Trypanosoma cruzi Epimastigotes Are Able to Store and Mobilize High Amounts of Cholesterol in Reservosome Lipid Inclusions

Abstract: Background Reservosomes are lysosome-related organelles found in Trypanosoma cruzi epimastigotes. They represent the last step in epimastigote endocytic route, accumulating a set of proteins and enzymes related to protein digestion and lipid metabolism. The reservosome matrix contains planar membranes, vesicles and lipid inclusions. Some of the latter may assume rectangular or sword-shaped crystalloid forms surrounded by a phospholipid monolayer, resembling the cholesterol crystals in foam cells. Methodology/Principal Findings Using Nile Red fluorimetry and fluorescence microscopy, as well as electron microscopy, we have established a direct correlation between serum concentration in culture medium and the presence of crystalloid lipid inclusions. Starting from a reservosome purified fraction, we have developed a fractionation protocol to isolate lipid inclusions. Gas-chromatography mass-spectrometry (GC-MS) analysis revealed that lipid inclusions are composed mainly by cholesterol and cholesterol esters. Moreover, when the parasites with crystalloid lipid-loaded reservosomes were maintained in serum free medium for 48 hours the inclusions disappeared almost completely, including the sword shaped ones. Conclusions/Significance Taken together, our results suggest that epimastigote forms of T. cruzi store high amounts of neutral lipids from extracellular medium, mostly cholesterol or cholesterol esters inside reservosomes. Interestingly, the parasites are able to disassemble the reservosome cholesterol crystalloid inclusions when submitted to serum starvation.

Abstract: How does one set about helping children and teenagers to understand the different musical styles and genres, the historical context pertaining to works of music and composers, or the historical aesthetic appreciation of the musical repertoire? Are these aims achievable and simple to attain with students? This article intends to focus on this very aspect, deemed important and included in all the national curricula throughout Europe. It will show that the teacher’s familiarity with the specialist musical code could lead him/her to take for granted technical knowledge that the student does not actually possess and that the problems of temporal conceptualization should represent an object of careful reflection for all teachers.
Variability-selected Active Galactic Nuclei in the VST-SUDARE/VOICE Survey of the COSMOS Field

Abstract: Optical variability has proven to be an effective way of detecting AGNs in imaging surveys, lasting from weeks to years. In the present work we test its use as a tool to identify AGNs in the VST multi-epoch survey of the COSMOS field, originally tailored to detect supernova events. We make use of the multi-wavelength data provided by other COSMOS surveys to discuss the reliability of the method and the nature of our AGN candidates. Our selection returns a sample of 83 AGN candidates; based on a number of diagnostics, we conclude that 67 of them are confirmed AGNs (81% purity), 12 are classified as supernovae, while the nature of the remaining 4 is unknown. For the subsample of AGNs with some spectroscopic classification, we find that Type 1 are prevalent (89%) compared to Type 2 AGNs (11%). Overall, our approach is able to retrieve on average 15% of all AGNs in the field identified by means of spectroscopic or X-ray classification, with a strong dependence on the source apparent magnitude. In particular, the completeness for Type 1 AGNs is 25%, while it drops to 6% for Type 2 AGNs. The rest of the X-ray selected AGN population presents on average a larger r.m.s. variability than the bulk of non variable sources, indicating that variability detection for at least some of these objects is prevented only by the photometric accuracy of the data. We show how a longer observing baseline would return a larger sample of AGN candidates. Our results allow us to assess the usefulness of this AGN selection technique in view of future wide-field surveys.

Abstract: One of the most peculiar characteristics of Active Galactic Nuclei (AGN) is their variability over all wavelengths. This property has been used in the past to select AGN samples and is foreseen to be one of the detection techniques applied in future multi-epoch surveys, complementing photometric and spectroscopic methods. In this paper, we aim to construct and characterise an AGN sample using a multi-epoch dataset in the r band from the SUDARE-VOICE survey. Our work makes use of the VST monitoring program of an area surrounding the Chandra Deep Field South to select variable sources. We use data spanning a six month period over an area of 2 square degrees, to identify AGN based on their photometric variability. The selected sample includes 175 AGN candidates with magnitude r < 23 mag. We distinguish different classes of variable sources through their lightcurves, as well as X-ray, spectroscopic, SED, optical and IR information overlapping with our survey. We find that 12% of the sample (21/175) is represented by SN. Of the remaining sources, 4% (6/154) are stars, while 66% (102/154) are likely AGNs based on the available diagnostics. We estimate an upper limit to the contamination of the variability selected AGN sample of about 34%, but we point out that restricting the analysis to the sources with available multi-wavelength ancillary information, the purity of our sample is close to 80% (102 AGN out of 128 non-SN sources with multi-wavelength diagnostics). Our work thus confirms the efficiency of the variability selection method in agreement with our previous work on the COSMOS field; in addition we show that the variability approach is roughly consistent with the infrared selection.

Abstract: This work makes use of the VST observations to select variable sources. We use also the IR photometry, SED fitting and X-ray information where available to confirm the nature of the AGN candidates. The IR data, available over the full survey area, allow to confirm the consistency of the variability selection with the IR color selection method, while the detection of variability may prove useful to detect the presence of an AGN in IR selected starburst galaxies.

Abstract: We explore the geodesic movement on an effective four-dimensional hypersurface that is embedded in a five-dimensional Ricci-flat manifold described by a canonical metric, in order to applying to planetary orbits in our solar system. Some important solutions are given, which provide the standard solutions of general relativity without any extra force component. We study the perihelion advances of Mercury, the Earth and Pluto using the extended theory of general relativity (ETGR). Our results are in very good agreement with observations and show how the foliation is determinant to the value of the perihelion's advances. Possible applications are not limited to these kinds of orbits.
Central syntropic effects elicited by trigeminal proprioceptive equilibrium in Alzheimer’s disease: a case report

Abstract: A 66-year-old Caucasian man, affected by Alzheimer’s disease and with a neuropsychological evaluation issued by the Alzheimer’s Evaluation Unit, underwent an electromyographic investigation of the masseter muscles in order to assess their functional balance. The patient showed a bilateral lack of all inferior molars. The extreme myoelectric asymmetry in dental occlusion suggested the rebalancing of masseter muscular functions through concurrent transcutaneous stimulation of the trigeminal nerve supramandibular and submandibular motor branches. The above-mentioned method allows detection of symmetric craniomandibular muscular relation that can be kept constant through the use of a cusp bite modeled on the inferior dental arch, called orthotic-syntropic bite. A few days later, the patient underwent a new neuropsychological investigation, together with a functional magnetic resonance imaging study, and saccadic, pupillometric video-oculographic examinations in occlusal un-balance and re-balance conditions.Comparative data analysis has shown that a re-balanced occlusal condition can improve a patient’s cognitive-attentive functions. Moreover, the saccadic and pupillometric video-oculographic investigations have proven useful both in analyzing reticulo-cerebellar subcortical systems, prematurely altered in Alzheimer’s disease, and in implementing neurological evaluations.
Cerebro-afferent vessel and pupillary basal diameter variation induced by stomatognathic trigeminal proprioception: a case report

Abstract: Introduction A patient affected by asymmetric hemodynamics of cerebro-afferent vessels underwent duplex color scanner investigations in occlusal proprioceptive un- and rebalance conditions. Pupillometric video-oculographic examinations were performed in order to spot connected trigeminal proprioceptive motor patterns able to interfere on sympathetic autonomic activity. The aim of this case report is to verify if involuntary jaw closing during swallowing, executed in unbalance and rebalance myoelectric activity, would be able to modify cerebral hemodynamics. Case presentation A 56-year-old Caucasian Italian woman affected by asymmetric blood flow of cerebro-afferent vessels underwent an electromyographic investigation of her occlusal muscles in order to assess their occlusal functional balance. The extreme asymmetry of myoelectric activity in dental occlusion evidenced by electromyographic values suggested the rebalancing of the functions of occlusal muscles through concurrent transcutaneous stimulation of the trigeminal nerve supra- and submandibular motor branches. The above-mentioned method allowed the detection of a symmetric craniomandibular muscular relation that can be kept constant through the use of a cusp bite modeled on the inferior dental arch: called orthotic-syntropic bite for its peculiar use of electrostimulation. A few days later, the patient underwent a duplex color scanner investigation and pupillometric video-oculographic examinations in occlusal unbalance and rebalance conditions. Conclusions A comparative data analysis showed that an unbalanced dental occlusal function may represent an interferential pattern on cerebral hemodynamics velocity and pupillometric evaluations have proved useful both in the analysis of locus coeruleus functional modalities and as a diagnostic tool in the assessment of pathologies involving locus coeruleus and autonomic systems. The inclusion of myoelectric masseter examinations can be useful in patients with asymmetric hemodynamics of cerebro-afferent vessels and dental occlusal proprioceptive rebalance can integrate the complex therapy of patients with increased chronic sympathetic activity.
