Pharmacometabolomics of Response to Sertraline and to Placebo in Major Depressive Disorder – Possible Role for Methoxyindole Pathway

Abstract: Therapeutic response to selective serotonin (5-HT) reuptake inhibitors in Major Depressive Disorder (MDD) varies considerably among patients, and the onset of antidepressant therapeutic action is delayed until after 2 to 4 weeks of treatment. The objective of this study was to analyze changes within methoxyindole and kynurenine (KYN) branches of tryptophan pathway to determine whether differential regulation within these branches may contribute to mechanism of variation in response to treatment. Metabolomics approach was used to characterize early biochemical changes in tryptophan pathway and correlated biochemical changes with treatment outcome. Outpatients with MDD were randomly assigned to sertraline (n = 35) or placebo (n = 40) in a double-blind 4-week trial; response to treatment was measured using the 17-item Hamilton Rating Scale for Depression (HAMD17). Targeted electrochemistry based metabolomic platform (LCECA) was used to profile serum samples from MDD patients. The response rate was slightly higher for sertraline than for placebo (21/35 [60%] vs. 20/40 [50%], respectively, χ2(1) = 0.75, p = 0.39). Patients showing a good response to sertraline had higher pretreatment levels of 5-methoxytryptamine (5-MTPM), greater reduction in 5-MTPM levels after treatment, an increase in 5-Methoxytryptophol (5-MTPOL) and Melatonin (MEL) levels, and decreases in the (KYN)/MEL and 3-Hydroxykynurenine (3-OHKY)/MEL ratios post-treatment compared to pretreatment. These changes were not seen in the patients showing poor response to sertraline. In the placebo group, more favorable treatment outcome was associated with increases in 5-MTPOL and MEL levels and significant decreases in the KYN/MEL and 3-OHKY/MEL; changes in 5-MTPM levels were not associated with the 4-week response. These results suggest that recovery from a depressed state due to treatment with drug or with placebo could be associated with preferential utilization of serotonin for production of melatonin and 5-MTPOL.

Abstract: Le style de vérité accessible à l’essai est perceptible dans son mode de circulation, les souvenirs que l’on en garde, la fa on dont on le résume, bref, l’usage que l’on en fait. Ce mémorable de l’essai tient à une pratique généralisée de la citation ; les essayistes partent de formes citables , comme d’autant de pensées héritées et de savoirs doxaux ou paradoxaux qu’ils exploitent, développent ou fragilisent ; et ils en produisent à leur tour, offrant non pas des modèles applicables mais des cas remobilisables, des pensées désormais disponibles. Cette disponibilité identifie une pratique intellectuelle, les objets qu’elle fait na tre (cette forme sensible que prennent dans un essai les opérations de l’esprit), un rythme propre à toute individualité pensante, bref une fa on qu’a la prose d’idées de se situer devant le temps. Elle explique aussi la colère suscitée dans le monde du savoir contemporain par un genre profondément temporel, c’est-à-dire rythmé, situé, mais aussi précaire et soup onnable à cet égard d’anachronisme. Equilibrium fluctuations for the disordered harmonic chain perturbed by an energy conserving noise

Abstract: We investigate the macroscopic behavior of the disordered harmonic chain of oscillators, through energy diffusion. The Hamiltonian dynamics of the system is perturbed by a degenerate conservative noise. After rescaling space and time diffusively, we prove that energy fluctuations in equilibrium evolve according to a linear heat equation. The diffusion coefficient is obtained from the non-gradient Varadhan's approach, and is equivalently defined through the Green-Kubo formula. Since the perturbation is very degenerate and the symmetric part of the generator does not have a spectral gap, the standard non-gradient method is reviewed under new perspectives.
Hydrodynamic limit for the velocity flip model

Abstract: We study the diffusive scaling limit for a chain of $N$ coupled oscillators. In order to provide the system with good ergodic properties, we perturb the Hamiltonian dynamics with random flips of velocities, so that the energy is locally conserved. We derive the hydrodynamic equations by estimating the relative entropy with respect to the local equilibrium state modified by a correction term.
Red Wine administration to Apolipoprotein E-deficient Mice reduces their Macrophage-derived Extracellular Matrix Atherogenic Properties

Abstract: proteoglycans (pgs) from the arterial extracellular matrix (ecm) contribute to the trapping of ldl and oxidized ldl (ox-ldl) in the arterial wall, a phenomenon called "lipoprotein retention". moreover, we have shown that subsequent to their binding to the matrix, ldl and ox-ldl are taken up by macrophages. oxidative stress significantly increases macrophage secretion of ecm-pgs, lipoprotein binding to the ecm and the uptake of ecm-retained lipoproteins by macrophages. the aim of the present study was to determine whether red wine administration to atherosclerotic mice would affect their peritoneal macrophage-derived extracellular matrix properties, such as the glycosaminoglycan content and the ability to bind ldl. in addition, we questioned the ability of ldl bound to the mice peritoneal macrophages-derived ecm to be taken up by macrophages. red wine administration to atherosclerotic mice did not affect the mice peritoneal macrophages-derived ecm glycosaminoglycan content but it significantly reduced the mice peritoneal macrophages-derived ecm ability to bind ldl and the subsequent uptake of ecm-retained ldl by the macrophages. the present study thus clearly demonstrated the inhibitory effect of red wine consumption by e0 mice on their peritoneal macrophage-derived extracellular matrix atherogenic properties.

Abstract: Cette recherche se propose d’aborder le mode de production du produit alimentaire sous l’angle des représentations mentales en France. Dans le langage commun on entend souvent les termes industriel ou artisanal pour caractériser les produits alimentaires, sans que l’on sache vraiment ce que ces termes désignent. Le contexte actuel d’incertitude et de méconnaissance des systèmes de production chez les consommateurs nous permet d’émettre l’hypothèse de l’existence d’une représentation mentale du mode de production. Nous tentons de vérifier cette hypothèse par une étude qualitative à l’issue de laquelle les premières caractéristiques du produit alimentaire industriel ou artisanal sont précisées. This research approaches the food product through the mental representations concerning manufacturing processes. In the common language we often find such terms as “industrial” or “traditional” to qualify production methods and characterize food products, but do we know what these terms really mean? Uncertainties and misunderstandings regarding food production enable us to highlight the existence of consumers’ mental representations regarding manufacturing processes. Such representations have been studied by means of a qualitative study which enabled us to define the primary characteristics of industrial or traditional food products.
