Acute Heat-Evoked Temperature Sensation Is Impaired but Not Abolished in Mice Lacking TRPV1 and TRPV3 Channels

Abstract: The discovery of heat-sensitive Transient Receptor Potential Vanilloid ion channels (ThermoTRPVs) greatly advanced our molecular understanding of acute and injury-evoked heat temperature sensation. ThermoTRPV channels are activated by partially overlapping temperatures ranging from warm to supra-threshold noxious heat. TRPV1 is activated by noxious heat temperature whereas TRPV3 can be activated by warm as well as noxious heat temperatures. Loss-of-function studies in single TRPV1 and TRPV3 knock-out mice have shown that heat temperature sensation is not completely abolished suggesting functional redundancies among these two channels and highlighting the need of a detailed analysis of TRPV1::TRPV3 double knock-out mice (V1V3dKO) which is hampered by the close proximity of the loci expressing the two channels. Here we describe the generation of a novel mouse model in which trpv1 and trpv3 genes have been inactivated using bacterial artificial chromosome (BAC)-based homologous recombination in embryonic stem cells. In these mice, using classical thermosensory tests such hot plate, tail flick and the thermotaxis gradient paradigms, we confirm that TRPV1 is the master channel for sensing noxious heat temperatures and identify a cooperative role of TRPV1 and TRPV3 for sensing a well-defined window of acute moderate heat temperature. Using the dynamic hot plate assay, we unravel an intriguing and unexpected pronounced escape behavior in TRPV1 knock-out mice that was attenuated in the V1V3dKO. Together, and in agreement with the temperature activation overlap between TRPV1 and TRPV3 channels, our data provide in vivo evidence of a cooperative role between skin-derived TRPV3 and primary sensory neurons-enriched TRPV1 in modulation of moderate and noxious heat temperature sensation and suggest that other mechanisms are required for heat temperature sensation.
Uncoupling of Molecular Maturation from Peripheral Target Innervation in Nociceptors Expressing a Chimeric TrkA/TrkC Receptor

Abstract: Neurotrophins and their receptors control a number of cellular processes, such as survival, gene expression and axonal growth, by activating multiple signalling pathways in peripheral neurons. Whether each of these pathways controls a distinct developmental process remains unknown. Here we describe a novel knock-in mouse model expressing a chimeric TrkA/TrkC (TrkAC) receptor from TrkA locus. In these mice, prospective nociceptors survived, segregated into appropriate peptidergic and nonpeptidergic subsets, projected normally to distinct laminae of the dorsal spinal cord, but displayed aberrant peripheral target innervation. This study provides the first in vivo evidence that intracellular parts of different Trk receptors are interchangeable to promote survival and maturation of nociceptors and shows that these developmental processes can be uncoupled from peripheral target innervation. Moreover, adult homozygous TrkAC knock-in mice displayed severe deficits in acute and tissue injury-induced pain, representing the first viable adult Trk mouse mutant with a pain phenotype.
Human Face Recognition, Information Processing and Social Behavior in Children with Autism  [PDF]

The aim of this study was to investigate whether children with high-functioning autism (HFA) and Asperger’s syndrome (AS) can be differentiated from each other and from typically developing children in ability to recognize a human face. The present study included 69 participants: children with autism (high-functioning autism or Asperger’s syndrome, n = 15 per group) and a control group of 39 typically developing children. It involved a face recognition task included in the NEPSY scale. Results showed that the autism groups performed poorer than the typically developing group, but no difference was found between high-functioning and Asperger groups. Both the information processing peculiarities of children with autism and their (interactional and communicative) social impairments could explain their difficulty to recognize human faces.

Abstract: In 2010, the World Health Organization supported the Lebanese Ministry of Education and Higher Education to establish a network of 10 Health Promoting Schools (HPS). This study was undertaken to address the extent to which the HPS model was able to produce changes in the risk behaviors of adolescents (Smoking, Drinking, and Drug Use) and prepare them to respond to evolving health challenges. A cross-sectional survey was carried out during 2011-2012 and compared Grade 6 - 9 students in HPS, and 10 other public and private schools. The Youth Risk Behavior Survey [1], and Carbon Monoxide measurements in adolescent breaths were used for data collection. Findings revealed that the current School Health Program failed to address issues of concern to adolescents with no significant differences in risk behaviors: smoking, alcohol and drug use. The HPS network needs reassessment to upgrade the outcomes of health education curricula. Expected learning outcomes and healthy practices must be designed to match students’ age, grade level, and developmental milestones.
Patients' perspectives on high-tech home care: a qualitative inquiry into the user-friendliness of four technologies

Abstract: Four home care interventions that are frequently used and vary in their technical and clinical features were selected: Antibiotic intravenous therapy, parenteral nutrition, peritoneal dialysis and oxygen therapy. Our qualitative study relied on the triangulation of three sources of data: 1) interviews with patients (n = 16); 2) interviews with carers (n = 6); and 3) direct observation of nursing visits of a different set of patients (n = 16). Participants of varying socioeconomic status were recruited through primary care organizations and hospitals that deliver home care within 100 km of Montreal, the largest urban area in the province of Quebec, Canada.The four interventions have both a negative and positive effect on patients' lives. These technologies were rarely perceived as user-friendly, and user-acceptance was closely linked to user-competence. Compared with acute I.V. patients, who tended to be passive, chronic patients seemed keener to master technical aspects. While some of the technical and human barriers were managed well in the home setting, engaging in the social world was more problematic. Most patients found it difficult to maintain a regular job because of the high frequency of treatment, while some carers found their autonomy and social lives restricted. Patients also tended to withdraw from social activities because of social stigmatization and technical barriers.While technology contributes to improving the patients' health, it also imposes significant constraints on their lives. Policies aimed at developing home care must clearly integrate principles and resources supporting the appropriate use of technology. Close monitoring of patients should be part of all technology-enhanced home care programs.The possibility of managing patients outside of the hospital has rarely been so widely considered. Abstract: even in the most precarious situations, the responses to the economic recessions are not limited to the practical application of survival resolution. these answers generate imaginaries, moral representations and prior experiences that give sense to the events and organize strategies. taking this fact into account, this paper proposes to analyze the idealistic aspects of the economic global crisis through the experiences of the cooperative workers in the mines of bolivia. their logic of interpretation is far from those proposed by the salary and the welfare state. thus, the crisis appears as an ideal heuristic situation to understand market and labour representations of the social actors. as a matter of fact, we can observe that for the bolivian miners the breakdown of the miner's market was less traumatic that the previously unseen prosperity. this assessment reminds us that the sudden excessive accumulation of wealth can be a crisis equal to that experienced by the economical recession.

Abstract: the psychodynamics of work is defined as the analysis of psychic processes mobilized by the gathering of a subject and the work organization constraints. in the 80s, the researches conducted in male workplace environments allowed to uncover the existence of collective defense strategies against suffering at the work. as from 1988, an important interdisciplinary work with work sociologies was launched. it is concerned with knowing whether the psychodynamics work was heuristic to equally analyze the situations of women at work and to reexamine the preexisting knowledge on men suffering in a sexualized problematic framework. the paper reports the main determinants which led to the establishment of sex social relationships and its challenge, the sexual division of work, a central dimension of the clinic research, the theory and methodology in workplace psychodynamics. an important issue in the paper is the situation of nurses, from which is constituted the main knowledge on the relations between health and work on the women's side.

Abstract: Bien que l’iconographie wordsworthienne s’attache à représenter le poète en promeneur solitaire, les paysages qu’il retrace lui-même dans le Prélude de 1805 sont fort peuplés. De multiples personnages — qu’il faudra bien appeler figures — animent ses paysages. Ils les animent au sens propre du terme : c’est-à-dire qu’ils conduisent le mouvement dont les paysages wordsworthiens sont à la fois le produit et la scène. Ces figures, loin de faire de la figuration, donc, configurent, dans un jeu typiquement wordsworthien d’activité passive, la nouvelle vision du monde qui se trame dans ces paysages. Ils effectuent et relayent les principes de la poétique wordsworthienne — combinaison, rayonnement, exaltation — par laquelle “la révolution copernicienne” du romantisme a “lieu”.
