Comparison of chromosomal and array-based comparative genomic hybridization for the detection of genomic imbalances in primary prostate carcinomas

Abstract: Our data demonstrate that the reliability of aCGH in the analysis of primary prostate carcinomas depends to some extent on the scoring approach used, with the breakpoint estimation method being the most sensitive and reliable. The pattern of copy number changes detected by aCGH was concordant with that of cCGH, but the higher resolution technique detected 2.7 times more aberrations and 15.2% more carcinomas with genomic imbalances. We additionally show that several aberrations were consistently overlooked using cCGH, such as small deletions at 5q, 6q, 12p, and 17p. The latter were validated by fluorescence in situ hybridization targeting TP53, although only one carcinoma harbored a point mutation in this gene. Strikingly, homozygous deletions at 10q23.31, encompassing the PTEN locus, were seen in 58% of the cases with 10q loss.We conclude that aCGH can significantly improve the detection of genomic aberrations in cancer cells as compared to previously established whole-genome methodologies, although contamination with normal cells may influence the sensitivity and specificity of some scoring approaches. Our work delineated recurrent copy number changes and revealed novel amplified loci and frequent homozygous deletions in primary prostate carcinomas, which may guide future work aimed at identifying the relevant target genes. In particular, biallelic loss seems to be a frequent mechanism of inactivation of the PTEN gene in prostate carcinogenesis.Prostate cancer is a frequent and heterogeneous malignancy with few established prognostic markers. Abstract: We examined 35 low- and 99 high-grade gliomas using quantitative methylation specific PCR (qMSP) and pyrosequencing. Gene expression level of MGMT was analyzed by RT-PCR.When examined by qMSP, 26% of low-grade and 37% of high-grade gliomas were found to be methylated, whereas 97% of low-grade and 55% of high-grade gliomas were found methylated by pyrosequencing. The average MGMT gene expression level was significantly lower in the group of patients with a methylated promoter independent of method used for methylation detection. Primary glioblastoma patients with a methylated MGMT promoter (as evaluated by both methylation detection methods) had approximately 5 months longer median survival compared to patients with an unmethylated promoter (log-rank test; pyrosequencing P = .02, qMSP P = .06). One third of the analyzed samples had conflicting methylation results when comparing the data from the qMSP and pyrosequencing. The overall survival analysis shows that these patients have an intermediate prognosis between the groups with concordant MGMT promoter methylation results when comparing the two methods.In our opinion, MGMT promoter methylation analysis gives sufficient prognostic information to merit its inclusion in the standard management of patients with high-grade gliomas, and in this study pyrosequencing came across as the better analytical method.Gliomas are histologically divided into several subgroups including astrocytomas, oligodendrogliomas, and oligoastrocytomas and are graded from I to IV according to the WHO classification [1]. Prognosis is highly variable depending on histopathology, grade, patient age, and genetic tumor factors such as the presence of a 1p/19q co-deletion, IDH1 and IDH2 mutations, and MGMT promoter methylation [1,2]. Abstract: Candidate biomarkers were subjected to quantitative methylation analysis in test sets of tissue samples from colorectal cancers, adenomas, and normal colonic mucosa. All findings were verified in independent clinical validation series. A total of 523 human samples were included in the study. Receiver operating characteristic (ROC) curve analysis was used to evaluate the performance of the biomarker panel.Promoter hypermethylation of the genes CNRIP1, FBN1, INA, MAL, SNCA, and SPG20 was frequent in both colorectal cancers (65-94%) and adenomas (35-91%), whereas normal mucosa samples were rarely (0-5%) methylated. The combined sensitivity of at least two positives among the six markers was 94% for colorectal cancers and 93% for adenoma samples, with a specificity of 98%. The resulting areas under the ROC curve were 0.984 for cancers and 0.968 for adenomas versus normal mucosa.The novel epigenetic marker panel shows very high sensitivity and specificity for both colorectal cancers and adenomas. Our findings suggest this biomarker panel to be highly suitable for early tumor detection.Colorectal cancer is the third most common cancer type in the US and is a major contributor to cancer-death [1]. Most cases of colorectal cancer develop from benign precursors (adenomas) during a long time interval. This provides a good opportunity for detection of colorectal cancer at an early curable stage and to screen for potentially pre-malignant adenomas [2]. Both flexible sigmoidoscopy and the Fecal Occult Blood Test (FOBT) have been tested in randomized trials and shown to reduce mortality from colorectal cancer [3]. By sigmoidoscopy adenomas may be detected and removed and thus the incidence of cancer will be reduced [4], however, this screening is invasive and cumbersome for the patient. Abstract: This paper aim to explain why the prenominal definiteness marker found in modified structures only is acquired much later than the suffixal definite article in Norwegian. The coexistence of the two definiteness markers is the result of the so-called double definiteness phenomenon in Norwegian which occurs in definite structures involving an attributive adjective. As the prenominal determiner only occurs in modified contexts, one obvious explanation that presents itself is that the omission of the prenominal determiner is related to the fact that it is much less frequent in the input than the suffixal article. However, if we consider the form of this determiner, it is clear that the form itself must be very frequent in the input, as it is homophonous with the demonstrative and with inanimate third person pronouns, which are all frequent in use. Consequently, a lexical insertion approach to the double definiteness phenomenon is proposed according to which the discrepancy in the order of acquisition is argued to be due to a combination of prosodic saliency and the lexicalisation of semantic features, the latter of which will be dealt with here.

Abstract: We conduct an on-line search of the Cochrane Database of Systematic Reviews (CDSR) and divide all Cochrane reviews according to their design inclusion criterion: (A) RCTs only or (B) RCTs and (some subset of) observational studies. We test statistically whether a shift in the proportion of category B reviews has occurred by comparing reviews published before 2008 with reviews published during 2008/09.We find that the proportion of Cochrane reviews choosing a broader inclusion criterion has increased, although by less than two percentage points. The shift is not statistically significant (P?=?0.08).There is currently not sufficient data to support a hypothesis of a significant shift in favour of including observational studies, neither at the aggregate level nor at the level of individual Review Groups within the Cochrane Collaboration.“Evidence based medicine is the conscientious, explicit, and judicious use of current best evidence in making decisions about the care of individual patients” [1]. Abstract: Da yrkesskoler, husflidsskoler og gymnas i Norge ble samlet i én felles videreg ende skole i 1976, var en av intensjonene at elevene skulle kunne kombinere praktisk og teoretisk utdanning. P ”husflidslinja” fikk kunst- og designfagene gode rammer for utvikling, men elevene fikk ingen sluttkompetanse. I perioden 1994-2006 fikk kunst- og designfagene stor oppslutning med utdannings--tilbudet Tegning, form og farge med studiekompetanse. Denne hybriden ble opprettet under den yrkesfaglige studieretningen Formgivingfag, og den fungerte som et studie--forberedende tilbud som b de ga studie-kompetanse og realkompetanse som grunnlag for opptak p h yere utdanning innen kunst, design og arkitektur. Ambisjonen fra 1976 om kombinere teoretiske og praktiske fag har blitt fulgt opp med ke antall obligatoriske teoretiske allmennfag p de yrkesfaglige studieretningene, spesielt etter 1994. Designers, decision makers, investors and consumers hold different positions in the design process, but they all make choices that will influence our future visual and material culture. To promote sustainability and meet global challenges for the future, professional designers are dependent on critical consumers and a design literate general public. For this purpose design education is important for all. We propose that design education in general education represents both a foundation for professional design education and a vital requirement for developing the general public’s competence for informed decision making.
