Variabilidade genética de acessos de pitaya com diferentes níveis de produ??o por meio de marcadores RAPD

Abstract: the most cultivated pitaya species nowadays is hylocereus undatus, red pitaya with white pulp. colombia and mexico are the major world producers and, due to its rusticity, pitaya is considered a potentially viable alternative to make good use of gravel, sandy and compact rocky soils. although the great demand, there is not yet a variety released on the market that attends production climatic needs and brazilian consumer exigencies. the present work is a part of embrapa cerrados pitaya cpac py-01 selection and improvement program. we have had the objective to realize genetic variability study of 16 pitaya accesses maintained at embrapa cerrados germoplasm collection, showing different phenotypic characteristics with special relation to production, through molecular markers rapd (random amplified polymorphic dna). each access genomic dna was extracted and eleven decamer primers indicators were used to obtain molecular markers rapd, that were converted in a binarium data matrix, from where it was estimated genetic distances between accesses and realized grouping analysis and graphic dispersion. one hundred and eleven rapd markers were obtained, making a 10,1 markers medium per primer, in witch 45 (40,54%) were polymorphic. genetic distances between 16 accesses varied within 0,006 and 0,148. the biggest genetic distances were obtained between accesses "52" and "61"; in 2007, the first one produced more than 25 fruits and the second one, none. the smallest genetic distances were verified between accesses "63" and "55" and between 19" and "59". both groups showed close production values. rapd molecular markers showed that, even inside the same species, there is genetic variability between plants with different productions, standing out molecular techniques importance as auxiliary instruments selection breeding programs.
Enraizamento de espécies silvestres de maracujazeiro utilizando cinco doses de ácido indolilbutírico

Abstract: the aim of this work was to evaluate under greenhouse conditions, the rooting potential of different wild species of passion fruit using different doses of indolebutyric acid. herbaceous cuttings of adult plants of passiflora setacea, p. coccinea, p. amethystina, p. edulis, p. edulis x p. setacea and p. coccinea x p. setacea were collected and treated with doses of 0, 250, 500, 750 and 1000 mgl-1 of iba, and then planted in trays with moistened substrate and held under intermittent mist. the experimental design was in randomized blocks with 5 replications and 6 x 5 factorial arrangement (six species and five iba levels) and 6 useful cuttings per experimental unit. the variables observed were rooted living cutting with sprouts (rlcws), rooted living cutting without sprouts (rlcwos), living cutting with callus (lcwc), living cutting without callus (lcwoc), total of rooted cuttings (trc), dead rooted cutting (drc), dead cutting with callus (dcwc), dead cutting without callus (dcwoc), total of dead cuttings (tdc), cutting with sprouds (cws), average cuttings with sprouts (acws), root dry matter (rootdm), sprout dry matter (sprdm),and total of dry matter (totaldm). the p. amethystina species presented better development than other species, showing rooting potential, with 88.67% of cuttings with roots and sprouts. the use of iba was efficient as the doses were increased, providing a good development for cuttings of the studied species.

Abstract: brazil is the largest world producer of passion fruit, however, it has been observed a reduction in the productivity in recent years due, mainly, to phytosanitary factors. at embrapa cerrados, the transfer of resistance genes from wild to commercial species of passion fruit has been made through interspecific hybridations, followed by a backcrossing molecular marker-assisted program. the objective this work was to verify the recovery of recurrent genome at the plants rc4 and rc5 [(passiflora edulis x passiflora setacea) x passiflora edulis] based on rapd markers. the study was developed at embrapa cerrados laboratory of genetics and molecular biology. dna samples of each genetic material (17 rc4 plants, 16 rc5 plants, passiflora edulis and passiflora setacea) were amplified to obtain rapd markers. there were used 12 decamer primers for plants rc4 and 14 decamer primers for plants rc5. the rapd markers generated were converted into a matrix of binary data. there were a high percentage of polymorphic markers as a result of interspecific base crossing. the smallest genetic similarity was observed between species p. edulis and p. setacea, highlighting the large genetic distance of these commercial and wild varieties, respectively.
Genetic variability of Passiflora spp. from commercial fields in the Federal District, Brazil

Abstract: this study aimed to characterize the genetic variability in commercial accessions of passion fruit from the federal district, brazil, by rapd markers. genetic analyses were done with leaf samples of 30 accessions. dna samples were amplified by rapd technique, and respective markers converted into a binary matrix, from which the genetic distances between the accessions were estimated. clustering analyis based on genetic distances allowed to detect a wide range of genetic variabillity among the accessions of sour passion fruit, and to separate them from the two sweet passion fruit. the graphical positioning of 'brs ouro vermelho' confirms its potential to improve the genetic variability of commercial varieties of sour passion fruit. dispersal of genetic distances among commercial accessions of sour passion fruit supports evidence for different genetic origins of the materials planted in the federal district. the verified genetic variability indicates the potential success of future breeding programs for this region.
Produtividade de seis cultivares de maracujazeiro-azedo durante três anos em Vargem Bonita, DF

Abstract: the productivity of sour-passionfruit cultivars was evaluated for the purpose of obtaining productive material adapted to the federal district conditions for consumption in natura. maguari, csb marília, nj3 vermelho, cbs marília x nj3 vermelho, roxo australiano and sele??o df cultivars were evaluated during three production years in a red-yellow latosol soil in the vertical stake system. maguari presented higher seed germination rate than that of the other cultivars. the medium plant height of sele??o df and roxo australiano was superior to that of the other four cultivars. maguari showed the greatest vegetative vigor during the three-year experiment and the mindest number of lost plants due to crop decay. csb marília was the most precocious and the first cultivar to produce fruits. by the first harvest, csb marília, csb marília x nj3 vermelho, roxo australiano and sele??o df were the most yielding. in the second year of harvesting, all six cultivars showed high productivity, and sele??o df surpassed all the other cultivars, with 50 ton/ha. in the third year, in which a considerable decay occurred, maguari presented the maximum productivity. in the three-year of production, the six cultivars showed to be superior in productivity in relation to the results of all other research ever seen in brazil.
Produtividade de seis cultivares de maracujazeiro-azedo durante três anos em Vargem Bonita, DF

Abstract: Avaliou-se a produtividade de cultivares de maracujazeiro-azedo (Passiflora edulis), com vistas à sele o de um material mais produtivo nas condi es do Distrito Federal para frutos destinados ao consumo in natura. As cultivares Maguari, CSB Marília, NJ3 Vermelho, CSB Marília x NJ3 Vermelho, Roxo Australiano e Sele o DF foram avaliadas durante três anos de produ o, em Latossolo Vermelho-Amarelo, no sistema de espaldeira vertical. A cultivar Maguari apresentou porcentagem de germina o das sementes superior à das demais cultivares. A média da altura das plantas das cultivares Sele o DF e Roxo Australiano foi superior à das outras quatro cultivares. A cultivar Maguari apresentou o maior vigor vegetativo durante os três anos de experimento, e o menor número de plantas perdidas por declínio da cultura. A cultivar CSB Marília foi a mais precoce e a primeira a produzir frutos. Na primeira safra, as cultivares CSB Marília, CSB Marília x NJ3 Vermelho, Roxo Australiano e Sele o DF foram as mais produtivas. No segundo ano de colheita, as seis cultivares apresentaram alta produtividade, destacando-se a cultivar Sele o DF, que produziu 50 t/ha. No terceiro ano de produ o, em que ocorreu um declínio considerável, a cultivar Maguari apresentou a maior produtividade. Na produ o média dos três anos, as seis cultivares mostraram-se superiores em produtividade aos resultados já conhecidos da pesquisa no Brasil.

Abstract: the objective of this study was to evaluate the reaction of 60 passionfruit (passiflora edulis sims f. flavicarpa degener) genotypes to septoriosis. the experimental design was randomized blocks with four replications and six plants by plot. the inoculation was made by spraying a pathogen suspension (105conidia/ml) on the plants. six evaluations were made weekly, 18 days after inoculation. the disease severity was evaluated following the disease index (1, no symptoms; 2, leaves showing until 10% of lesion area; 3, leaves showing 10 to 33% of lesion area; leaves showing over 35% of lesion area; 5, leaf fall) and the percentage of leaf fall. the genotypes which showed less susceptibility were mar.20.27, joseph and mar.20.36, whereas the genotypes mar.20.59 and mar.20.48 presented the higher index of susceptibility. between the score obtained by the less susceptible genotype (3.4) and the score obtained by the most susceptible genotype (4.5) in the lesion scale, there was a difference of 31.8%, indicating that there is variability among the genotypes for resistance to septoriosis. the difference between the highest (81.6%) and the lowest (49.9%) percentage of leaf fall was of 31.7%.
Rea??o de genótipos de maracujazeiro-amarelo ao Colletotrichum gloeosporioides

Abstract: the present work was carried out in order to evaluate the resistance of passion fruit genotypes to c. gloeosporioides, the agent of anthracnose. c. gloeosporioides was inoculated in the lineages of passion fruit plants at a concentration of 5x106 spores on wounds pre-established by abrasion with a fine steel brush. the first evaluation was carried out 20 days after the inoculation and the others in intervals of seven days, ending in the seventh evaluation 62 days after the inoculation. to evaluate the degree of incidence and severity of anthracnose, a grade scale was established, where 1 = absence of symptoms and 7 = leaf loss. average grades < 2 = resistant (r); > 2 and < 3 = moderately resistant (mr); > 3 and < 4 = susceptible (s) and > = highly susceptible (hs). in general, two genotypes were classified as moderately resistant; eight as susceptible and sixty-two were highly susceptible.
Efeito dos substratos artificiais no enraizamento e no desenvolvimento de estacas de maracujazeiro-azedo (Passiflora edulis Sims f. flavicarpa Deg)

Abstract: the experiments were carried out (august to november of 1999 and 200) under greenhouse conditions in embrapa cerrados, planaltina-df. in the first assay, passion fruits (cv. marília-msc) cuttings containing two internodes were immersed in iba (indolbutiric acid) solution at 2000 ppm for 10 seconds and planted in expanded polystyrene trays, with 72 units containing eight organic substrates. the evaluation was made 90 days after planting, by determining the percentage of rooted and budded cuttings and the incidence (% of died cuttings) of colletotrichum gloeosporioides. the plantmax florestal estacaò exhibited higher level of budding and rooting and lower incidence of c. gloeosporioides compared to the substrate plantmax hortali?asò. the nutriplantaò combined with osmocoteò was more efficient than the plantmax florestal estacaò. in the second assay, the cuttings of hybrid rc1 ((f1: marilia x roxo australiano) x marília) were collected in august 2000 and treated with 2000 ppm of iba. the organic substrates used were twelve. the results indicated that the rooting level of rc1 cutting was similar for all substrates. the addition of 1g of osinocote per cutting, 30 days after they were planted in substrate plantmax florestal estaca, prorrided the better development to aerial portion for the rc1 hybrid.

Abstract: with the purpose of evaluating the influence of substrate on the development of yellow passion fruit, an experiment in a greenhouse at embrapa cerrados station has been conducted. the experimental design was a randomized block in a factorial scheme 2 x 4 x 3 x 2, totalizing 48 treatments, 18 useful plants by plot and four replications. the treatments had consisted of combinations of: two commercial substrate (plantmaxr ? to the base of vermiculiter plus pines bark and vermiculiter ? to the base of vermiculiter); three organic sources ? f.o. (humus, cows manure and nutriplantar (product base of bacteria) and absence of f.o., in the ratio of 3:1 of the basic substrate for f.o.; two formulated fertilizers [osmocoter as 14-14-14 (product of slow release of nutrients) and 4-14-8, of normal release], besides the absence of fertilizer and, absence and presence of glomus etunicatum. the artificial substrate plantmaxr was upper than the vermiculiter in all the analyzed features. among the f.o., the nutriplantar with cows manure had provided the best performance. the osmocoter promoted the biggest development of seedlings, followed by 4-14-8. the presence of absence of f.o. matched with plantmaxr practically did not influence the analyzed features. it must not use glomus etunicatum associated the plantmaxr, due to the high phosphorus content in this substrate.
