Biochemical evaluation of a series of synthetic chalcone and hydrazide derivatives as novel inhibitors of cruzain from Trypanosoma cruzi

Abstract: chagas' disease, a parasitic infection widely distributed throughout latin america, is a major public health problem with devastating consequences in terms of human morbidity and mortality. the enzyme cruzain is the major cysteine protease from trypanosoma cruzi, the etiologic agent of american trypanosomiasis or chagas' disease, and has been selected as an attractive target for the development of novel trypanocidal drugs. in the present work, we describe the synthesis and inhibitory effects of a series of thirty-three chalcone and seven hydrazide derivatives against the enzyme cruzain from t. cruzi. most of the compounds showed promising in vitro inhibition (ic50 values in the range of 20-60 μm), which suggest the potential of these compounds as lead candidates for further development. twelve compounds have not been reported before, and four of them (7, 13, 16 e 18) are among the most potent inhibitors of the series.

Abstract: Las reuniones de medicina internacionales constituyen en la actualidad uno de los acontecimientos celebrados con más frecuencia en Espa a, y los organizadores se caracterizan por recurrir a los servicios de interpretación ante la confluencia de distintas lenguas y culturas. Por consiguiente, la medicina representa un ámbito de especialización que ofrece amplias posibilidades a los intérpretes en formación, especialmente a aquellos cuya combinación lingüística es inglés-espa ol, ya que en las últimas décadas se constata el auge del inglés como lingua franca de la comunidad médica internacional. No obstante, son pocos los estudios realizados en el ámbito de la interpretación médica, especialmente los que tratan de profundizar en el mercado para adaptar la formación a las necesidades profesionales reales. Por este motivo, decidimos realizar un estudio téorico-práctico con el objetivo de establecer una base sólida a partir de la cual dise ar posibles programas de formación. La relevancia del presente estudio estriba en la aplicabilidad de los aspectos teóricos y prácticos en la formación del intérprete de medicina, ya que consideramos que dichos aspectos ayudan a conocer las necesidades, requisitos y exigencias tanto de intérpretes profesionales como de médicos usuarios, orientando la preparación del futuro intérprete al ejercicio de la profesión. --------------------------------------------------- A training proposal in interpretation in the medical field. International medical meetings are nowadays one of the events most frequently held in Spain at which organisers commonly employ interpreting services due to the confluence of different languages and cultures. Consequently, medicine constitutes a scientific field that offers a good opportunity for interpreters in training, particularly those who language combination is English-Spanish. As a matter of fact, English has become during the last decades the language par excellence of international medical community. Nevertheless we detected a lack of research projects in the field of medical interpreting, especially those that study in depth the market in order to adapt training to real professional demands. Abstract: La medicina constituye actualmente uno de los ámbitos en los que se celebran con más frecuencia reuniones internacionales multilingües, y los organizadores suelen contratar servicios de interpretación. Por lo tanto, cabría afirmar que la medicina representa un campo de interés creciente para el intérprete de conferencias, en especial para aquel cuya combinación lingüística es inglés-espa ol, ya que no podemos olvidar que la lengua inglesa se ha convertido, en las últimas décadas, en la 'lingua franca' de la comunidad médica internacional. No obstante, a pesar de las posibilidades que ofrece la medicina a los intérpretes de conferencias, se trata de un ámbito olvidado tanto en la investigación como en interpretación como en los planes de estudio de las facultades espa olas de Traducción e Interpretación. Este vacío en la formación lleva al intérpreta a prepararse y a documentarse exhaustivamente antes de la interpretación. En el artículo que nos ocupa, analizaremos la situación de la formación médica y de la especialización de los intérpretes en Espa a, y describiremos el proceso de preparación y documentación que éstos realizan antes de cumplir con un determinado encargo de interpretación. --------------------------------------------------- Interpreting in the field of Medicine; training and specialization. Medicine is currently one of the fields in which international multilingual meetings are most frequently held, and the organisers commonly employ interpreting services. For this reason, we could say that medicine represents an interesting field for conference interpreters, especially those whose language combination is English-Spanish. As a matter of fact, English has become in the last decades the language par excellence of medical science. However, despite the possibilities that this field offers to conference interpreters, medical interpreting is a forgotten area in interpreting research and in the syllabus of Spanish Faculties of Translation and Interpreting. This lack of medical training leads the interpreter to develop a deep process of preparation and documentation. In the present article we will analyze the situation of medical training for interpreters and we will describe the preparation and documentation process developed by the interpreter before undertaking a given interpreting project.

Abstract: bradykinin (bk) a nonapeptide generated in plasma during tissue injury, is involved in many physiological and pathological states. kinin actions are mediated by specific membrane receptors and involve a complex signal transducer and also second messager mechanisms. due to its inequivocal relevance, an intensive effort has been focused in recent years to develop selective and competitive bk antagonists. thus, the development of a new series of peptide bk antagonists has made an important contribution to the understanding of the pharmacological, physiological and pathophysiological role of bk, and this is certain to provide a firm basis for developing new drugs to relieve pain and inflammation. however, bk antagonists derived from peptide origin reported to date have limited clinical use due to their poor oral absortion and short duration of effect. thus, considerable effort has also been made in developing stable nonpeptide bk antagonists. up to now, most nonpeptide compounds reported to exhibit bk antagonistic activity have been derived from plants, including many flavonoids, terpenes, and also synthetic substances with various molecular structures. amongst them, the pregnane glycoside compounds isolated from the plant mandevilla velutina are the most promising. these compounds are effective in antognizing bk responses in a variety of preparations, and they also exhibit potent and long-lasting analgesic and anti-inflammatory activities. the exact mechanism underlying their action however, is not yet completely understood.

Abstract: Nine 3,3’-(arylmethylene)bis(2-hydroxynaphthalene-1,4-dione) derivatives were synthesized through the reaction between 2-hydroxy-1,4-naphthalen-1,4-dione and different aromatic alde-hydes in water applying ultrasonic irradiation for 5 min at room temperature and microwave irradiation for 15 min at 70°;C. Two of the nine derivatives, compounds 3-e and 3-i, obtained from 3-bromo-hydroxybenzaldehyde and 5-methylfuran-2-carbaldehyde, respectively, are previously unpublished. The structures of all compounds were established on the basis of their spectral data and mass analysis. The attractive features of this synthesis protocol include mild conditions, high atom-economy and excellent yields with the elimination of water as the only by-product.
