Abstract: the objective of this work was to study the effect of 2,4-d and bap in the induction of callus formation from leaf segments of coffea arabica l. cv rubi plantlets obtained through in vitro germination as well as to determine its growth curve, protein pattern and levels of total proteins and reducing sugars. the results indicated that explants inoculated in the absence of growth regulators or in the presence of bap only showed no callus formation. maximum callus production is obtained when 1.0 mg l-1 2,4-d is used. the callus growth curve showed a sigmoidal shape with 3 distinct phases: lag (0-42 d), exponential (42-77 d) and linear (77-84 d). it was not observed accumulation of proteins in any of the phases of callus growth and maximum levels of reducing sugars were observed in explants at the inoculation day. the callus protein pattern presented quantitative (intensity of bands) and qualitative (presence or absence of bands) differences.
Linfonodo sentinela em melanoma de crian a: relato de caso

Abstract: Objetivo: apresentar um caso de melanoma cutaneo na infancia, submetido à biópsia de linfonodo sentinela. Descri o: crian a de 12 anos de idade, portadora da síndrome do nevo displásico, desenvolveu melanoma em dorso. A biópsia excisional revelou melanoma (Breslow = 1,5 mm), sendo submetida a tratamento cirúrgico da les o, com amplia o de margem de 2cm e biópsia de linfonodo sentinela. O exame anatomopatológico n o mostrou doen a residual. O exame do linfonodo sentinela n o mostrou metástases ao exame histopatológico por hematoxilina eosina, nem à imunohistoquímica (S100 e HMB45). No entanto, a pesquisa de RNA mensageiro da tirosinase por RT-PCR se mostrou positiva. A paciente n o mostrou sinais de metástase ou recidiva local nestes doze meses iniciais de seguimento. Comentários: o melanoma é raro em crian as, corresponde a menos de 1% dos tumores da infancia. Inúmeros trabalhos mostram o aumento da incidência mundial do melanoma. Em cerca de 70%, se originam de nevos melanocíticos pré-existentes, e nos 30% restantes, eles surgem de novo. Dada a agressividade do melanoma, a sobrevida depende do diagnóstico precoce. A biópsia do linfonodo sentinela tem contribuído na decis o de se realizar ou n o a linfadenectomia completa, e alguns autores já est o utilizando a mesma em crian as.

Abstract: salix is an ornamental tree adapted to humid soils being considered an important species used in depleted areas. its wood may be used in all kinds of industries. although the production of seeds is large, the germination is reduced. the objective of this work was to establish salix in vitro through the induction and formation of friable callus of leaf explants to provide future regeneration of plants for mass propagation of the species. leaf explants were inoculated in ms medium (murashige & skoog, 1962) supplemented with different concentrations of 2-4-diclorofenoxiacetic acid (0; 1.0; 2.0; 4.0; 6.0; 8.0 and 12.0mg l-1) and combinations of 1-naphthaleneacetic acid and 6-benzylaminopurine both with the concentrations of 0; 1.0; 2.0; 4.0; 6.0 e 8.0mg l-1. the results showed that explants inoculated in the absence of growth regulators had no formation of friable callus. significant production of friable callus (90%) was obtained with 6.0mg l-1 of 2-4-diclorofenoxiacetic acid. individual concentrations of 1-naphthaleneacetic acid or 6-benzylaminopurine induced callogenesis and rooting.
Experimental model in rat for sentinel node biopsy

Abstract: although sentinel node procedure has been used world wide, there are many aspects to be defined and better standardized. this study address if the experimental model in rats is appropriate for sentinel node biopsy. in this model, the lymph nodes are showed by lymphoscintigraphy, they are dyed by patent blue and identified by intraoperative gamma probe detection. it isn?t necessary to use magnification for the procedure. the model demonstrated that sentinel node biopsy in rats is feasible. so, besides allowing researches in this field, the model is useful for training and diffusing this technique.

Abstract: the sexual propagation of smilax japecanga grisebach is limited, due to seed dormancy which may take 6 to 8 months to overcome, making the production of seedlings a difficult task. in order to promote seed germination, experiments on dormancy breakage were performed. mechanical scarification using an emery; chemical scarification with 98% sulfuric acid for 30 seconds, 1, 5 and 10 minutes; imbibition for 48 hours with ga3 0.55, 1.1, 1.65 and 2.2 mm and a combination of chemical scarification for 6 minutes plus imbibition in ga3 1.92 mm for 48 hours were used. the combination of chemical scarification plus imbibition with ga3 1.65 mm promoted the highest germination (86%), followed by imbibition in 1.65 mm ga3 (56%). lower germination percentage was obtained using mechanical scarification (8%) and chemical scarification for 1 minute (7%).

Abstract: aspects of leaf anatomy, extraction and characterization of essential oils present in leaf tissues of pindaíba (xylopia brasiliensis), were studied. the anatomical cuts were manually performed and their visualization were made through a coloration process using safranine and astra blue. the extraction and characterization of the essential oil used the steam drag’s method. with these studies it was observed the presence of oily idioblasts filled mainly with cariophylene oxide.

Abstract: due the presence of seed dormancy, the propagation of caryocar brasiliense requires studies in order to obtain young asexually plants. this work had as objective contribute to the establishment of a "pequizeiro" micropropagation protocol. for shoot induction, explants were inoculated in wpm medium with different concentrations and interactions of bap and naa. for rooting induction, shoots obtained in vitro were transferred to wpm medium supplemented with different concentrations of iba in the presence or absence of activated charcoal. the best treatment for shoot induction was the use of 0,05mg l-1 ana + 0,75mg l-1 bap, which produced an average of 6 shoots/explant (17,4) providing the highest rate of multiplication (8,7). the use of 3mg l-1 iba promoted root induction in all explants generating average of 12,87 roots per explant. roots developed in the presence of activated charcoal presenting higher length (33,16 mm) and higher number of secondary roots (19,53).

Abstract: caffeine, the alkaloid known as 1,3,7 - trimetilxantina, is found in quiescent coffee seeds, amounting to a total of 1.1 to 1.7% in coffea arabica l. and 2 to 3% in coffea canephora pierre, mostly localized in theo endosperm, in the free cytoplasm of cells or complexed with chlorogenic acids. with the physiological function in plants not yet completely understood, caffeine causes an allelophatic effect, either inhibiting germination of a number of species or as an anti-herbivore or a natural pesticide agent. the slow germination of coffee seeds has not yet been elucidated and several causes are pointed to, such as the presence of the endocarp, low water and o2 uptake and presence of natural inhibitors and hormonal balance. although suggested, studies on coffee seed inhibition by the action of endogenous or exogenous caffeine are scarce. thus, the present study was designed to investigate the effect of exogenous caffeine on both germination and embryo development of coffea arabica l. and coffea canephora pierre. the experiment was conducted by utilizing seeds from red ripe berries of the cultivars rubi and apoat? iac-2258. after disinfecting the berries for 30 minutes by soaking in sodium hypochlorite (2% a.i.) and washing three times in distilled water and autoclaved, the embryos were removed and inoculated in a aseptic manner, in petrri dishes with 50% ms medium with the addition of sucrose and supplemented with different of concentrations of caffeine (0.00; 0.05; 0.10; 0.15; 0.20; 0.25; 0.30 and 0.40%). the embryos were maintained in a growth room at 27±20oc and photon flow density of 13μmol.m-2.s-1 for 23 days, when shoot length, root length and seedling fresh mass were evaluated. five days after cultivation, the percentage of radicle emission and that of cotyledons were evaluated, calculating the embryos with open cotyledons and with expanded radicles. a complete randomized experimental design was used six replications per treatment, each replicate consisting of

Abstract: objective: to present a case of a child who was subjected to sentinel lymph node biopsy for cutaneous melanoma. description: a 12 year-old child with dysplastic nevus syndrome developed melanoma on the lumbar region. the excision biopsy revealed a melanoma with depth of 1.5 mm. the patient was submitted to amplification of the margins 2 cm in all directions and the sentinel node was also excised. the histopathological exam did not show residual disease. sentinel on exam did not show metastases either under hematoxylin-eosin stain or immunohistochemistry (s-100 and hmb45). therefore, rt-pcr for tyrosinase mrna was positive. the patient has been followed for twelve months without evidence of recurrence. comments: childhood melanoma is rare, corresponding to less than 1% of malignant tumors in children. data point to a worldwide increase in its incidence. melanoma occurs in melanocytic lesions in 70% and in the remaining 30% it occurs de novo. melanoma is very aggressive, so the survival depends on an early diagnosis. sentinel lymph node biopsy has selected patients to complete lymphadenectomy. some authors have been using this technique in childhood melanoma.
