Abstract: colorectal cancer is a common malignancy in our clinical practice and it often evolves to a metastatic stage. cutaneous dissemination, however, is a rare form of presentation of this disease. this study reports the case of a 38-year-old female patient that, even after neoadjuvant chemotherapy, presented cutaneous metastases of signet-ring cell colorectal adenocarcinoma. the malignancy proved to be extremely aggressive, without response to clinical therapy nor allowing surgical management, leading the patient to death about six months after the diagnosis.

O cancer colorretal é uma neoplasia comum em nossa prática clínica e frequentemente evolui ao estágio metastático. Dissemina o cutanea, entretanto, é uma forma rara de manifesta o da doen a. Apresentaremos aqui o caso de uma paciente de 38 anos que, mesmo após a realiza o de quimioterapia neoadjuvante, apresentou metástases cutaneas de um adenocarcinoma colorretal com células em anel de sinete. A neoplasia demonstrou-se extremamente agressiva, n o respondendo ao tratamento medicamentoso nem permitindo o tratamento cirúrgico, levando a paciente ao óbito cerca de seis meses após o diagnóstico.

Flow cytometry analysis of the effect of allopurinol and the dinitroaniline compound (Chloralin) on the viability and proliferation of Leishmania infantum promastigotes

Abstract: Background Leishmaniasis is a major parasitic disease in the tropical regions. However, Leishmania infantum has recently emerged as a very important cause of opportunistic infections for individuals positive for human immunodeficiency virus (HIV). However, there is a lack of in vitro tests for assessing the effect of anti-parasitic drugs on the viability and proliferation of Leishmania infantum. The aim of this study is to assess the efficacy of anti-parasitic drugs like allopurinol and Chloralin on the viability and proliferation of L. infantum promastigotes by utilizing two complementary flow cytometric approaches after exposure of the promastigotes to various concentrations of the drugs. Results The density of the cultures in the presence and absence of allopurinol was determined by haemocytometer enumeration. The two flow cytometric approaches used to monitor the drug effect were: (i) a quantitative method to measure cell division using 5-,6-carboxyfluorescein diacetate succinimidyl ester (CFSE) staining and (ii) evaluation of cell viability by dual-staining with the membrane-permeable nuclear stain, SBRY-14 and propidium iodide. It was found that concentrations of allopurinol above 50 μg/ml yielded a clear decrease in the proliferation rate of the promastigotes. However, the viability results showed that about 46.8% of the promastigotes incubated in the presence of 800 μg/ml of allopurinol were still alive after 96 hours. In sharp contrast, more than 90% of promastigotes treated with Chloralin 10 μM (2.7 μg/ml) were dead after 48 hours of treatment. These flow cytometric findings suggest that allopurinol has a leishmaniostatic effect while the dinitroaniline compound (Chloralin) has a leishmaniocidal effect against promastigotes. Conclusions The flow cytometric data on proliferation and viability were consistent with results obtained from haemocytometer counts and allowed us to develop a model for assessing in vitro the effects of medicaments like allopurinol and chloralin on L. infantum promastigotes on a cellular level.

Abstract:

Phytochemicals (PHT) are biologically active chemicals produced by plants, non-essential nutrients, with medicinal properties. In this short communication we report the dramatic improvement in the physical and clinical well-being of 9 terminal AIDS patients that received Phyto V7, a PHT mix, for a period of 3 months. All patients living in the Dr. Cruz Jiminian Foundation (hospice) in Santo Domingo, Dominican Republic, were in an emaciated condition—most could not eat, sit down, shower, stand up or dress alone; all had high viremia (from ~50,000 to above 500,000; 243,816 ± 176,724 HIV-1 RNA copies/ml) and very low CD4+ T-cells counts (142 ± 51 counts/mm3). The clinical status of all patients was C3 according to the United States Centers for Disease Control (CDC) status index. As antiretroviral treatment was not available to the Foundation at the time of the study, the only treatment that they received was Phyto V7 supplementation. Each individual received 5 tablets 3 times a day, each tablet containing 750 mg of Phyto V7. At the end of the 3 months, Phyto V7 supplementation radically improved the well-being of all 9 patients. All patients could eat, sit down, shower, stand up and dress alone. This study supports the notion that PHT supplementation can improve significantly the well-being of terminally ill AIDS patients and is the foundation to conducting further control studies to substantiate this notion.

Abstract: El artículo analiza los conceptos y círculos de negación que el teólogo, médico y psicoanalista francés Marc Oraison desarrolló en su pensamiento sobre sexualidad, particularmente en lo referido a sus estudios y reflexiones sobre homosexualidad. Desde una mirada antropológica, el texto pretende deshilar la red de interpretaciones psicosociales y morales que Oraison fue tejiendo en torno a los ideales estereotipados y estigmatizados de la homosexualidad y de l@s homosexuales, de su presunta sexualidad patológica y condición desviada. Una lectura detenid del pensamiento de Marc Oraison permite apreciar en el caso de este autor, pero extrapolable a otros muchos coetáneos suyos, un amplio marco del basamento intelectual que da argumentos a la reflexión y actitud homófobas que podemos encontrar aún hoy en día en muchos sectores de la sociedad occidental. Tomando como guión principal su obra "El problema homosexual", el artículo hace una parada sobre los distintos círculos de negación que el autor francés construyó en torno a la homosexualidad, pasando tanto por negarla como por afirmarla para llegar siempre a la presión social sobre la persona homosexual. Finalmente, el artículo analiza la negación de los homosexuales como colectivo con identidad reivindicativa, por parte del pensamiento sociorreligioso, que observa a los homosexuales como "criaturas de Dios".
