New methods in mammary gland development and cancer: proteomics, epigenetics, symmetric division and metastasis

Abstract: Speaking of Weggis, where the European Network for Breast Development and Cancer (ENBDC) meeting is held, Mark Twain said 'Sunday in heaven is noisy compared to this quietness'. Indeed, people working on breast development and cancer took full advantage of this wonderfully quiet and beautiful place again this year for in-depth discussions. The meeting, held in April, started with an inspiring keynote lecture by Nancy Hynes and included the following sessions.The proteomics session consisted of two lectures, one by Bernd Bodenmiller (University of Zurich, Switzerland) and the other by Arzu Umar (Erasmus University Medical Center, Rotterdam, The Netherlands). Bodenmiller talked about mass cytometry, a new technology that combines flow cytometry and atomic mass spectrometry and that allows simultaneous measurement of up to 100 parameters [1]. This method allows the analysis of complex regulatory networks activated in response to various environmental signals at the single-cell level and can provide a system-wide picture of signaling events. Cellular signaling intermediates are recognized by antibodies, as used for other techniques, but become labeled with rare earth metals, non-radioactive and non-biological, instead of fluorochromes, and this generates a significantly larger range of reporter tags. In the mass cytometer, cells are introduced into the plasma, atomized, and analyzed by mass spectrometry. At present, a shortage of suitable antibodies against cellular signaling compounds is one of the limiting factors in mass cytometry. Abstract: The European Network for Breast Development and Cancer (ENBDC) organized its second meeting to foster interactions and the sharing of protocols between groups working on breast development and cancer. Graduate students, postdocs and research associates were encouraged to attend. The meeting included discussions on genomics, bioinformatics, intravital microscopy, disseminated tumor cells, ex vivo culture and in vivo models for studying breast cancer.Nuno Barbosa-Morais (Cancer Research UK, Cambridge Research Institute) discussed the importance of the correct annotation of microarray probes and of being sure that a probe truly maps to the gene of interest. Further problems to consider are probes that map to intron-exon boundaries, the presence of SNPs, and alternative splicing, which, as is becoming apparent, occurs on a far wider scale than previously appreciated. Splicing may lead to difficulties when summarizing data from multiple probes apparently mapping to the same gene but which in fact detect different splice variants. Overall, it is clear that whatever array platform is being used, application of the latest, most reliable annotation is important. In a test of annotation reliability, it was found that Refseq annotations are a more reliable guide to probe identity than GenBank/UniGene.Britta Weigelt (Cancer Research UK, London Research Institute) spoke on the design of gene expression microarray studies, which fall into three types. The first, class comparison, is a supervised analysis to define molecular differences between predefined groups. The second, class prediction, is a supervised analysis where, after identifying the transcriptional differences between predefined groups, a genomic classifier (signature) is defined to classify new samples. Abstract: The mammary gland consists of secretory alveoli connected by a system of branching ducts embedded in connective tissue. The epithelial cells that compose the gland are arranged in two layers, the luminal epithelial layer and the basal myoepithelial layer. The whole structure is surrounded by a basement membrane.The myoepithelial layer is organised differently in the ducts and in the lobules. In the ducts, elongated myoepithelial cells form a more or less continuous layer and are in direct contact with the basement membrane, and hence with the stroma. The interaction between ductal luminal cells and the extracellular matrix (ECM) is largely mediated by the myoepithelium, although some of the luminal cells in the mammary ducts may reach the basement membrane. Alveolar myoepithelial cells are of stellate shape and form a basket-like structure around the acini, resulting in the exposure of most of the basal surface of the luminal cell to the basement membrane.Differentiated myoepithelial cells are highly contractile and their ultrastructure is reminiscent of that of smooth muscle cells. Myoepithelial cells contain large amounts of micro-filaments, dense plaques (cell–matrix adherens junctions characteristic of smooth muscle cells) and smooth muscle-specific cytoskeletal and contractile proteins. Abstract: Several archaic features of interpreting the surrounding world arestill present in Udmurt folk culture. Calendar-related customs and feasts still preserve the oldest elements on all levels of rituals: in activities, artefacts, verbal and acoustic fields, etc. The dismissal of pests and caterpillars, and their wedding rituals, are deeply rooted in calendar customs. The thorough study of the codes of these rituals would help to determine the semantics of rituals,ascertaining the synchronic-diachronic aspects of the calendar and provide an integral imagination with regard to the mythopoetic foundations of popular worldview. Relying on the analysis of the specificity of Udmurt calendar feasts and customs, it becomes obvious that the tradition of warding off insects and caterpillars takes place in different seasons and is an inseparable part of the calendar cycle. Having analysed the specificity of Udmurt calendar feasts andcustoms, the rituals associated with the dismissal of insects is intrinsically polyfunctional, whereas the most archaic feature therein is the idea of conjuring the rain.

A Community Success Model for Gaming Communities

Abstract: Community satisfaction has become a crucial aspect for success of emerging multimedia information systems. For measuring community satisfaction we propose a community success model (CSM) based on the seminal DeLone & McLean IS Success Model. CSM combines quantitative data collected through automatic monitoring of service usage with qualitative extracted from automatically generated questionnaires. As proof of concept we propose to apply our CSM to evaluate innovative community supporting services created within our regular lab course “Gaming Communities” at RWTH Aachen University. Video gaming is part of our new media culture, entertaining people of all ages. The satisfaction of gaming community members can influence the success of a game and therefore presents a relevant object of investigation for CSM.
Membrane Orientation and Binding Determinants of G Protein-Coupled Receptor Kinase 5 as Assessed by Combined Vibrational Spectroscopic Studies

Abstract: G-protein coupled receptors (GPCRs) are integral membrane proteins involved in a wide variety of biological processes in eukaryotic cells, and are targeted by a large fraction of marketed drugs. GPCR kinases (GRKs) play important roles in feedback regulation of GPCRs, such as of β-adrenergic receptors in the heart, where GRK2 and GRK5 are the major isoforms expressed. Membrane targeting is essential for GRK function in cells. Whereas GRK2 is recruited to the membrane by heterotrimeric Gβγ subunits, the mechanism of membrane binding by GRK5 is not fully understood. It has been proposed that GRK5 is constitutively associated with membranes through elements located at its N-terminus, its C-terminus, or both. The membrane orientation of GRK5 is also a matter of speculation. In this work, we combined sum frequency generation (SFG) vibrational spectroscopy and attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR) to help determine the membrane orientation of GRK5 and a C-terminally truncated mutant (GRK51-531) on membrane lipid bilayers. It was found that GRK5 and GRK51-531 adopt a similar orientation on model cell membranes in the presence of PIP2 that is similar to that predicted for GRK2 in prior studies. Mutation of the N-terminal membrane binding site of GRK5 did not eliminate membrane binding, but prevented observation of this discrete orientation. The C-terminus of GRK5 does not have substantial impact on either membrane binding or orientation in this model system. Thus, the C-terminus of GRK5 may drive membrane binding in cells via interactions with other proteins at the plasma membrane or bind in an unstructured manner to negatively charged membranes.

Abstract:

Between 1928 and 1953, Joseph Stalin was the undisputed totalitarian dictator of the former Soviet Union whose “reign of fear” continues to maintain its egregious reputation. An examination of Stalin’s documented behaviors attempts to evaluate any signs of psychopathology in accordance with DSM-IV-TR criteria. Evidence of a troubled upbringing, depression, paranoia, and alcohol abuse suggests psychopathology as an implicating factor behind Stalin’s actions. Utilizing such a perspective may allow for future distinctions of individuals deemed responsible for horrendous atrocities.

The Role of Group Size and Correlated Project Outcomes in Group Lending  [PDF]

Abstract: This paper analyzes contract efficiency with regard to correlated project realization and the size of the borrowers in group lending. Firstly, I show that under the standard assumption of independent project payoffs, the expected group cost of default decreases with group size. Secondly, I show that small groups can also optimize group efficiency if individual payoffs and credit risks are correlated. The results outline that social cost minimization occurs due to a common interest in forming optimal borrower groups between lenders and borrowers.

Abstract: This paper fits into the research stream started by Aumann and Serrano (2008) with their index RAS, and introduces a new index of riskiness called Iθ. In particular, our intuition moves from the observation that the RAS index is defined over the set of gambles whose expected value of losses is lower than the one of gains. We then restrict the attention on investment opportunities in a proper relation with a benchmark value \"\" and we build the index Iθ. The mathematical features of the new index are discussed in deepest detail, providing evidence that Iθ can be used by the investor in comparing risks: θ plays the role of a threshold value such that once fixed in the range (0,1] , Iθ suggests to take into consideration a gamble (an investment, an asset) g only if the ratio between the expected value of its losses and the expected value of its gains is lower than such θ. Moreover, Iθ nests RAS as special case when θ = 1, and it satisfies both homogeneity and duality properties. In the light of these features, Iθ seems to satisfy the common need among practitioners for flexible index of riskiness.
