Despite better understanding of the biology and intense research in the treatment of these cancers, the long-term survival remains poor both in the locally advanced and metastatic settings. The addition of combined modality strategies has resulted in modest improvement in 5-year survival rates. A number of biologic agents targeting epidermal-derived growth factor receptor, vascular endothelial derived growth factor and its receptor, and mammalian target of rapamycin (mTOR) are being currently evaluated in Phase II and III clinical trials. Some of these, like trastuzumab, cetuximab, and bevacizumab, have shown promising results. This review provides a brief overview of the recent developments in biologic agents for the treatment of esophagogastric cancers.

Fruiting Body Production and Aroma Profile Analysis of Agrocybe aegerita Cultivated on Different Substrates  [PDF]

Abstract:

The industrial cultivated basidiomycete Agrocybe aegerita, also known as “Pioppino” in Italy or “Samthaube” in Germany, is a high quality mushroom with a delicious aroma. Cultivation of A. aegerita on wheat straw supplemented with different residues of the food industry showed the highest yields in fruiting body production with a biological efficiency of 36% when black tea pomace was added. The addition of other substrates resulted in biological efficiencies of 23% to 33%. Besides the mushroom yields, the number and size of fruiting bodies harvested on the different substrates were determined. A comparison of the aroma profiles of A. aegerita grown on wheat straw and on wheat straw supplemented with black tea by means of GC/MS/MS/O is presented.

Coupling of cytoplasm and adhesion dynamics determines cell polarization and locomotion

Abstract: Observations of single epidermal cells on flat adhesive substrates have revealed two distinct morphological and functional states, namely a non-migrating symmetric unpolarized state and a migrating asymmetric polarized state. These states are characterized by different spatial distributions and dynamics of important biochemical cell components: F-actin and myosin-II form the contractile part of the cytoskeleton, and integrin receptors in the plasma membrane connect F-actin filaments to the substratum. In this way, focal adhesion complexes are assembled, which determine cytoskeletal force transduction and subsequent cell locomotion. So far, physical models have reduced this phenomenon either to gradients in regulatory control molecules or to different mechanics of the actin filament system in different regions of the cell. Here we offer an alternative and self-organizational model incorporating polymerization, pushing and sliding of filaments, as well as formation of adhesion sites and their force dependent kinetics. All these phenomena can be combined into a non-linearly coupled system of hyperbolic, parabolic and elliptic differential equations. Aim of this article is to show how relatively simple relations for the small-scale mechanics and kinetics of participating molecules may reproduce the emergent behavior of polarization and migration on the large-scale cell level.
QUANTIFICATION OF BIOFILMS IN MULTI-SPECTRAL DIGITAL1 VOLUMES FROM CONFOCAL LASER-SCANNING MICROSCOPES

Abstract: Populations of bacteria in sludge flocs and biofilm marked by fluorescence marked with fluorescent probes are digitised with a confocal laser scanning microscope. These data are used to analyse the microbial community structure, to obtain information on the localisation of specific bacterial groups and to examine gene expression. This information is urgently required for an in-depth understanding of the function and, more generally, the microbial ecology of biofilms. Methods derived from quantitative image analysis are applied to digitised data from confocal laser scanning microscopes to obtain quantitative descriptions of volumetric, topological (and topographical) properties of different compartments of the components under research. In addition to free-moving flocs, also biofilms attached to a substratum in an experimental environment are analysed. Growth form as well as interaction of components are quantitatively described. Classical measurements of volume and intensity (shape, distribution) and distance dependent interaction measurements using methods from mathematical morphology are performed. Mainly image (volume) processing methods are outlined. Segmented volumes are globally and individually (in terms of 3Dconnected components) measured and used for distance mapping transform as well as for estimation of geodesic distances from the substrate. All transformations are applied on the 3D data set. Resulting distance distributions are quantified and related to information on the identity and activity of the probe-identified bacteria.
Improved success rate and stability for phase retrieval by including randomized overrelaxation in the hybrid input output algorithm

Abstract: In this paper, we study the success rate of the reconstruction of objects of finite extent given the magnitude of its Fourier transform and its geometrical shape. We demonstrate that the commonly used combination of the hybrid input output and error reduction algorithm is significantly outperformed by an extension of this algorithm based on randomized overrelaxation. In most cases, this extension tremendously enhances the success rate of reconstructions for a fixed number of iterations as compared to reconstructions solely based on the traditional algorithm. The good scaling properties in terms of computational time and memory requirements of the original algorithm are not influenced by this extension.
Combined Pulse Electroporation – A Novel Strategy for Highly Efficient Transfection of Human and Mouse Cells

Abstract: The type of a nucleic acid and the type of the cell to be transfected generally affect the efficiency of electroporation, the versatile method of choice for gene regulation studies or for recombinant protein expression. We here present a combined square pulse electroporation strategy to reproducibly and efficiently transfect eukaryotic cells. Cells suspended in a universal buffer system received an initial high voltage pulse that was continuously combined with a subsequent low voltage pulse with independently defined electric parameters of the effective field and the duration of each pulse. At comparable viable cell recoveries and transfection efficiencies of up to 95% of all cells, a wide variety of cells especially profited from this combined pulse strategy by high protein expression levels of individual cells after transfection. Long-term silencing of gene expression by transfected small interfering RNA was most likely due to the uptake of large nucleic acid amounts as shown by direct detection of fluorochromated small interfering RNA. The highly efficient combined pulse electroporation strategy enables for external regulation of the number of naked nucleic acid molecules taken up and can be easily adapted for cells considered difficult to transfect.
Review of the Use of Electroencephalography as an Evaluation Method for Human-Computer Interaction

Abstract: Evaluating human-computer interaction is essential as a broadening population uses machines, sometimes in sensitive contexts. However, traditional evaluation methods may fail to combine real-time measures, an "objective" approach and data contextualization. In this review we look at how adding neuroimaging techniques can respond to such needs. We focus on electroencephalography (EEG), as it could be handled effectively during a dedicated evaluation phase. We identify workload, attention, vigilance, fatigue, error recognition, emotions, engagement, flow and immersion as being recognizable by EEG. We find that workload, attention and emotions assessments would benefit the most from EEG. Moreover, we advocate to study further error recognition through neuroimaging to enhance usability and increase user experience.
Kontekstuele bepalers by die beplanning van skoolwoordeboeke

Abstract: Die doel van hierdie artikel is om tot die metaleksikografiese gesprek oor skoolwoordeboeke as 'n tipe pedagogiese woordeboek by te dra. Die fokus is op kontekstuele bepalers wat by die beplanning van skoolwoordeboeke verreken behoort te word: Binne die konteks van 'n veeltalige Namibi word enkele onderwyskundige sisteme verken wat 'n impak op die ontwerp van gedrukte skoolwoordeboeke vir die sekondêre skoolfase kan hê, naamlik skoolkurrikula, die nasionale taalbeleid vir skole en die beleid van leerdergerigte onderwys. Hierdie artikel voer ook aan dat die voor die hand liggende teikengebruiker van 'n skoolwoordeboek, naamlik die leerder, nie die enigste teikengebruiker is nie: Die onderwyser word as medeteikengebruiker gestel wanneer dit by die gebruik van woordeboeke in die onderrigleersituasie kom. Verskillende tipes gevalle van woordeboekgebruik in die mikrokonteks van die onderrigleersituasie word onderskei, naamlik woordeboekonderrig, woordeboekge ntegreerde taalonderrig en woordeboekgeondersteunde taalonderrig. Hierdie tipes gebruik behoort komponente van 'n woordeboekpedagogie te wees, wat uiteindelik behoort te lei tot outonome woordeboekgebruik en die gepaardgaande vestiging van 'n individuele woordeboekkultuur by die leerder. Die leksikograaf moet al hierdie aspekte inreken wanneer 'n skoolwoordeboek beplan word ten einde die nodige leksikografiese vernuwing in te voer. Enkele voorstelle vir sodanige vernuwings en vir verdere navorsing word ten slotte aangebied. Abstract: Adresseringsekwivalensie geld in 'n woordeboekartikel wanneer al die vertaalekwivalente in die vertaalekwivalentparadigma neerslag vind in die doeltaalvoorbeelde in die koteksafdeling en elke doeltaalvoorbeeld 'n vertaalekwivalent bevat wat in die vertaalekwivalentparadigma voorkom. Die versteuring van hierdie ekwivalensie vind plaas deur prosedures van onderadressering (wanneer nie al die vertaalekwivalente in die vertaalekwivalentparadigma in doeltaalvoorbeelde in die koteksafdeling neerslag vind nie) en ooradressering (wanneer doeltaalvoorbeelde vertaalekwivalente bevat wat nie in die vertaalekwivalentparadigma voorkom nie). In 'n polifunksionele tweetalige woordeboek verteenwoordig adresseringsekwivalensie die ideale adresseringstruktuur tussen vertaalekwivalentparadigma en koteksafdeling. In Afrikaanse tweetalige woordeboeke is adresseringsekwivalensie egter skaars, maar dit blyk in tweetalige woordeboeke van ander tale ook die geval te wees. Hoewel gevalle van onderadressesing relatief maklik geneutraliseer kan word, bevind 'n dieper ondersoek na gevalle van ooradressering dat die meeste doeltaalvoorbeelde wat simptomaties van ooradressering blyk te wees, wel tot kommunikatiewe ekwivalensie bydra ten spyte van 'n o nskynlik gebrekkige adresseringstruktuur. Dít is omdat hierdie doeltaalvoorbeelde nie as ware voorbeelde (wat die optrede van vertaalekwivalente illustreer) funksioneer nie, maar in der waarheid optree as kotekstuele spesifiseerders wat 'n funksionele prosedure van kontekstuele vertaalekwivalentuitsluiting verteenwoordig, waardeur die teikengebruiker gewys word op die nié-bruikbaarheid van die aangebode vertaalekwivalente in die vertaalekwivalentparadigma in bepaalde kontekste. Hierdie bevinding noop 'n herwaardering van adresseringstrukture in die algemeen en die beginsel van adresseringsekwivalensie in die besonder.
