Lactobacillus reuteri Maintains a Functional Mucosal Barrier during DSS Treatment Despite Mucus Layer Dysfunction

Abstract: Treatment with the probiotic bacterium Lactobacillus reuteri has been shown to prevent dextran sodium sulfate (DSS)-induced colitis in rats. This is partly due to reduced P-selectin-dependent leukocyte- and platelet-endothelial cell interactions, however, the mechanism behind this protective effect is still unknown. In the present study a combination of culture dependent and molecular based T-RFLP profiling was used to investigate the influence of L. reuteri on the colonic mucosal barrier of DSS treated rats. It was first demonstrated that the two colonic mucus layers of control animals had different bacterial community composition and that fewer bacteria resided in the firmly adherent layer. During DSS induced colitis, the number of bacteria in the inner firmly adherent mucus layer increased and bacterial composition of the two layers no longer differed. In addition, induction of colitis dramatically altered the microbial composition in both firmly and loosely adherent mucus layers. Despite protecting against colitis, treatment with L. reuteri did not improve the integrity of the mucus layer or prevent distortion of the mucus microbiota caused by DSS. However, L. reuteri decreased the bacterial translocation from the intestine to mesenteric lymph nodes during DSS treatment, which might be an important part of the mechanisms by which L. reuteri ameliorates DSS induced colitis.
iNOS-Dependent Increase in Colonic Mucus Thickness in DSS-Colitic Rats

Abstract: Aim To investigate colonic mucus thickness in vivo in health and during experimental inflammatory bowel disease. Methods Colitis was induced with 5% DSS in drinking water for 8 days prior to experiment, when the descending colonic mucosa of anesthetized rats was studied using intravital microscopy. Mucus thickness was measured with micropipettes attached to a micromanipulator. To assess the contributions of NOS and prostaglandins in the regulation of colonic mucus thickness, the non-selective NOS-inhibitor L-NNA (10 mg/kg bolus followed by 3 mg/kg/h), the selective iNOS-inhibitor L-NIL (10 mg/kg bolus followed by 3 mg/kg/h) and the non-selective COX-inhibitor diclofenac (5 mg/kg) were administered intravenously prior to experiment. To further investigate the role of iNOS in the regulation of colonic mucus thickness, iNOS ?/? mice were used. Results Colitic rats had a thicker firmly adherent mucus layer following 8 days of DSS treatment than untreated rats (88±2 μm vs 76±1 μm). During induction of colitis, the thickness of the colonic mucus layer initially decreased but was from day 3 significantly thicker than in untreated rats. Diclofenac reduced the mucus thickness similarly in colitic and untreated rats (?16±5 μm vs ?14±2 μm). While L-NNA had no effect on colonic mucus thickness in DSS or untreated controls (+3±2 μm vs +3±1 μm), L-NIL reduced the mucus thickness significantly more in colitic rats than in controls (?33±4 μm vs ?10±3 μm). The importance of iNOS in regulating the colonic mucus thickness was confirmed in iNOS?/? mice, which had thinner colonic mucus than wild-type mice (35±3 μm vs 50±2 μm, respectively). Furthermore, immunohistochemistry revealed increased levels of iNOS in the colonic surface epithelium following DSS treatment. Conclusion Both prostaglandins and nitric oxide regulate basal colonic mucus thickness. During onset of colitis, the thickness of the mucus layer is initially reduced followed by an iNOS mediated increase.

Abstract: i believe that john locke's defense of the right to punish is in tension with his defense of human equality. the main purpose of this article is to show that this tension could be resolved if locke's justification of the right to punish is based on the concept of trust. this concept is clarley employed by locke to justify the right of resistance. i think that it is possible to use it also to justify the existence of the right to punish. but, even if this justification were succesful, some problems remain.

Abstract: This article investigates web-based user education, considering it as a forum where the shaping of librarians’ professional expertise is negotiated. By conveying a profession’s knowledge on the Web, it is possible to define and emphasise those “problems” that the profession is striving to solve. The aim of the article is to present various approaches to the expertise of university librarians as conveyed by the librarians’ pedagogical working methods. The empirical basis for the study is an analysis of 31 web-based guides to information seeking, available at Nordic higher education and research libraries.The content of the web-based user education is presented and discussed in terms of twelve recurrent themes. Then, four approaches to user education are characterised: a source-oriented approach, a behaviour-oriented approach, a process-oriented approach and a communication-oriented approach. The approaches convey different definitions of important concepts such as information, information seeking, and user, and a point of departure for this article is that these approaches have practical consequences for the user education.

Abstract: The present article is an analytical review of studies about user education (in both traditional and web-based form)aimed primarily at college and university students, and of studies about professionals' information seeking. A theoretical framework discussing the concepts of information, profession and professional identity form a critical background against which the mainly empirically oriented studies are presented. Librarians and nurses, two occupational groups with important similarities and differences, are used as examples of user groups in the study. A point of departure for this article is that user education and information seeking should be understood as socio-cultural practices. Thus, the symbolic value of professional information and its potential to create, sustain and alter power relations, identities and interests is underlined. The analysis of previous studies concludes in a number of research questions that are addressed in a presently ongoing research project about ICT-based information seeking in the transition from educational to occupational practice. The project aims to uncover how norms, values and expectations about information seeking and information literacy are formed within occupational groups, are mediated through user education and are appropriated or mastered by students for use in their working life.

Abstract: Since at least three decades, the concept of equality —in particular, distributive equality— has been at the centre of the contemporary discussion on distributive justice. The idea that underserved inequalities should be eliminated and that deserved inequalities should be preserved is one of the most relevant ways in which the ideal of equality has been recently defended. The main scope of this article is to argue against the intrinsic value of equality thus understood by pointing to an important confusion: the one between the value of equality and the value of people having what they deserve. Desde hace al menos tres décadas el concepto de igualdad —en particular, el concepto de igualdad distributiva— ha estado en el centro de la discusión contemporánea sobre justicia distributiva. La idea que las desigualdades inmerecidas deberían ser eliminadas y que las desigualdades merecidas deberían ser preservadas es una de las maneras más relevantes a través de la cual el ideal de la igualdad ha sido recientemente defendido. El objetivo principal de este trabajo es argumentar contra el valor intrínseco de la igualdad entendida de esta manera apuntando a una importante confusión: aquélla entre el valor de la igualdad y el valor de que las personas tengan lo que merecen.
Calibration Systems of the ATLAS Tile Calorimeter

Abstract: TileCal is the hadronic calorimeter covering the most central region of the ATLAS experiment at the LHC. This sampling calorimeter uses iron plates as absorber and plastic scintillating tiles as the active material. A multi-faceted calibration system allows to monitor and equalize the calorimeter response at each stage of the signal production, from scintillation light to digitization. This calibration system is based on signal generation from different sources: a Cs radioactive source, laser light, charge injection and minimum bias events produced in proton-proton collisions. A brief description of the different TileCal calibration systems is given and the latest results on their performance in terms of calibration factors, linearity and stability are presented.
