Neiva, Elaine Rabelo;Paz, Maria das Gra?as Torres da;
Revista de Administra??o (S?o Paulo) , 2012, DOI: 10.5700/rausp1023
Abstract: the purpose of this study was to evaluate the individual and organizational changes perceived by the employees of a public sector organization that suffered interventions for four years. the study had three stages: measurement of organizational characteristics prior to the interventions using questionnaires; investigation of the objectives of a specific intervention and the demand for change mentioned by the organization's employees in order to construct a tool to evaluate the perceived change; and the investigation of the factors that influenced the perception of individual and organizational change. the factors that foretold the perception of organizational changes were organizational characteristics, taken as cultural variables - organizational values and power configurations (measured before and after the intervention); organizational characteristics that were change facilitators; and individual variables, such as attitudes in the face of organizational change (measured after the intervention). the criterion or dependent variable for the study was the perception of organizational and individual change. hierarchical regressions were used to analyze the relations postulated in the study. the results indicated that the organizational values of autonomy and egalitarianism and the attitudes of acceptance and fear were the most frequent predictors of the perceived changes.

Abstract: Two procedures have been proposed and compared for separation and preconcentration of trace amounts of manganese, copper, and zinc in saline samples. The procedures are based in the use of Amberlite XAD-7 and an anion-exchanger Dowex 1X8-50 loaded with Alizarin Red S (ARS). In order to obtain quantitative recoveries of metal ions, various experimental parameters such as pH, shaking time, sample volume, amounts of solid phase, effects of concomitants, capacity and cations desorptions from solid phases were optimized. For Amberlite XAD-7 impregnated with ARS (XAD 7-ARS), Mn, Cu, and Zn were quantitatively retained, in the pH range 8.5-10.0, by using 0.50 g of solid phase, stirring time of 5 min and a total mass up to 200 μg of each cation. The sorbed elements were subsequently eluted with 5 mL of 3 mol L-1 HNO3, with recovering over 95%, and a fifty-fold preconcentration factor for Zn and a ten-fold preconcentration factor for Mn and Cu were obtained. For Dowex 1X8-50 impregnated with ARS (Dowex-ARS), Cu and Mn were quantitatively retained in the pH range 8.1 to 9.0, and Zn pH 8.1, by using 0.75 g of solid phase, 90 min of stirring time and a total mass up to 50 μg of each cation. The sorbed elements were subsequently eluted with 20 mL of 2 mol L-1 HNO3, and a five-fold preconcentration factor to Cu and Zn was obtained. The precision of the procedure was determined by running 10 replicate samples, each one containing 250 μg L-1 of each element and the relative standard deviations were 0.2% (Cu), 0.4% (Mn), and 0.4% (Zn), to XAD 7-ARS and 0.3% (Cu), 0.5% (Mn), and 0.3% (Zn), to Dowex-ARS. The procedures were used for determining of Mn, Cu, and Zn in physiological solutions and seawater samples, from Salvador-Bahia. The analyte addition technique was used and the recoveries obtained (3 95%) revealed that the proposed procedure shows good accuracy and precision.

Abstract: xylosereductase (xr) (e.c., produced by candida guilliermondii, grown in sugar cane bagasse hydrolysate, was separated directly from the cell homogenate by reversed micelles of cetyl trimethyl ammonium bromide (ctab), attaining a recovery yield of 100% and enrichment factor of 5.6 fold. the extraction conditions were: ph=7.0, electrical conductivity= 14 ms/cm, t=5 oc, 5% (w/w) of hexanol, 22% (w/w) of butanol and 0.15 m ctab. the xr after extraction was stable in ph interval of 6.0-6.5 and its heat inactivation constant was about 6.5 fold higher than that before extraction. the vmax values against both xylose and nadph for xr before and after extraction by reversed-micelles differed about 6%, whereas the difference on km values were more pronounced. the (km)xylose for xr after extraction was about 35% higher than before extraction, meanwhile (km)nadph was about 30% lower after than before extraction. as the km variations indirectly signaled, the xr affinity simultaneously diminishes for xylose and increases for nadph. this could probably explain why the vmax values for xr before and after extraction were quite similar.
