Development of a drtqPCR Assay for Differentiation of PCV-2a and 2b in Shanghai Area  [PDF]

Abstract: A dual real-time quantitative polymerase chain reaction assay (drtqPCR) was established to detect and differentiate between Porcine circovirus-2a (PCV-2a) and Porcine circovirus-2b (PCV-2b). Genotype-specific primer sets and probes were designed by using sequence data published for different PCV-2 strains. Specificity and sensitivity of the drtqPCR were examined by using PCV-2 isolates with known genotype. Among 367 tissue samples, 44.69% (164/367) were PCV-2 positive. From 164 PCV-2 positive samples, 10.98% (18/164), 92.56% (137/164), and 3.31% (9/164) were positive for PCV-2a, PCV-2b, and both genotypes, respectively. These results suggest that the dif-ferential drtqPCR can be used to detect PCV-2 and to differentiate the 2 genotypes from field sam-ples. The PCV-2 infection is quite common in swine of Shanghai area. Furthermore, the PCV-2b infective ratio is far higher than PCV-2a, and PCV-2a/2b mixed infections are also observed but at a lower prevalence in Shanghai area.

Abstract: Gross errors make skewness influence on mathematical expectation of series,which will transfer to the residual sequence.The gross errors detection only with 3σ criterion may make a mistake of discarding the truth or accepting the wrong.The relationship between the poor of observations or error correction and gross errors was analyzed and the formulae to judge gross errors were deduced.The judgment condition matrix based on bio-poor for detecting gross errors was constructed and the difference of small sample with large sample in detecting gross errors was compared.This method is verified effective through example analyses.

Abstract: The coordinate transformation between different coordinate systems includes four parameters: two translations,one rotation,and one scale paramete.When there is ill-condition transformation matrix,the common points' little errors may cause big fluctuation of parameters' results.Considering the unity between coordinate transformation model and centre of gravity datum of free net adjustment,the two translation parameters may be elimineted under auxiliary condition.This method is validated with an example.
Regional differences of vulnerability of food security in China

Abstract: In view of food affordability and the threshold for food security, this paper has established an integrated index for assessing the vulnerability of food security in China, which is composed mainly of the balance between food supply and demand, the reserve for food security and the economic capacity for offset food demand. Six types of food security regions are identified based on the data from county-level statistics. At regional scale, China’s food security is not optimistic. Under normal conditions without the emergence of extreme disasters and decline of grain-sown areas, China’s most vulnerable areas (Type VI) account for 30.3% of the total number of counties (cities), which are unable to meet the ends by food productivity or market based measures at subsistence levels. In China, there are only 14.5% of the counties (cities) that could guarantee well-off food security through grain production (Type I) or economic measures to meet the demand by themselves (Type III). According to the different vulnerable levels of food security and its dominant forming factors, vulnerable regions of food security in China could be classified into three categories: (1) Vulnerable regions of food security dominated by natural factors (including Type IV and Type VIa), which account for 39.4% of the total number of counties (cities), mainly located in fragile ecologic zones, i.e., farming-grazing transitional zones in the marginal areas of summer monsoon, the poor hilly areas in southern China and so on; (2) Vulnerable regions of food security dominated by low ratio of grain-sown areas (including Type VIb and Type V), which account for 16.7% of the total number of counties (cities), mostly located in the developed areas in the eastern coast of China; (3) Potential vulnerable regions of food security with underdeveloped local economies (Type II), of which 57% are the main grain-surplus regions in China, mainly located in the areas of plains and basins with favorable climate.
3-Port incisionless laparoscopic surgery for rectal cancer with a transrectal assistance  [PDF]

Abstract: Introduction: To present the initial experience of 3-port incisionless laparoscopic surgery for rectal cancer with a transrectal assistance by using a toothed oval clamp. Case Presentation: One patient received 3-port incisionless laparoscopic surgery for rectal cancer with a transrectal assistance by using a toothed oval clamp. Better direct vision and exposure could be acquired for performing laparoscopic surgical procedure, avoiding additional port inserted. Using this procedure, with strictly adhering to the principles of laparoscopic colectomy and oncological procedure, along with the specimen exteriorized via recta, transacted and a stapled anastomosis performed, no incision can be achieved at the end of an operation. The operative time was 180 minutes. The estimated blood loss in the course of an operation was 80 ml. The patient recovered quickly after surgery, with no post-operative pain and no incision. The patient was dischanged home on the 6th postoperative day. Conclusions: With a transrectal assistance by using a toothed oval clamp, 3-port laparoscopic surgery for rectal cancer could be achieved without no incision at the end of the operation, the same as NOTES. It is enormously advantageous to the patient and suitable for application in developing countries, especially in a rural area.

First-principles theory of coloration of WO$_3$ upon charge insertion

Abstract: We report first-principles investigations of the coloration of WO$_3$ upon charge insertion, using sodium tungsten bronze (Na$_x$WO$_3$) as a model system. Our results explain well the systematic color change of Na$_x$WO$_3$ from dark blue to violet, red-orange, and finally to golden-yellow as sodium concentration $x$ increases from 0.3 to unity. Proper accounts for both the interband and the intraband contributions to the optical response are found to be very important for a detailed understanding of the coloration mechanism in this system.

Abstract: In this brief report we consider couplings of the dark energy scalar, such as Quintessence to the neutrinos and discuss its implications in studies on the neutrino mass limits from Baryogenesis. During the evolution of the dark energy scalar, the neutrino masses vary, consequently the bounds on the neutrino masses we have here differ from those obtained before.
Hole-lattice Coupling and Photo-induced Insulator-Metal Transition in VO$_2$

Abstract: Photo-induced insulator-metal transition in VO$_2$ and the related transient and multi-timescale structural dynamics upon photoexcitation are explained within a unified framework. Holes created by photoexcitation weaken the V-V bonds and eventually break V-V dimers in the M$_1$ phase of VO$_2$ when the laser fluence reaches a critical value. The breaking of the V-V bonds in turn leads to an immediate electronic phase transition from an insulating to a metallic state while the crystal lattice remains monoclinic in shape. The coupling between excited electrons and the 6.0 THz phonon mode is found to be responsible for the observed zig-zag motion of V atoms upon photoexcitation and is consistent with coherent phonon experiments.
Innate Immune Response of Human Plasmacytoid Dendritic Cells to Poxvirus Infection Is Subverted by Vaccinia E3 via Its Z-DNA/RNA Binding Domain

Abstract: Plasmacytoid dendritic cells (pDCs) play important roles in antiviral innate immunity by producing type I interferon (IFN). In this study, we assess the immune responses of primary human pDCs to two poxviruses, vaccinia and myxoma virus. Vaccinia, an orthopoxvirus, was used for immunization against smallpox, a contagious human disease with high mortality. Myxoma virus, a Leporipoxvirus, causes lethal disease in rabbits, but is non-pathogenic in humans. We report that myxoma virus infection of human pDCs induces IFN-α and TNF production, whereas vaccinia infection does not. Co-infection of pDCs with myxoma virus plus vaccinia blocks myxoma induction effects. We find that heat-inactivated vaccinia (Heat-VAC; by incubating the virus at 55°C for 1?h) gains the ability to induce IFN-α and TNF in primary human pDCs. Induction of IFN-α in pDCs by myxoma virus or Heat-VAC is blocked by chloroquine, which inhibits endosomal acidification required for TLR7/9 signaling, and by inhibitors of cellular kinases PI3K and Akt. Using purified pDCs from genetic knockout mice, we demonstrate that Heat-VAC-induced type I IFN production in pDCs requires the endosomal RNA sensor TLR7 and its adaptor MyD88, transcription factor IRF7 and the type I IFN feedback loop mediated by IFNAR1. These results indicate that (i) vaccinia virus, but not myxoma virus, expresses inhibitor(s) of the poxvirus sensing pathway(s) in pDCs; and (ii) Heat-VAC infection fails to produce inhibitor(s) but rather produces novel activator(s), likely viral RNA transcripts that are sensed by the TLR7/MyD88 pathway. Using vaccinia gene deletion mutants, we show that the Z-DNA/RNA binding domain at the N-terminus of the vaccinia immunomodulatory E3 protein is an antagonist of the innate immune response of human pDCs to poxvirus infection and TLR agonists. The myxoma virus ortholog of vaccinia E3 (M029) lacks the N-terminal Z-DNA/RNA binding domain, which might contribute to the immunostimulating properties of myxoma virus.
Synthesis of novel temperature/pH responsive polymer via oxyanionic polymerization

Abstract: Hyperbranched poly(3-ethyl-3-oxetanemethanol)-graft-poly (2-dimethylaminoethyl methacrylate) (HP-g-DMA) with a three-demensional structure was synthesized via oxyanionic polymerization. The hydroxyl groups of hyperbranched poly(3-ethyl-3-oxetanemethanol) (HP) reacted with KH and conversed into potassium alcoholate macroinitiators with high initiating efficiencies. High monomer conversion (>95%) was obtained and no residual macroinitiators or monomer was observed. UV-visible spectra indicate that the aqueous solution of the HP-g-DMA exhibited the lowermost critical solution temperature (LCST). The LCST was influenced by the chain length of DMA and pH condition of solution. It is found that LCST decreased with increasing DMA chain length or increasing pH value of solution.
