area of myofibroblasts was similar between the groups SL and CL and Si and Ci and higher in SL than in Si, and in Cl than in and Ci, and in SL than in Ci. All statistical analysis assumed significance when p < 0.05. From these results, we conclude that lactoferrin increases the number and size of the pericrypt myofibro-blasts and stimulates rapidly the regeneration of atrophied villi.
Entomological investigation of a sylvatic yellow fever area in S?o Paulo State, Brazil

Abstract: following reports of two autochthonous cases of sylvatic yellow fever in the state of s?o paulo, brazil, in 2000, entomological surveys were conducted with the objective of verifying the occurrence of vector species in forest environments close to or associated with riparian areas located in the western and northwestern regions of the state. culicidae were captured in 39 sites distributed in four regions. haemagogus leucocelaenus and aedes albopictus were the most abundant species and were captured in all the regions studied. h. leucocelaenus was the most abundant species in the municipalities of santa albertina and ouroeste, where the two cases of sylvatic yellow fever had been reported. mosquitoes from the janthinomys/capricornii group were only found at eight sites in the s?o josé do rio preto region, while sabethes chloropterus was found at one site in ribeir?o preto. h. leucocelaenus showed its capacity to adapt to a secondary and degraded environment. our results indicate a wide receptive area for yellow fever transmission in the state of s?o paulo, with particular emphasis on the possibility of h. leucocelaenus being involved in the maintenance of this sylvatic focus of the disease.
On the numerical integration of rigid body nonlinear dynamics in presence of parameters singularities

Abstract: one of the main complexities in the simulation of the nonlinear dynamics of rigid bodies consists in describing properly the finite rotations that they may undergo. it is well known that, to avoid singularities in the representation of the so(3) rotation group, at least four parameters must be used. however, it is computationally expensive to use a four-parameters representation since, as only three of the parameters are independent, one needs to introduce constraint equations in the model, leading to differential-algebraic equations instead of ordinary differential ones. three-parameter representations are numerically more efficient. therefore, the objective of this paper is to evaluate numerically the influence of the parametrization and its singularities on the simulation of the dynamics of a rigid body. this is done through the analysis of a heavy top with a fixed point, using two three-parameter systems, euler's angles and rotation vector. theoretical results were used to guide the numerical simulation and to assure that all possible cases were analyzed. the two parametrizations were compared using several integrators. the results show that euler's angles lead to faster integration compared to the rotation vector. an euler's angles singular case, where representation approaches a theoretical singular point, was analyzed in detail. it is shown that on the contrary of what may be expected, 1) the numerical integration is very efficient, even more than for any other case, and 2) in spite of the uncertainty on the euler's angles themselves, the body motion is well represented.
