Role of cytokines and nitric oxide in the induction of tuberculostatic macrophage functions

Abstract: The aim of this study was to determine phenotypic differences when BCG invades macrophages. Bacilli prepared from the same BCG primary seed, but produced in different culture media, were analysed with respect to the ability to stimulate macrophages and the susceptibility to treatment with cytokines and nitric oxide (NO). Tumour necrosis factor (TNF) activity was assayed by measuring its cytotoxic activity on L-929 cells, interleukin-6 (IL-6) and interferon γ (IFN-γ) were assayed by enzyme-linked immunosorbent assay (ELISA), whereas NO levels were detected by Griess colorimetric reactions in the culture supernatant of macrophages incubated with IFN-γ , TNF or NO and subsequently exposed to either BCG-I or BCG-S. We found that BCG-I and BCGS bacilli showed different ability to simulate peritoneal macrophages. Similar levels of IL-6 were detected in stimulated macrophages with lysate from two BCG samples. The highest levels of TNF and IFN-γ were observed in macrophages treated with BCG-S and BCG-I, respectively. The highest levels of NO were observed in cultures stimulated for 48h with BCG-S. We also found a different susceptibility of the bacilli to ex ogenous treatm ent w ith IFN-γ and TNF which were capable of killing 60 and 70% of both bacilli, whereas NO was capable of killing about 98 and 47% of BCG-I and BCG-S, respectively. The amount of bacilli proportionally decreased with IFN-γ and TNF, suggesting a cytokine-related cytotox ic effect. Moreover, NO also decreased the viable number of bacilli. Interestingly, NO levels of peritoneal macrophages were significantly increased after cytokine treatment. This indicates that the treatment of macrophages with cytokines markedly reduced bacilli number and presented effects on NO production. The results obtained here emphasize the importance of adequate stimulation for guaranteeing efficient killing of bacilli. In this particular case, the IFN-γ and TNF were involved in the activation of macrophage bactericidal activity.

Abstract: in brazil, public health specialization courses with the characteristics presently adopted by the national school of public health, i.e., short duration and great amplitude, are relatively recent, having started approximately fifteen years ago. in brasilia, nine such courses have been already completed, having, six of them, directive and/or teaching envolvement by the authors. results on a final assessment of one the courses are herewith presented in order to introduce our experience as a contribution to future analogous iniciatives. the assessment was applied to a random sample of students, teachers and coordinators, with the objective of identifying positive and negative appreciations about methodological, operational, administrative and institutional aspects. answers were quite variable, with the highest frequencies scored to field work and to the apprehension on admitting graduates to the health system. such aspects deserve final differentiated discussion.

Abstract: this article compares food intake by women 35 years or older in two population-based cross-sectional studies in rio de janeiro, brazil, in 1995-1996 (n = 1,014) and 2004-2005 (n = 1,001). food intake was assessed with a food-frequency questionnaire, and nutritional status was defined according to body mass index (bmi = weight/height2). prevalence of obesity (bmi 30kg/m2) increased in the ten-year period (16.6% to 24%). many high energy density (kcal/g) foods showed a statistically significant increase in the period, such as crackers, candies, bacon, sausage, and hamburger. some high energy density items were reported less frequently: butter, mayonnaise, potato chips, and sugar. the intake of fruits, milk, beans, roots and potatoes, and meat decreased in the 10-year period. women with more education showed a larger reduction in fruit and meat intake and a smaller reduction in fish, dairy product, and root and potato intake. changes in prevalence of obesity were associated with numerous changes in food intake, depending on the level of schooling.
Raz?o cintura/quadril como preditor de hipertens?o arterial

Abstract: this study aims to define cut-off points for the waist:hips girth ratio (whr), using arterial hypertension as the outcome. the data refer to 3,282 individuals over twenty years of age examined in a survey conducted in the municipality of rio de janeiro in 1995-1996, using a two-stage sample. sixty census tracts were drawn initially; subsequently, 34 households were selected systematically from each tract. stature, weight, waist and hips girths, and blood pressure were measured in the households. the criterion for hypertension was a systolic blood pressure of 140 mmhg or diastolic pressure of 90 mmhg, or use of medication to reduce blood pressure. the sensitivity and specificity of different cut-off points for whr were calculated in the prediction of arterial hypertension according to sex, age, and presence of overweight, classified according to world health organization guidelines. the best cut-off points for whr were 0.95 for men and 0.80 for women. compared to the waist:stature ratio and waist circumference, the whr proved more capable of predicting arterial hypertension and less correlated with body mass index.

Abstract: this work has the aim of investigating, in a psychogenetic context, the possibilities of a game of rules to be introduced in the psycho-pedagogical diagnosis with the purpose of evaluating the construction of classification awareness. forty students of both genders with ages between 4 and 17 years old attending primary (kindergarten and 1st grade) and elementary (3rd to 8th grade) public schools have participated on this research, 10 in each developing level: figural collection, non-figural collection, hierarchical inclusion of classes and logical thinking. spontaneous classification tests were applied, along with hierarchical inclusion of classes, matching cards of various colors and four matches with the game "find out the animal". the results point out that the game utilized was effective to help on psycho-pedagogical diagnosis for the classification.

Abstract: Este trabalho tem como objetivo definir pontos de corte para a raz o cintura/quadril (RCQ), usando como desenlace a hipertens o arterial. Os dados obtidos referem-se a 3.282 indivíduos com idade acima de vinte anos, examinados numa pesquisa realizada no Município do Rio de Janeiro, em 1995-1996, utilizando amostra em dois estágios. Inicialmente, foram sorteados sessenta setores censitários; posteriormente, 34 domicílios foram selecionados sistematicamente em cada setor. Altura, peso, perímetros da cintura e do quadril e press o arterial foram medidos nos domicílios. Foram considerados hipertensos os que apresentavam press o sistólica 140 mmHg ou press o diastólica 90 mmHg, ou, ainda, os que faziam uso de medicamento para reduzir a press o arterial. Foram calculadas a sensibilidade e a especificidade de diferentes pontos de corte para a RCQ na predi o de hipertens o arterial segundo sexo, idade e presen a de sobrepeso, classificado segundo proposta da Organiza o Mundial da Saúde. Os melhores pontos de corte para a RCQ foram 0,95 para homens e 0,80 para mulheres. Comparada com a raz o cintura/altura e com o perímetro da cintura, a RCQ apresentou maior capacidade preditiva de hipertens o e menor correla o com o índice de massa corporal.
Acurácia da circunferência da cintura e da rela??o cintura/quadril como preditores de dislipidemias em estudo transversal de doadores de sangue de Cuiabá, Mato Grosso, Brasil

Abstract: this study evaluated the accuracy of anthropometric fat location indices (waist circumference and waist-to-hip ratio) as predictors of dyslipidemia, in addition to defining their respective cut-offs. the sample consisted of 416 healthy male blood donors from 20 to 58 years of age at the blood center in cuiabá, capital of mato grosso state, brazil. serum lipids were determined through the enzymatic method. data analysis through the roc curve identified the best indicator and the cut-off with the largest statistically significant area under the roc curve. the best cut-off points for waist-to-hip ratio and waist circumference were 0.90 and 85cm, respectively. the area under the roc curve between waist circumference and high cholesterol/hdl relation included the 0.50 value, so waist circumference could not be considered a predictor of dyslipidemia. the best sensitivity and specificity were in the youngest group. waist-to-hip ratio was a better predictor of dyslipidemia than waist circumference, although the latter showed better discriminating power to detect hypertriglyceridemia. the most accurate cut-off points were lower those set by the world health organization.

Abstract: objective: to evaluate the association between alcohol consumption and abdominal fat. methods: cross-sectional study carried out in a sample of male blood donors (n=1,235), aged 20-59 years, in the city of cuiabá, central-west brazil, between august 1999 and january 2000. waist circumference and waist-to-hip ratio were indicators of abdominal fat, adjusted for total adiposity. weight, height, waist and hip circumferences were measured. alcohol consumption was evaluated using a questionnaire collecting information on type, frequency, and amount of consumption. the association between alcohol consumption and abdominal fat was assessed through multiple linear regression models adjusted for age, physical activity, smoking, and percent of body fat. results: after adjustment, waist circumference and waist-to-hip ratio were positively associated with beer (p = 0.02) and total alcohol consumption (p=0.01; p=0.03, respectively). waist circumference was positively associated with spirit consumption (p=0.04). conclusions: alcohol intake, particularly beer, was positively associated to abdominal fat.

Abstract: objective:to assess the effect of exposition to tobacco's smoke on infants' growth. methods:a cross-sectional population-based study was carried out with infants up to two years old who went to public primary healthcare units for immunization. ten units with 200 infants each were randomly selected. the parents or people in charge of 1,437 children answered a questionnaire. weight and length were measured before the immunization. for multivariate analysis two models were built, whose dependent variable were the continuous ones, weight-for-age and length-for-age, which were built using the conceptual hierarchical modeling. results:prevalence of low-length was 4.7%, and of low-weight was 3.0%. among all children in the study, 35.8% were exposed to cigarette smoking of people in the household. after adjustments for socio-demographic variables and biological variables of the child, only the mother's smoking showed independent effect on the nutritional status of children, where the smoking habits during pregnancy remained associated with length-for-age (β=-0.226; p=0.02) and smoking of the mother with weight-for-age (β=-0.235; p=0.02). the smoking of the father and other people in the household's smoking did not present statistically significant association for any studied outcomes. conclusion:smoking during pregnancy showed independent effect on the length-for-age indicator and mother's smoking with weight-for-age of infants.
Acurácia da circunferência da cintura e da rela o cintura/quadril como preditores de dislipidemias em estudo transversal de doadores de sangue de Cuiabá, Mato Grosso, Brasil

Abstract: Avaliou-se a acurácia dos indicadores antropométricos de localiza o de gordura (circunferência da cintura e rela o cintura/quadril) como preditores de dislipidemias, apresentando seus pontos de corte. A amostra consistiu-se de 416 homens saudáveis, de 20 a 58 anos, doadores de sangue do hemocentro de Cuiabá, Mato Grosso, Brasil. Os lipídios séricos foram determinados por método enzimático. A análise dos dados foi feita através da curva ROC para identificar o melhor ponto de corte dos indicadores e a maior área estatisticamente significante sob a curva. Os melhores pontos de corte para rela o cintura/quadril e circunferência da cintura foram 0,90 e 85cm, respectivamente. O IC95% da área sob a curva ROC entre a circunferência da cintura e a rela o colesterol/HDL elevada incluiu o valor 0,50, limitando a capacidade da circunferência da cintura como preditor de dislipidemias. Valores de sensibilidade e especificidade foram melhores no grupo mais jovem. A rela o cintura/quadril foi mais associada às dislipidemias do que a circunferência da cintura. Circunferência da cintura mostrou melhor poder discriminatório na detec o de hipertrigliceridemia. Os pontos de corte de melhor acurácia foram menores do que os estabelecidos pela Organiza o Mundial da Saúde.
