P53 protein expression and cell viability in irradiated peripheral blood mononuclear cells as bioindicators of radiosensitivity  [PDF]

Abstract: Cellular radiosensitivity is directly correlated with the mechanism of DNA repair, in which p53 protein plays a major role. In this context, this study correlated cell death with p53 expression in lymphocytes irradiated in vitro with different doses of gammaradiation. For this, peripheral blood samples were collected from 10 healthy subjects. Each sample was divided in aliquots and, separately, irradiated with doses of 0,5; 2 and 4 Gy. After this, peripheral blood mononuclear cells (PBMCs) were isolated and cultivated during 72 hours in 5% CO2 at 37ºC without mitogen stimulation. The expression of p53 protein was evaluated by flow cytometry. In parallel, cell viability was determined by trypan blue staining. Statistical analysis was performed us-ing analysis of variance (ANOVA), differences were considered as statistically significant when p < 0.05. The results showed an increase of p53 expression with the absorbed dose, which was proportional to cell death, suggesting that p53 can be used as bioindicator of individual radiosensitivity.

Abstract: Publicamos a seguir um documento dirigido ao Imperador dom Pedro II, em 1861, denunciando, sob o ponto de vista de um eclesiástico católico, as interferências praticadas por James Newell Gordon, superintendente da Saint John d'el Rey Mining Company, Limited (Morro Velho), nas práticas religiosas dos escravos (que à época eram quase 3000) e os atritos que ocorriam entre catolicismo e protestantismo dentro da companhia. Suas entrelinhas, no entanto, acabam por revelar outros importantes aspectos do mundo do trabalho na minera o aurífera inglesa em Minas Gerais, na segunda metade do século XIX, como por exemplo, os castigos, pris es, descanso dominical, assistência religiosa aos enfermos, mendicancia, rela es de hierarquia, dentre outros. We are publishing a document sent to the Emperor of Brazil, D. Pedro II, in 1861, denouncing, under a catholic clergyman's point of view, the interferences practiced by James Newell Gordon, superintendent of Saint John d'el Rey Mining Company, Limited (Morro Velho), in the slaves' religious practices and the attritions which happened between Catholicism and Protestantism in this company. Between the lines, however, we can observe that it ends for revealing other important aspects of the world of the work in the English auriferous mining in Minas Gerais, in the second half of the century XIX, as for instance, punishments, prisons, Sunday rest, religious attendance to patients, begging, hierarchy relationships, among others.
Does low dose13C-urea breath test maintain a satisfactory accuracy in diagnosing Helicobacter pylori infection?

Abstract: context: the standard doses of 13c-urea in 13c-urea breath test is 75 mg. objective: to assess the diagnostic accuracy of 13c-urea breath test containing 25 mg of 13c-urea comparing with the standard doses of 75 mg in the diagnosis of helicobacter pylori infection. methods: two hundred seventy adult patients (96 males, 174 females, median age 41 years) performed the standard 13c-urea breath test (75 mg 13c-urea) and repeated the 13c-urea breath test using only 25 mg of 13c-urea within a 2 week interval. the test was performed using an infrared isotope analyzer. patients were considered positive if delta over baseline was >4.0‰ at the gold standard test. results: one hundred sixty-one (59.6%) patients were h. pylori negative and 109 (40.4%) were positive by the gold standard test. using receiver operating characteristic analysis we established a cut-off value of 3.4% as the best value of 25 mg 13c-urea breath test to discriminate positive and negative patients, considering the h. pylori prevalence (95% ci: 23.9-37.3) at our setting. therefore, we obtained to 25 mg 13c-urea breath test a diagnostic accuracy of 92.9% (95% ci: 88.1-97.9), sensitivity 83.5% (95% ci: 75.4-89.3), specificity 99.4% (95% ci: 96.6-99.9), positive predictive value 98.3% (95% ci: 92.4-99.4), and negative predictive value 93.0% (95% ci: 88.6-96.1). conclusions: low-dose 13c-urea breath test (25 mg 13c-urea) does not reach accuracy sufficient to be recommended in clinical setting where a 30% prevalence of h. pylori infection is observed. further studies should be done to determine the diagnostic accuracy of low doses of 13c-urea in the urea breath test.

Abstract: the objective of the present study was to evaluate the lambari?s performance (oligusarcus argenteus) using fly worms (musca domesticus) as a feeding source in substitution to ration. six hundred fingerlings of lambari bocarra were used with initial mean weight of 0.88 ± 0.14 g and an average length of 3.62 ± 0.30 cm. a complete randomized experimental design was used with five treatments, t1: 100% of ration, t2: 25% of fly worms and 75% of ration, t3: 50% of fly worms and 50% of ration, t4: 75% of fly worms and 25% of ration, and t5; 100% of fly worms with four repetitions and 30 fingerlings in each aquarium. fingerlings were kept in an aquarium of 1.000 l volume. t1 treatment showed no significant differences (p>0.05) for final weight, weight gain, daily weight gain, and total length compared to the other treatments, obtaining the worst results. for the standard length, treatments t2, t3, and t5 obtained the best results (p<0.05). there was no significant difference (p>0.05) for specific growth rate for all treatments. for the carcass yield the best results were observed in treatments t2 and t4. treatment t1 obtained the worst result for viscer somatic index (p<0.05). for the production of lambari, it could be used 75% of fly worms instead of 32% pb ration, which provides better results for carcass yield. for final weight, weight gain, daily weight gain and total length it could be used up to 100% of fly worms in substitution to 32% crude protein ration.

Abstract: this article discusses the paving of the br-174 roadway in the mid-1990s and socio-spatial consequences in the state of roraima. the case is presented as an example of the transformation that territorial interventions have evinced in the amazon region within a context of state neoliberalization and the production of a transnational economic space. federal investments for roadway development and ancillary infrastructure brought about unprecedented connectivity and spearheaded economic growth. yet, the article argues that, besides new discourses of sustainable regional development, the logics dominating projects such as the br-174 upgrade are those of neoliberal transnational expansion. the entrepreneurial state becomes a partner for the establishment of external markets rather than the main agent for the integration of the national territory. therefore, in the br-174 plan, roraima appears mainly as strategic space for crossboundary integration and the linking of the brazilian amazon to caribbean ports. nevertheless, after over a week since the project was concluded, territorial concentration deepened and new socio-spatial inequalities have emerged in close relation to the functions and flows that roadway integration created.

Abstract: Este artigo discute a pavimenta o da BR-174 na metade da década de 1990 e as suas consequências socioespaciais no estado de Roraima. O caso é apresentado como um exemplo da transforma o das interven es territoriais na Amaz nia no contexto da neoliberaliza o do Estado e da produ o de um espa o econ mico transnacional. Os investimentos federais para o desenvolvimento rodoviário e infraestruturas auxiliares trouxeram conectividade sem precedentes para o estado de Roraima, além de gerar crescimento econ mico. Todavia, o artigo argumenta que, além dos novos discursos de desenvolvimento regional sustentável, as lógicas dominantes dos projetos, tais como o melhoramento da BR-174, s o as de um neoliberalismo em expans o transnacional, no qual o Estado empreendedor torna-se um parceiro para o estabelecimento de mercados externos mais do que o principal agente da integra o do território nacional. Assim, no desenho da BR-174, Roraima aparece, principalmente, como um espa o estratégico de conex o transfronteiri a que liga a Amaz nia brasileira aos portos do Caribe. Entretanto, passada mais de uma década desde a conclus o do projeto, a concentra o territorial tem se aprofundado ao passo que emergem novas desigualdades socioespaciais intimamente atreladas às fun es e fluxos que a integra o rodoviária criou.
