Drug Screening for Autophagy Inhibitors Based on the Dissociation of Beclin1-Bcl2 Complex Using BiFC Technique and Mechanism of Eugenol on Anti-Influenza A Virus Activity

Abstract: Autophagy is involved in many human diseases, such as cancer, cardiovascular disease and virus infection, including human immunodeficiency virus (HIV), hepatitis C virus (HCV), influenza A virus (IAV) and coxsackievirus B3/B4 (CVB3/B4), so a drug screening model targeting autophagy may be very useful for the therapy of these diseases. In our study, we established a drug screening model based on the inhibition of the dissociation of Beclin1-Bcl2 heterodimer, an important negative regulator of autophagy, using bimolecular fluorescence complementation (BiFC) technique for developing novel autophagy inhibitors and anti-IAV agents. From 86 examples of traditional Chinese medicines, we found Syzygium aromaticum L. had the best activity. We then determined the anti-autophagy and anti-IAV activity of eugenol, the major active compound of Syzygium aromaticum L., and explored its mechanism of action. Eugenol could inhibit autophagy and IAV replication, inhibited the activation of ERK, p38MAPK and IKK/NF-κB signal pathways and antagonized the effects of the activators of these pathways. Eugenol also ameliorated the oxidative stress and inhibited the expressions of autophagic genes. We speculated that the mechanism underlying might be that eugenol inhibited the oxidative stress and the activation of ERK1/2, p38MAPK and IKK/NF-κB pathways, subsequently inhibited the dissociation of Beclin1-Bcl2 heterodimer and autophagy, and finally impaired IAV replication. These results might conversely display the reasonableness of the design of our screening model. In conclusion, we have established a drug screening model for developing novel autophagy inhibitor, and find eugenol as a promising inhibitor for autophagy and IAV infection.
The Progresses of preparation and modification of graphene on substrate

Abstract: It is significant to prepare large area of high quality graphene for the study of the characteristics of graphene and the research of the nano-devices based on graphene. This paper summarizes the experiment progresses and mechanism of graphene grown on different substrates. Nowadays, we can obtain the large area of high quality graphene by using the methods, such as CVD, epitaxial growth, etc. The interaction between the graphene and the substrates is closely related to the mismatch of the lattice, weakness of the bonds and the transformation of the electrons, which has a great influence on geometry, energy band and the properties of electrons of the graphene. The combination of the experiment and the calculation can make deeper understanding of the mechanism of the effects between graphene and different substrates, which can be served as a guide for further study.

Abstract: Rough集理论是一种处理不完备信息系统的数学工具,但是Pawlak的经典rough集理论似乎是不可行的,因为它要求论域中数据间有很强的等价关系。在产生基本集(相似类)时,一般相似关系的分类误差较大,集对分析会把两个对立度不为O的个体划分在一起。汲取两者的优点,给出相同度的概念,只有满足一般相似关系并且相同度大于或等于闽值α的两个对象才能划分在一个基本集中,在此基础上建立基于α相同度相似关系的rough集模型。通过实例验证效果要比基于集对分析或者一般相似关系的模型更好。
Coarsening behavior of MX carbonitrides in type 347H heat-resistant austenitic steel during thermal aging

Abstract: In this work, the growth kinetics of MX (M = metal, X = C/N) nanoprecipitates in type 347H austenitic steel was systematically studied. To investigate the coarsening behavior and the growth mechanism of MX carbonitrides during long-term aging, experiments were performed at 700, 800, 850, and 900°C for different periods (1, 24, 70, and 100 h). The precipitation behavior of carbonitrides in specimens subjected to various aging conditions was explored using carbon replicas and transmission electron microscopy (TEM) observations. The corresponding sizes of MX carbonitrides were measured. The results demonstrates that MX carbonitrides precipitate in type 347H austenitic steel as Nb(C,N). The coarsening rate constant is time-independent; however, an increase in aging temperature results in an increase in coarsening rate of Nb(C,N). The coarsening process was analyzed according to the calculated diffusion activation energy of Nb(C,N). When the aging temperature was 800–900°C, the mean activation energy was 294 kJ·mol?1, and the coarsening behavior was controlled primarily by the diffusion of Nb atoms.

Abstract: This paper,using Huygens' equivalent principle,study the UWB rectangular aerial transient state both in time -domain and frequently-domain and the arithmetic formula of the energetic diffusion on the axis.In the end,the 2D simulation is utilized to prove the relation among spread characteristic in the free space of the encouraging pulse and aerial requirements geometry,the form,main radiation,and the radiation wave form.And then we can get that the radiated waveform on the axis has nothing to do with the shape of the aperture,but in direct proportion to the size of the aperture.This research provided a theoretical basis for designing the radiated property of UWB aperture antennas and helping engineers to design UWB electromagnetic radiation.
Stanniocalcin-1 promotes tumor angiogenesis through up-regulation of VEGF in gastric cancer cells

Abstract: BGC/STC cells (high-expression of STC-1) and BGC/shSTC cells (low- expression of STC-1) were constructed to investigate the effect of STC-1 on the xenograft tumor growth and angiogenesis in vitro and in vivo. ELISA assay was used to detect the expression of vascular endothelial growth factor (VEGF) in the supernatants. Neutralizing antibody was used to inhibit VEGF expression in supernatants. The expression of phosphorylated -PKCβII, phosphorylated -ERK1/2 and phosphorylated -P38 in the BGC treated with STC-1protein was detected by western blot.STC-1 could promote angiogenesis in vitro and in vivo, and the angiogenesis was consistent with VEGF expression in vitro. Inhibition of VEGF expression in supernatants with neutralizing antibody markedly abolished angiogenesis induced by STC-1 in vitro. The process of STC-1-regulated VEGF expression was mediated via PKCβII and ERK1/2.STC-1 promotes the expression of VEGF depended on the activation of PKCβII and ERK1/2 pathways. VEGF subsequently enhances tumor angiogenesis which in turn promotes the gastric tumor growth.Development of gastric cancer involves multiple factor changes that lead to the transformation of human gastric epithelial cells to gastric cancer cells [1]. Angiogenesis is a critical hallmark of malignancy and can occur at different stages of the tumor progression [2]. Acquisition of the angiogenic phenotype can result from genetic changes or local environmental changes such as the secretion of pro-angiogenic growth factors by tumor that lead to the activation of endothelial cells. Stanniocalcin-1(STC-1) is a glycoprotein hormone originally discovered in the corpuscles of Stannius of bony fish [3]. Effect of cold rolling and first precipitates on the coarsening behavior of γ″-phases in Inconel 718 alloy

Abstract: The coarsening behaviors of γ″-phase particles in Inconel 718 alloy aged at 750, 800, and 850°C were investigated by scanning electron microscopy (SEM). Detailed observations and quantitative measurements were conducted to characterize the coarsening behavior of the γ?-phase under various aging conditions. The experimental results indicate that the existence of the δ-phase retards the formation and coarsening of the γ″-phase, without influencing its final particle size or amount. Moreover, when cold rolled with a reduction of 50%, the dimensions of the γ″ particles in Inconel 718 alloy decrease with increasing aging time. Furthermore, the coarsening behavior of the γ″-phase in the Inconel 718 alloy after a normal aging treatment (sample A) and that of the primary δ-phase (sample B) follow the Lifshitz–Slyozov–Wagner (LSW) diffusion-controlled growth theory; the thus-obtained activation energies for the γ″-phase are 292 kJ·mol-1 and 302 kJ·mol-1, respectively.
Study of expression of Pneumococcal surface protein and Immunogenicity

Abstract: The specific fragment of Pneumococcal surface protein A( PspA) and Pneumococcal Surface Adhesin A(PsaA) gene was amplified by PCR from Streptococcus pneumonia 5. The amplified fragnent of PspA and PsaA gene was ligated into pET- 27b (+) vector and transformed into BL 21 E.coli for expression and obtain the expressive production of PspA and PsaA. Induced by IPTG, the expression level was as high as 75 % of total protein. The result showed that the recombinant plasmid could express a specific 75 kDa and 37 kDa fusion protein in E. coli BL 21, which showed the good immunogenicity and a broadly cross reactivity with the other serotypes.
