BLMH and APOE genes in Alzheimer Disease: A possible relation  [PDF]

Abstract: Alzheimer disease (AD) is a progressive and irreversible neurodegenerative disorder that is characterized by cognitive decline, memory loss and confusion. The E4 allele of the apolipoprotein E gene (APOE) is associated with AD and it is the main genetic risk factor for disease. Although the exact physiological function is unknown, bleomycin hydrolase (BLMH) may also be associated with AD development, although previous immunohistochemical findings havebeen inconsistent. Therefore, the purpose ofthis study was to evaluate the genotypic and allele frequencies of theAPOEgene andBLMH1450 G> A polymorphism and assessBLMHexpression using PCR-RFLP and RT-qPCR analyses ofblood samples from patients with Alzheimer disease (AD), healthy elderly adults (EC) andhealthyyoung subjects(YC). BLMHexpression wassignificantly different among groups (p= 0.015) and there was substantial reduction with age and with AD. TheAPOEandBLMHgenotype frequency did not diverge from the Hardy-Weinberg equilibrium. There was a higher frequency of genotype 3/3 inall subjects (61.1%) and the AD group demonstrated a higher frequency of allele 4; however, differences ingenotype and allele distributions were statistically different among groups.

A study on the action of two calcium channel blockers (verapamil and flunarizine) upon an experimental model of tardive dyskinesia in rats

Abstract: tardive dyskinesia (td), a serious complications of neuroleptic chronic use, has no effective therapy yet. we performed an experiment to study the action on td, of the calcium channel blockers (ccb) drugs, verapamil and flunarizine. we obtained the td model in rats, administering haloperidol for a 21-day period. after this, the stereotyped movement induced by apomorphyne was rated. the ccb drugs were administered in acute (in the 28th. day) and chronic (for 8 days, after the 25th day) experiments. acutely, verapamil increased the stereotyped behaviour, and promoted a reduction of it in the chronic experiment. the results suggest that ccb drugs should be tested in clinical trials of td.

Abstract: the cognitive subscale of the "alzheimer's disease assessment scale" (adas-cog) is widely used for the evaluation of dementia and is very popular in dementia drug trials because of the characteristics of the scale. the objective of the present study was to adapt the adas-cog for use for the brazilian population. a major problem is that education is variable, a fact that may influence performance in cognitive evaluation. this study was conducted on a control group (cg) of 96 subjects (25 males and 71 females aged 68 ± 8.6 years) and on 44 alzheimer's disease (ad) patients (19 males and 25 females aged 72 ± 6.4 years) with mild dementia (clinical dementia rating score 1). for statistical analysis groups were divided into three educational levels: i, 0-4 years of schooling (65 cg/20 ad); ii, 5-11 years of schooling (19 cg/7 ad), and iii, 12 or more years of schooling (12 cg/17 ad). the battery was applied according to original instructions. total mean score for cg and ad was 10.9 and 22.9 for level i, 7.8 and 22.4 for level ii, and 6.2 and 15.4 for level iii, respectively. these results indicate that our version of the adas-cog is useful to identify mild dementia, though there may be an overlapping when comparing high education demented with low education non-demented subjects.

Abstract: this study aimed to evaluate the effects of organic fertilization at planting and dressing on the aboveground part dry biomass and on the content and yield of essential oil of mint (mentha arvensis). the experiment consisted of two stages (harvests). a randomized complete block design was used, with three replications. at the first stage, five different cattle manure dosages (0, 2.5, 5.0, 7.5 and 10.0 kg m-2) applied to the plots at planting, were evaluated and the first harvest was carried out 120 days later. at the second stage after the first harvest, the plots were subdivided in four split-plot and four cattle manure dosages (0, 2.5, 5.0 and 7.5 kg m-2) were applied at dressing, following a split-plot design. the second harvest was carried out 120 days after the first one. the fresh biomass was dried in the oven at 37oc and the essential oil was extracted in a clevenger apparatus. aboveground part dry biomass (g), essential oil content (w/w %) and essential oil yield were determined. the levels of cattle manure applied at planting and dressing affected in a positive linear way the aboveground part dry biomass production and the essential oil yield. essential oil content was not affected by different organic manure levels.

Abstract: a case of acute methanol intoxication is presented, in which bilateral putaminal hemorrhage developed after hemodialysis. even though the patient was initially comatose and profoundly acidotic, favorable outcome was achieved, with long-term neurologic impairments essentially restricted to mild crural paraparesis, retrograde amnesia, and marked visual deficit. a comparative literature review is evaluated.

Abstract: hyptis marrubioides epling ("hortel?-do-campo") is a medicinal species known for its activities against gastrointestinal and skin infections, pains and cramps. the aim of this work was to evaluate essential oil content and chemical composition in h. marrubioides of different genotypes (purple and white), as well as fresh plant parts (leaves, inflorescences and stem). the studied genotypes were identified based on the inflorescences, which are purple and white. the experimental design was completely randomized, including the three parts of both genotypes, in a 3x2 factorial arrangement, with four replicates. essential oil was extracted through hydrodistillation for two hours and analyzed using gas chromatography attached to a mass spectrometry (gc-ms). the highest essential oil content was found for the inflorescence of purple genotype. hyptis marrubioides essential oil composition quantitatively varied among the analyzed plant parts and between genotypes. the compound α-thujone had the highest percentage of apex area in inflorescences of purple genotype. the percentages of purple genotype were on average higher than those of white one.

Abstract: a 45 years-old woman presented with loss of iniciative and memory, imotivated ciy and behaviour disturbance with childish traits, quickly progressive along 6 months until total apathy. an eeg showed periodic activity with bilateral triphasic waves against a flattened background activity suggesting creutzfeldt-jacob disease (cjd), but investigation for treatable causes of dementia disclosed hypercalcemia and hypophosphatemia. further investigation showed a mass at the thyroid region that at surgery was identified as an oxyphilic ceils adenoma. with electrolyte disturbance correction and posterior surgery there was normalization of both eeg and clinical status. though it was previously reported mental confusion and eeg alteration associated with hyperparathyroidism we do not know of any previous case o>: confusion associated with periodic activity in eeg as in this disease. hyperparathyroidism should be a differential diagnosis in every ?de novo? case of cjd.

Abstract: given increasing research into new therapies for dementia, especially for alzheimer disease, it has become clear that traditional methods of evaluation, centered on cognition, have proved insufficient. thus, scales for behavioral disturbances and activities of daily living have been added. nevertheless, some observations of clinical significant changes, as reported by caregivers, could be overlooked. clinician's impression of global change scales (cigic) are based on broader information and may detect more subtle changes. typically they take into consideration caregiver information, that which is seen by the health professional during the interview, and may also include brief cognitive tests. we present a brazilian portuguese version of a widely used scale of this type, specifically designed for alzheimer disease (adcs-cgic), being the result of the recommended method of translation, back-translation and version by panel consensus.

Abstract: Com o crescente volume de pesquisas sobre novas terapias para as demências, em particular a doen a de Alzheimer, tornou-se evidente que a avalia o mais tradicional, centrada nas altera es da cogni o, n o seria suficiente para detectar algumas das altera es significativas que novos tratamentos podem trazer. Deste modo, foram acrescentadas escalas para avaliar altera es do comportamento e da dependência funcional. Ainda assim, restavam mudan as, percebidas pelos familiares, que poderiam n o ser captadas por essas escalas. As escalas de impress o clínica de mudan a (CIGIC) baseiam-se nas informa es fornecidas pelo informante, no que é observado durante a consulta, e, se necessário, em breves testes de cogni o. Apresentamos a vers o de uma escala deste tipo, largamente utilizada, desenvolvida para a doen a de Alzheimer (ADCS-CIGIC), que foi obtida pelo método recomendado de tradu o, retro-tradu o e vers o final por consenso.
