Galectin-3 Impairment of MYCN-Dependent Apoptosis-Sensitive Phenotype Is Antagonized by Nutlin-3 in Neuroblastoma Cells

Abstract: MYCN amplification occurs in about 20–25% of human neuroblastomas and characterizes the majority of the high-risk cases, which display less than 50% prolonged survival rate despite intense multimodal treatment. Somehow paradoxically, MYCN also sensitizes neuroblastoma cells to apoptosis, understanding the molecular mechanisms of which might be relevant for the therapy of MYCN amplified neuroblastoma. We recently reported that the apoptosis-sensitive phenotype induced by MYCN is linked to stabilization of p53 and its proapoptotic kinase HIPK2. In MYCN primed neuroblastoma cells, further activation of both HIPK2 and p53 by Nutlin-3 leads to massive apoptosis in vitro and to tumor shrinkage and impairment of metastasis in xenograft models. Here we report that Galectin-3 impairs MYCN-primed and HIPK2-p53-dependent apoptosis in neuroblastoma cells. Galectin-3 is broadly expressed in human neuroblastoma cell lines and tumors and is repressed by MYCN to induce the apoptosis-sensitive phenotype. Despite its reduced levels, Galectin-3 can still exert residual antiapoptotic effects in MYCN amplified neuroblastoma cells, possibly due to its specific subcellular localization. Importantly, Nutlin-3 represses Galectin-3 expression, and this is required for its potent cell killing effect on MYCN amplified cell lines. Our data further characterize the apoptosis-sensitive phenotype induced by MYCN, expand our understanding of the activity of MDM2-p53 antagonists and highlight Galectin-3 as a potential biomarker for the tailored p53 reactivation therapy in patients with high-risk neuroblastomas.
Great Expectations goes to Hollywood. La rilettura di un classico da Lean a Cuarón

Abstract: L'articolo intende dare una lettura dell'adattamento di Great Expectations, da parte di Alfonso Cuaron, in rapporto non solo al romanzo di Dickens ma soprattutto nei confronti dell'adattamento di David Lean del 1946. Il film di Lean funziona, infatti, per Cuaron da ipotesto da riadattare per un pubblico contemporaneo ed hollywoodiano. Il tempo in Lean rimane quello ottocentesco con tutto ciò che ne comporta (differenze di classe, obblighi sociali), Cuarón, invece, tradisce fin dalla prima inquadratura il romanzo d'origine, spostando l'azione in Florida e ai giorni nostri. E non è più Londra il luogo da raggiungere per compiere la propria formazione ma New York. Il bianco e nero che è in un certo senso già del libro si trasforma in un'esuberanza del colore che trascina con sé personaggi ed azioni, recitazione, montaggio, colonna sonora. Nonostante i presupposti lascerebbero pensare ad un chiaro happy end, il film del regista messicano non lascia, però, entrare metaforicamente la luce dalla finestra come avviene in Lean e ciò perché il dopoguerra di Lean può ancora avere delle grandi speranze da realizzare, il postmodernismo di Cuarón fatica a crederci.
INSEGNAMENTO E APPRENDIMENTO DELLA STORIA IN UNA CLASSE PLURILINGUE E MULTICULTURALE

Abstract: Apprendere una disciplina non significa esclusivamente assimilarne i contenuti, ma anche parlarne la lingua specifica: si tratta di una questione che la presenza sempre maggiore di studenti non madrelingua nella scuola italiana ha reso solo più evidente e riproposto in tutta la sua urgenza. Da recenti indagini, tra i linguaggi disciplinari, quello della storia è risultato essere uno dei più complessi; a partire quindi da un’analisi delle caratteristiche specifiche della lingua della storia (dal lessico alla morfo-sintassi, dall’organizzazione testuale alla tipologia dei concetti della disciplina), il presente intervento si propone di individuare le possibili strategie di semplificazione e di facilitazione didattica, evidenziandone anche gli eventuali limiti o risvolti negativi. L’articolo si propone quindi di riflettere sulla necessità di rinnovare l’insegnamento disciplinare, rivedendo programmi e metodologie didattiche, anche al fine di un’educazione linguistica globale per l’apprendimento delle lingue a scuola. Teaching and learning history in a multilingual, multicultural class Studying a subject does not mean merely assimilating the content, but also using specific language: the ever-increasing presence of non-mothertongue students in Italian schools makes this appear more and more evident and urgent. Recent surveys regarding different disciplines showed that history was among the most complex areas. Starting from an analysis of the specific characteristics of the language of history (from the lexis to the morphosyntax, the organization of the texts to the types of disciplinary concepts), this article singles out the possible ways to simplify and facilitate didactics, also highlighting the limits or negative aspects. The article is a reflection on the necessity to renew the teaching of disciplines, up-dating didactic programs and methods, with the aim of arriving at a global linguistic education for learning languages at school.

Para ello será analizada la recepción de las fotografías del antropólogo e indigenista mexicano Julio de la Fuente entre los indígenas zapotecos del estado de Oaxaca, al sur de este país. Abstract: Angiotensin II plays a crucial role in pathologic processes of chronic allograft nephropathy (CAN) leading to chronic and progressive renal allograft dysfunction (CRAD). Systemic and glomerular hypertensions together with proteinuria occur in CAN under conditions of JGA hypertrophy with up-regulated RAS activity in the renal allograft, and they represent independent factors of rapid progression of chronic renal allograft failure. ACEi are safe and efficient antihypertensives with renoprotective effects in patients with CAN. Favorable response to ACEi has been reflected in diminished proteinuria, slow increase of creatininemia, regulation of arterial hypertension and better long-term survival of patients and kidney allografts. Our pilot study highlights the importance of ACEi therapy in CAN after renal transplantation from elder donors (>55 years).

Abstract: Many studies have been performed previously to investigate different theories of motivation and, more specifically, how to motivate technical professionals. Through a questionnaire survey carried out on 376 development engineers, identified as “knowledge workers” by managers in their firms, this study identifies what methods are used in industry to motivate these individuals, what works and what does not. While many aspects of standard incentives and reward systems used to motivate technical professionals in general are also motivating for technical visionaries, these results indicate that they are motivated by additional factors not generally discussed in the literature. Many studies have been performed previously to investigate different theories of motivation and, more specifically, how to motivate technical professionals. Through a questionnaire survey carried out on 376 development engineers, identified as “knowledge workers” by managers in their firms, this study identifies what methods are used in industry to motivate these individuals, what works and what does not.
