Análise de diversidade genética do gene da osteopontina em bovinos da ra?a girolando

Abstract: the objective was to obtain the indices of genetic diversity for the snp (single nucleotide polymorphism) of the 4 intron osteopontin gene (opn) for 434 animals (87 bulls and 347 cows) participants in the teste de progênie da ra?a girolando (girolando progeny test) in brazil. for amplification, primers used were described for the holstein breed, and differentiation of alleles c/t snp that was obtained by pcr-rflp. genotype frequencies of tt (52.53%), ct (38.71%) and cc (8.76%) and allele frequencies of t (71.9%) and c (28.1%) indicate that the population is in hardy-weinberg principle (hwp). although the opn gene locus is in hwp, the higher frequency of allele t of snp in these animals may suggest a setting-trend of allele t in the race. no difference was observed between the group of bulls and cows (fst = -0.018), supporting the estimate of population balance. considering the values estimated by the fis (0.043), it is likely that high numbers of individuals homozygous for the t allele observed in the population occur because of possible inheritance of this allele coming from the zebu breed, rather than inbreeding. thus, to better characterize the opn gene polymorphism, assessments in a larger number of animals must be performed, since only animals that participated in the progeny test were assessed.
Detection and quantification of Roundup Ready? soybean residues in sausage samples by conventional and real-time PCR

Abstract: the increasing presence of products derived from genetically modified (gm) plants in human and animal diets has led to the development of detection methods to distinguish biotechnology-derived foods from conventional ones. the conventional and real-time pcr have been used, respectively, to detect and quantify gm residues in highly processed foods. dna extraction is a critical step during the analysis process. some factors such as dna degradation, matrix effects, and the presence of pcr inhibitors imply that a detection or quantification limit, established for a given method, is restricted to a matrix used during validation and cannot be projected to any other matrix outside the scope of the method. in brazil, sausage samples were the main class of processed products in which roundup ready? (rr) soybean residues were detected. thus, the validation of methodologies for the detection and quantification of those residues is absolutely necessary. sausage samples were submitted to two different methods of dna extraction: modified wizard and the ctab method. the yield and quality were compared for both methods. dna samples were analyzed by conventional and real-time pcr for the detection and quantification of roundup ready? soybean in the samples. at least 200 ng of total sausage dna was necessary for a reliable quantification. reactions containing dna amounts below this value led to large variations on the expected gm percentage value. in conventional pcr, the detection limit varied from 1.0 to 500 ng, depending on the gm soybean content in the sample. the precision, performance, and linearity were relatively high indicating that the method used for analysis was satisfactory.
Tannin-tolerant bacteria from crossbred Holstein x Zebu cows

Abstract: the objective of this work was to isolate and characterize tannin-tolerant ruminal bacteria from crossbred holstein x zebu cows fed a chopped mixture of elephant grass (pennisetum purpureum), young stems of "angico-vermelho" (parapiptadenia rigida), and banana tree (musa sp.) leaves. a total of 117 bacteria strains were isolated from enrichment cultures of rumen microflora in medium containing tannin extracts. of these, 11 isolates were able to tolerate up to 3 g l-1 of tannins. classical characterization procedures indicated that different morphological and physiological groups were represented. restriction fragments profiles using alu1 and taq1 of 1,450 bp pcr products from the 16s rrna gene grouped the 11 isolates into types i to vi. sequencing of 16s rrna pcr products was used for identification. from the 11 strains studied, seven were not identifiable by the methods used in this work, two were strains of butyrivibrio fibrisolvens, and two of streptococcus bovis.
Differential expression of genes in follicular cells of swines

Abstract: the main purpose of the present study was to identify for candidate genes related to ovulation in swines. to do so, it was investigated in ovarian follicular cells through quantitative real-time pcr the differential expression of the following genes: steroidogenic acute regulator (star), gata-binding protein 4 (gata), prostaglandin f2α (pgf2α), progesterone receptor (p4r), follicle-stimulating hormone receptor (fshr), and cytochrome p450 aromatase (cyp19). these genes encode hormone receptors (fshr and p4r), hormone (pgf2α), steroidogenic proteins (star and cyp19) and transcription factor (gata). folicular cells were collected from sows with high and low number of piglets/litters during the follicular phase of the estrus cycle. there was difference in transcript abundance among low and high prolific sows for the star, gata, pgf2α, p4r and cyp19 genes. for the fshr gene, the fold change was not considered to be significantly different. because in the present study only the transcript level of the above mentioned genes was analyzed, no inference can be made regarded to protein translation or activity. therefore, gene sequence trials and other functional studies will be necessary to complement the present results, allowing a better understanding on biological complexity of these genes and their use as markers for prolificity in swines.

Abstract: manuscripts submitted at meetings do not necessarily qualify as scientific papers since they do not adjust themselves to the objectives and structures required. it was concluded through this analysis that a scientific paper is an essential part of the proceds of scientific research, which is not complete unless their results are published. such a reason motivated us to conduct this study aimed at updating the methodological structure of the scientific paper, allowing those nursing professionals who review and prepare these texts to develop abilities in this sense, increasing the number and quality of the articles to be published. this bibliographic or documentary review was based on the study on paho's documents and from other cuban authors that devote their time to examine these aspects. the nursing personnel will be able to improve and update their knowledge in this field and on the methodological structure of the scientific paper by this schematic information. this will influence on the quality, scientific and technical level, and national and international projection of the cuban journal of nursing.

Abstract: Los manuscritos presentados en reuniones no califican necesariamente como artículo científico, ya que no se ajustan a los objetivos y estructuras requerida. De este análisis se concretó que el artículo científico es una parte esencial del proceso de investigación científica, y que ésta no está completa mientras sus resultados no se publican. Por tal razón nos motivó a realizar este estudio, con el fin de actualizar la estructura metodológica del artículo científico, que permita a aquellos profesionales de enfermería que se dedican a la revisión y elaboración de estos textos, desarrollen habilidades en este sentido, proporcionando el aumento en número y calidad de los artículos que se publicarán. Esta revisión bibliográfica o documental se efectuó mediante estudios de documentos de la OPS y otros autores cubanos que dedican su tiempo al estudio de estos aspectos. Dicha revisión nos permitirá contribuir en mejorar el conocimiento en este campo, en el personal de enfermería y a su vez actualizarlo acerca de la estructura metodológica del artículo científico, mediante esta información esquemática. Esto influirá en la calidad, nivel científico-técnico y proyección nacional e internacional en la Revista Cubana de Enfermería. Impact of subclinical hypothyroidism treatment in systolic and diastolic cardiac function

Abstract: objective: to evaluate the effects of levothyroxine (l-t4) replacement in echocardiographic parameters of middle-aged women with subclinical hypothyroidism (sh). subjects and methods: this was a randomized, double-blind, placebo-controlled study. echocardiographic evaluation was carried out at baseline and one year after restoration of euthyroidism. thirty-three women with sh were assigned to one of two groups (l-t4 or placebo). results: the two groups had similar basal characteristics. there was a significant deterioration of left ventricular tei index after one year of placebo use, which differed from the effect of l-t4 replacement (+0.086 ± 0.092 vs. -0.014 ± 0.012; p = 0.047). there was also a slight reduction in cardiac output and cardiac index with placebo use, which was not different from l-t4 effect. conclusion: results suggest a positive impact of l-t4 replacement in cardiac function of middle-aged women with sh.
AS UNIDADES DE CONSERVA O GERENCIADAS PELO IEF REGIONAL DIAMANTINA (MG): UM ENFOQUE METODOLOGICO ACERCA DOS INVENTáRIOS TURíSTICOS

Abstract: Sabe-se que Minas Gerais possui significativa relevancia ambiental, diante das riquezas e variedades de paisagens, recursos hídricos, diversidade da fauna e flora e sítios arqueológicos. A necessidade de preserva o ambiental é incentivada com a implanta o das unidades de conserva o (UC’s). S o necessárias algumas estratégias no processo de preserva o e apropria o da sociedade pelo meio ambiente e UC’s, como o uso público, por meio da atividade ecoturística. E, para que o turismo ocorra de forma responsável é primordial a realiza o de pesquisas que viabilizem o planejamento e avalia o constante acerca dos tipos e formas mais apropriados de uso. Nesse contexto, foi firmado um convênio entre a Universidade Federal dos Vales do Jequitinhonha e Mucuri (UFVJM), por meio do Núcleo de Estudos em Turismo, e o Instituto Estadual das Florestas (IEF) – Regional Alto Jequitinhonha, no intuito de desenvolver uma pesquisa-a o dos atrativos turísticos de quatro UC′s: Parque Estadual de Biribiri (PEBI), Parque Estadual do Pico do Itambé (PEPI), Parque Estadual da Serra do Intendente (PESI) e Parque Estadual da Serra do Cabral (PESC). A metodologia consistiu em quatro etapas: 1) Pesquisa Bibliográfica: referencial teórico, considerando o conhecimento cientifico publicado para instruir os discentes envolvidos no desenvolvimento do projeto além do embasamento teórico do inventário; 2) Pesquisa Documental: levantamento de gabinete em documentos técnicos, como Plano de Manejo, características gerais, aspectos da constitui o, usos sociais e históricos do território e área circunvizinha. Implicou na caracteriza o dos aspectos físico-naturais da UC, analise das cartas topográficas e planejamento da estratégia da pesquisa in loco; 3) Pesquisa in loco: atua o direta dos discentes nas UC’s. Foi realizada, a priori, a descri o de todos os atrativos (turísticos efetivos e,ou potenciais) com identifica o da: localiza o, acesso, distancias, coordenadas geográficas, grau de conserva o, características de vegeta o, relevo e hidrografia, índices de eleva o, medidas e características fision micas, conforme equipamentos eletr nicos disponibilizados pelos parceiros envolvidos. Estabeleceu-se, ainda, o georeferenciamento das trilhas, pontos de referências, atrativos e infra-estrutura existente, bem como registro sistemático em mídia digital das imagens e paisagens das UC’s; 4) Tabula o dos dados e elabora o do documento final: digitaliza o dos dados coletados para elabora o do inventário, com todas as informa es e imagens dos atrativos visitados.
