Compositional changes of proteins and amino acids in germinating coffee seeds

Abstract: endosperm is the main reserve tissue in coffee seeds. coffee (coffea arabica l.) seeds were germinated for six weeks and qualitative and quantitative changes in amino acids and proteins were investigated. the total content of free amino acids were reduced during germination, however, protein content remained constant. sds-page profiles showed that legumin-like proteins became less stained in the last weeks. asparagine, glutamic acid, aspartic acid, alanine and lysine were the major free amino acids, although serine and glutamine were also significant. except for tyrosine, which increased with germination, all other amino acids were reduced. analysis of the amino acid composition of the total soluble protein showed glutamic acid/glutamine and glycine as the main amino acids. however, other amino acids such as leucine, aspartic acid/asparagine, alanine, lysine, serine were also found in reasonable amounts.

Abstract: polyphenoloxidase (pfo) activity has been used to indicate the quality of coffee beverage. a traditional extraction method used for coffee polyphenoloxidase was compared to another in which phenol oxidation was avoided and phenols were eliminated by exclusion chromatography. an intermediate method was also tested. activities were obtained by spectrophotometry, which is commonly used for coffee pfo determination, and o2 consumption. the traditional extraction method and spectrophotometry are inacurate to measure pfo activity since there is a significant interference of phenols present in the extracts. consequentely, the data reported in the literature are not reproducible. pfo activity differentiated soft coffee from hard and rio, but not between the lost two. soft coffee presented higher pfo activity. for an accurate activity determination, antioxidants and phenol complexation is essential during extraction, as well as their elimination by exclusion chromatography. however, using this procedure and o2 consumption, pfo activity could still not differentiate the three coffee qualities, except soft from the other two. instead of 3,4 - dihydroxyphenylalanine, it is suggested that chlorogenic acid (5-caffeoylquinic acid) should be used as substrate.
Extra o e dosagem da atividade da polifenoloxidase do café

Abstract: A atividade de polifenoloxidase (PFO) tem sido usada como indicadora da qualidade de bebida de café. Foram feitas compara es entre o método tradicionalmente usado para extra o de polifenoloxidase de café e outro, onde impediu-se a oxida o de fenóis durante a extra o, com posterior elimina o por filtra o em coluna de exclus o. Um método intermediário entre os dois também foi testado. As medi es de atividade foram feitas por espectrofotometria e por consumo de O2. Os métodos tradicionais de extra o e dosagem da atividade de PFO sofrem forte interferência de fenóis presentes no extrato, n o permitindo reprodu o dos dados publicados na literatura. Por espectrofotometria foi possível diferencia o apenas entre café de bebida Mole de cafés de bebida Dura e Rio, mas n o entre as duas últimas. Café de melhor qualidade, bebida Mole, apresentou maior atividade de PFO. Extra o de PFO na presen a de antioxidantes e complexadores de fenóis, seguida pela elimina o dos mesmos por cromatografia de exclus o é essencial para uma avalia o correta. Entretanto, usando este procedimento e consumo de O2, a atividade de PFO também n o diferenciou os três cafés testados, exceto o Mole dos dois outros. No lugar de DOPA (3,4 - dihidroxifenilalanina), usado comumente nas dosagens por espectrofotometria, sugere-se o emprego de ácido clorogênico (ácido 5-cafeoilquínico).

Abstract: background: over the last 3 years, there has been an increase in the number of patients requesting gluteoplasty. despite this increase, there are no published data on the durability of gluteal prostheses. this study evaluated the mean useful life of silicone implants for gluteoplasty. methods: this was a retrospective observational study. a total of 380 medical records were reviewed to obtain data on age, the date of prosthesis insertion, and the date of reoperation. the mean time between the first surgery for insertion of the implant and the second surgery for exchanging it for a new one or to remove it was calculated. results: of the 380 cases evaluated, 70 patients were reoperated on by the senior author; only 2.8% of these cases showed an intact prosthesis during the reoperation, and all others presented with a ruptured prosthesis. the right and left buttock prostheses were ruptured in 100% and 80% of the cases, respectively. conclusions: the useful life of gluteal implants is shorter than that of breast implants despite being manufactured from the same material. this is because gluteal prostheses are subjected to more tension and compression, which are associated with the lifestyle and job of the patient.

Abstract: coffee is one of the most valuable agricultural commodities and ranks second on international trade exchanges. the genus coffea belongs to the rubiaceae family which includes other important plants. the genus contains about 100 species but commercial production is based only on two species, coffea arabica and coffea canephora that represent about 70 % and 30 % of the total coffee market, respectively. the brazilian coffee genome project was designed with the objective of making modern genomics resources available to the coffee scientific community, working on different aspects of the coffee production chain. we have single-pass sequenced a total of 214,964 randomly picked clones from 37 cdna libraries of c. arabica, c. canephora and c. racemosa, representing specific stages of cells and plant development that after trimming resulted in 130,792, 12,381 and 10,566 sequences for each species, respectively. the ests clustered into 17,982 clusters and 32,155 singletons. blast analysis of these sequences revealed that 22 % had no significant matches to sequences in the national center for biotechnology information database (of known or unknown function). the generated coffee est database resulted in the identification of close to 33,000 different unigenes. annotated sequencing results have been stored in an online database at http://www.lge.ibi.unicamp.br/cafe. resources developed in this project provide genetic and genomic tools that may hold the key to the sustainability, competitiveness and future viability of the coffee industry in local and international markets.
Late surgical correction of anomalous left coronary artery from pulmonary trunk in children, using autogenous aortic and pulmonary endotheliazed tube  [PDF]

Abstract: Background: Direct re-implantation of an anomalous left coronary artery into the aorta is the preferred surgical option for creating a dual coronary arterial system in patients in whom the anomalous artery originated from the pulmonary trunk. This technique, however, is applicable only when the anomalous ar- tery arises from the left posterior pulmonary sinus. We report the successfully late follow-up of series of patients employing a new technique using combined autogenous aortic and pulmonary endotheliazed tube. Patients and Methods: We have treated 3 patients, aged 2 months, 6 months and 8 months respectively, who presented with anomalous origin of the left coronary artery from the left posterior pulmonary sinus and moderated mitral valve insufficiency by left ventricle dysfunction. We used a trapdoor like coronary artery elongation with autogenous aortic and pulmonary endotheliazed tube to connect the left coronary ostium to ascending aorta, avoiding the mitral valve intervention. Results: There was no early or late death. All patients are in functional class I, with good biventricular function and competent mitral valve at a median follow-up of 94 months, ranged from 108 to 132 months. Postoperative Computerized tomogramphy of aorta in our 1st patient showed good arterial flow, without any distortion. A local and distal stenosis of the left pulmonary artery was observed and submitted to stent treatment. Conclusions: The potential benefits of the trapdoor like and its modification technique are excellent operative exposure. The use of autogenous endothelized tube is a viable tissue capable of further growth, avoidance of injury to the aortic and pulmonary valvar apparatus or production of obstruction within the right ventricular outflow tract.

Abstract: Objective: Long-term results after the Senning operation for transposition of the great arteries are little known. Sinus node dysfunction and systemic ventricular dysfunction are crucial in patient survival. We evaluated the results, long term outcome and quality of life in a group of 39 patients. Methods: The study was a retrospective analysis, of 39 (39/40 = 97.5%) surgical surviving patients, submitted to Senning operation, with a mean follow-up time of 14.7 +/– 3.1 years. Electrocardiogram, echocardiogram, and chest radiograph series were performed every 6 months. Thirty six patients of this series underwent Holter study and ergometric test, to evaluate the physical capacity. Three patients living overseas were excluded. Results: There was only one late death (1/39) (late mortality = 2.5%): a 16 year-old patient had a no cardiac death. The actuarial survival was 95.0% (38/40) (simple or with little VSD, TGA). The probability of staying in sinus rhythm, in 39 surviving patients was 77.1% (30) or normal right ventricular function was 76.5% (29), 10 to 20 years after operation. The incidence of sinus node and right ventricular dysfunction increased gradually over time. No re-operations and pacemaker implantation, was performed. Functional class: I = 30 (85.7%) cases and functional class II = 5 (14.3%) cases. Conclusions: Patients with simple TGA submitted to Senning procedure in our experience, presented during late follow-up: 1—Low incidence of right ventricular dysfunction and active arrhythmias; 2—Low mortality and no sudden death recorded; 3—Good quality of life and 4—Satisfactory surgical results (free of re-operation or definitive pacemaker implantation).

Abstract: contrary to the idea that the technical knowledge of the teacher of a native tongue serves primordially to the content of its teaching or to the planning of classes, this article discusses the precept that such knowledge should be seen as the basis for the creation of an attitude to be maintained in face-to-face interactions in the classroom. this attitude relates to the constant need to make decisions before the unexpected, and points to the construction of a specific discursive place for the language teacher - that of someone who listens to the student's words and enlaces them to his or her own words, so as to make sure that the assumption of the position of subject follows a reflection about the linguistic means available to such end. as an example to fuel this debate, the text describes the case of the unusual interpretation made by some students of the word "rataria" present in one of monteiro lobato's works. some of the possible attitudes before this reading mistake are discussed, as well as their implications: to request a change in the answer, to modify the didactic material, to make explicit the linguistic work underlying the mistake, or to use the mistake as a pretext to other activities. the procedures discussed are based on the premise that mistakes and other unforeseen manifestations not only reveal procedures of construction of knowledge, but also offer important opportunities for the teacher to include him/herself into the student's word, being, therefore, a fundamental aspect in the construction of a relationship in which teaching becomes possible.
Training Needs of International Medical Graduates [IMGs] in Psychiatry  [PDF]

Abstract: The potential shortage of psychiatrists over the next 5 - 10 years has focused attention on the need to recruit more IMGs to fill the needs rather than use nurse practitioners or physician assistants. IMGs make up about 1/3 of first year psychiatry residents. These individuals have been found to provide services to the poor, the elderly and the psychotic. The quality of their medical work has been found to be satisfactory. The training needs of these physicians require an understanding on the part of their teachers that they come from cultures with different values that we have. The extended families of these primarily Asian residents clash with our strong commitment to individualism. It leads to a We-self rather than our I-Self. This difference coupled with the stress of leaving to come to a new culture is a great stress. Their exposure to psychiatry has been limited. They request and need more interview demonstration and practice, ore feedback and examinations. They should have help in accent reduction. They should be exposed to the working of the hospital by sitting on departmental and hospital committees. The faculty should extend their social opportunities and work as mentors on joint projects. Courses on the history of American culture should be taught. Psychotherapy for them should be encouraged as well as teaching medical ethics. They must become the major educational concern for the department that they are in.
